Effects of Basella alba and Hibiscus macranthus extracts on testosterone production of adult rat and bull Leydig cells

Aim： To determine the androgenic effects of Basella alba and Hibiscus macranthus extracts in the rat and the bull, and to develop a novel in vitro test system using Leydig cells from bull testes. Methods： The effect of methanol extracts from both plants on testosterone production in isolated Leydig cells from the rat and the bull was analyzed using ^125I-radioimmunoassay （^125I-RIA）. Rat Leydig cells were obtained by common methods, whereas a novel technique was used to purify Leydig cells from bull testes. Results： Bull testes from the slaughter house were a cheap source of pure Leydig cells. In culture, these cells produced testosterone for 5-6 days, which can be stimulated by human chorionic gonadotrophin （hCG）. Basella alba extracts significantly enhanced testosterone production in bull and rat Leydig cells in a concentration-dependent manner. Hibiscus macranthus showed no androgenic effect but was shown to inhibit testosterone production at higher concentrations. Conclusion： Leydig cells purified from bull testes can be used as an alternative tool in experimental animal research. Certain fractions of Basella alba extract demonstrated androgenic potential whereas Hibiscus macranthus extracts did not.

We question whether,in men with an abnormal rate of sperm DNA fragmentation,the magnetic-activated cell sorting(MACS)could select spermatozoa with lower rates of DNA fragmentation as well as spermatozoa with unbalanced chromosome *** spermatozoa from six males were separated into nonapoptotic and apoptotic *** determined the percentages of spermatozoa with(i)externalization of phosphatidylserine(EpS)by annexin V-Fluorescein isothiocyanate(FITC)labeling,(ii)DNA fragmentation by TdT-mediated-dUTp nick-end labeling(TUNEL),and(iii)numerical abnormalities for chromosomes X,Y,13,18,and 21 by fluorescence in situ hybridization(FISH),on the whole ejaculate and selected spermatozoa in the same *** to the nonapoptotic fraction,the apoptotic fraction statistically showed a higher number of spermatozoa with EpS,with DNA fragmentation,and with numerical chromosomal *** to the whole ejaculate,we found a significant decrease in the percentage of spermatozoa with EpS and decrease tendencies of the DNA fragmentation rate and the sum of disomy levels in the nonapoptotic ***,we observed statistically significant higher rates of these three parameters in the apoptotic *** may help to select spermatozoa with lower rates of DNA fragmentation and unbalanced chromosome content in men with abnormal rates of sperm DNA fragmentation.

AIM： To assess the long-term clinical benefit of sustained virological response （SVR） in patients with hepatitis C virus （HCV） cirrhosis treated by antiviral therapy using mostly ribavirin plus interferon either standard or ***： One hundred and thirteen patients with uncomplicated HCV biopsy-proven cirrhosis, treated by at least one course of antiviral treatment ≥ 3 mo and followed ≥ 30 mo were included. The occurrence of clinical events [hepatocellular carcinoma （HCC）, decompensation and death] was compared in SVR and non SVR ***： Seventy eight patients received bitherapy and 63 had repeat treatments. SVR was achieved in 37 patients （33%）. During a mean follow-up of 7.7 years, clinical events occurred more frequently in non SVR than in SVR patients, with a significant difference for HCC （24/76 vs 1/37, p = 0.01）. No SVR patient died while 20/76 non-SVR did （p = 0.002）, mainly in relation to HCC （45%）.CONCLUSION： In patients with HCV-related cirrhosis, $VR is associated with a significant decrease in the incidence of HCC and mortality during a follow-up period of 7.7 years. This result is a strong argument to perform and repeat antiviral treatments in patients with compensated cirrhosis.

Near-infrared and mid-infrared spectroscopies were currently used to analyze natural compounds. During the last ten years various multiblocks methods were developed such as Concatenated pLS, Hierarchical-pLS (H-pLS), and MultiBlock-pLS (MB-pLS). These three algorithms were used to analyze 55 lavender (Lavandula angustifolia) essential oil samples. The results obtained were compared to the ones obtained respectively in NIR and MIR ranges. The accuracies of the models depend on the spectroscopic technique, pretreatment and the pLS methods. The results showed that the choice of the factor numbers used to build the multiblock models was the most important parameter for the H-pLS and MB-pLS methods.

What Is the Role of Work-Related Factors in Self-Reported Health Inequalities among Employed Individuals? A Longitudinal Study in Luxembourg

Objectives: We measured health inequalities among employed Luxembourg residents over time and the socio-economic and work-related determinants. Design and Setting: Longitudinal data were obtained from the Socio-economic Liewen zu Lëtzebuerg/European Union Survey on Income and Living Conditions, which has been conducted each year since 2003 in Luxembourg. participants: participants comprised 727 Luxembourg residents (58% men), aged between 21 and 55 years in 2003, who were employed between 2003 and 2012. primary and Secondary Outcomes Measured: The variable of interest was self-reported health. We used transition indicators on work-related factors to consider changes that individuals may have experienced in their job over this period. Results: people who moved from a part-time to a full-time contract (odds ratio (OR): 5.52, confidence interval (CI): 1.55 - 19.73), and those who moved from the 3rd or 4th quartile of earnings to the 1st or 2nd quartile (OR: 2.48, CI: 1.02 - 6.05) between 2003 and 2012, had a higher risk of being in poor health in 2012. The risk of deterioration in self-reported health in 2012 among people who were healthy in 2003 was associated with the type of contract, economic activity, and occupation. Conclusion: Health inequalities occur among employed people in Luxembourg. Their importance varies according to work-related characteristics and economic activity. Our findings showed that declined health status was associated with contract type, profession, and economic activity. This suggests that measures should be taken to maintain good health for people working in these specific occupations or economic sectors (e.g. preventive action, reduction of risk exposure, change of occupation in the same company, and so on).

The influence of different carbon and nitrogen sources, of ethanol concentration, the optimal pH, temperature and medium composition were evaluated on extracellular polysaccharides (EpS) synthesis and bacterial growth of Gluconacetobacter hansenii LMG1524, and preliminary characterization of EpS was investigated. The highest EpS yields were obtained using glycerol and ammonium sulphate as carbon and nitrogen sources, respectively. The increase of ethanol concentration in the medium did not influence the EpS synthesis but reduced the bacterial growth. The optimum temperature and pH for polysaccharides production were respectively 25℃ and 5;whereas for cell growth were respectively 30℃ and 4. The optimal culture medium composition was determined as follows: 10 g/L sucrose, 0.892 g/L (NH4)2SO4, 0.34 g/L NaNO3, 3 mL acetic acid, 1.5 g/L KH2pO4, 1.5 g/L K2HpO4 and 0.6 g/L MgSO4. The polysaccharides produced were of 14 and 10 polymerization degrees (Dp) and constituted mainly of glucose, galactose and mannose, in relative percent of 36.36, 33.94 and 22.42, respectively.
