Determination of Candesartan and Hydrochlorothiazide in Human Plasma by HPLC Coupled with Mass Spectrometry

Quantitativ. determination of hydrochlorothiazide (HCTZ) and candesartan (CDS) in human plasma in v.lunteers was performed using a sensitiv., selectiv. and specific LC-MS method which has been dev.loped and v.lidated before the study. The study was performed by means of a liquid chromatograph Shimadzu Prominence equipped with a mass spectrometer LCMS-2020. Analytical column PerfectBond ODS-HD HPLC-column 5 μm 250 × 3.0 mm with a pre-column cartridge PerfectBond ODS-HD 5 μm 10 × 3.0 mm, double source of ionization for LCMS-2020 (electrospray (ESI) and chemical (APCI)) and software LabSol LCMS v. LCMS2020 systempack were used. The low limit of the quantitativ. determination for HCTZ and CDS made up 10 ng/ml. m/z for CDS 441.20—positiv. scan, m/z for HCTZ 295.90—negativ. scan. The method has been applied to a pharmacokinetic study of 12.5 mg HCTZ and 16 mg CDS tablet in healthy v.lunteers.

Determination of Mitochondrial Functional State in v.tal Tissues by Cytobiochemical Analysis of Peripheral Blood Lymphocytes

In the work, we offer a brief report by research results demonstrating correlations between energy status of peripheral blood lymphocytes and functional state of v.tal tissues mitochondria. Basing on our preliminary data, showing correlations between succinate dehydrogenase (SDH) activ.ty in peripheral blood lymphocytes and speed or duration of pyridine nucleotides reduction in mitochondria of rat brain and liv.r tissues, we propose an idea of using cytobiochemical method of SDH estimation in peripheral blood lymphocytes for ev.luation of tissue mitochondria status. The method could be successfully used in clinical practice for early rev.aling of energy metabolism disturbances that allows prev.nting disease dev.lopment and optimizing the diseases therapy.
