The inhibitory effects of NKX3.1 on IGF- 1R expression and its signalling pathway in human prostatic carcinoma PC3 cells

NKX3.1, which is a prostate-specific homeobox gene, plays an important role in prostate cancer and usually functions as a tumour suppressor gene. In this study, we investigated the inhibitory effect of NKX3,1 on insulin-like growth factor （IGF）- 1R expression and its downstream signalling pathway in PC3 cells, PC3 cells were stably transfected with NKX3.1 expression plasmid （pcDna3.1-NKX3.1） or vector plasmid （pcDna3.1＋）. The IGF-IR mRna and protein expression levels were assessed in PC3-NKX3.1 transfectants by reverse transcriptase-polymerase chain reaction （RT-PCR） and Western blotting. The expression and activation of IGF-1/IGF-1R downstream signalling targets were examined by Western blotting and luciferase reporter assay. The cells were subsequently treated with relevant concentrations of IGF-1. The effect of IGF-1 on cell growth was examined by 3-（4,5）-dimethylthiahiazo（-z-yl）-3, 5-diphenytetrazoliumromide （MTT） assay and flow cytometry analysis. A significant suppression of IGF-1R mRna and protein expression was observed after forced expression of NKX3.1 in PC3 cells. Correspondingly, the forced expression of NKX3.1 decreased IGF-l-induced phosphorylation of extracellular signal-regulated kinase 1/2 （ERK1/2） and protein kinase B （AKT） and activation of the Elk-1 transcription factor and downregulated the expression of the downstream target genes c-fos and cyclin D1. Furthermore, the forced expression of NKX3.1 inhibited IGF-l-induced cell growth. In conclusion, NKX3.1 could downregulate IGF-1R expression and could inhibit IGF-1R-mediated mitogen-activated protein kinase （MAPK）/ERK and AKT signalling pathways, which might partially leads to the inhibition of IGF-l-induced cell growth. This study provides new insights into the molecular mechanisms that NKX3,1 exerts against prostate cancer and ultimately expands the scope of alternative approaches in advanced prostate cancer therapy.

Electrochemical reduction of acetonitrile（ACN） to ethylamine（ETA） is a new strategy for producing high-value ***,the ultrathin nickel sulfide nanosheets（nixSyNSs） anchored on nickel foam（NF） nanohybrid（nixSyNSs/NF） were designed as an efficient bifunctional electrocatalyst for the waste *** to the introduction of the S element,the ultrathin nanosheet structure,and the three-dimensional architecture,nixSyNSs/NF simultaneously reveals excellent electrocatalytic activity for both electrochemical ACN reduction reaction（EACNRR） at the cathode and electrochemical sulfur ion(S2-) oxidation reaction（ESOR） at the *** the EACNRR,nixSyNSs/NF exhibits a Faradaic efficiency of 95.5% and the ETA yield of 923.1 mmol h-1g-1at-0.05 V *** the ESOR,the S2-ion is oxidized to the value-added S8product,in which the oxidation potential is only 0.16 V at 50 mA ***,the assembled nixSyNS s/NF||nixSyNSs/NF electrolytic cell is successfully established for the ESOR-assisted EACNRR system that only needs a cell voltage of 0.32 V to reach the 50 mA cm-2current *** work provides an effective and energy-saving strategy for the co-production of value-added chemicals from pollutants.

In this study we aim to investigate target-regulatory miRna of cutaneous malignant melanoma（CMM） key disease-causing gene CCL18 and analysis expression of predicted miRna and CCL18,as well as their *** analysis was performed to predict target-regulatory miRna of CCL18,real-time quantitative PCR was used to quantify the expression of CCL18 and selected miRnas in fourteen CMM tissues and luciferase reporter gene assays was used to verify the interaction of each miRna and CCL18-3’*** results showed that CCL18 with miR-183、miR-128 or miR-33 a would have a conserved and stable *** fourteen CMMs,the expression of these miRna declined,while that of CCL18 ***=-0.367,-0.548 and-0.185,*** only the relevance of miR-128 and CCL18 has statistical significance（P＜0.05）.The mut-128 3’UTR group showed a significantly higher expression of luciferase,in contrast with WT 3’UTR group and blank control（P＜0.05）.miR-128 is likely to negatively regulate CCL18,which is one of key disease-causing genes in CMM.

Fabrication of ni-Zn spinel ferrite with superior magnetic performance from electric arc furnace dust and iron scale

The ni-ZnFe_(2)O_(4)(nixZn_(1-x)Fe_(2)O_(4),x=0.4-0.7)spinel was synthesized using Zn2+extracted from electric arc furnace dust(EAFD),nickel chloride hexahydrate,and Fe^(3+)extracted from iron scale as raw *** zinc was selectively extracted from EAFD using CaO roasting followed by NH_(4)Cl solution *** ferric ion was leached from iron scale using HCl solution as acid *** experimental results demonstrate a high level of efficiency in the extraction of zinc,with a rate of 97.5%,and the leaching rate of ferric ion is 96.89%.The composition of the leaching solution is primary zinc and iron with low calcium,which is beneficial to the preparation of spinel *** influence of ni content(x)and calcination temperature on the synthesis and magnetic properties of nixZn_(1-x)Fe_(2)O_(4)compounds was investigated by X-ray diffraction,scanning electron microscopy,and vibrating sample *** results revealed that both ni content and calcination temperature significantly affect the synthesis and magnetic properties of spinel nixZn_(1-x)Fe_(2)O_(4).Under the conditions of ni content set at x=0.6,calcination temperature of 1100℃,and a duration of 2 h,a spinel nixZn_(1-x)Fe_(2)O_(4)with high saturation magnetization(Ms=65.7 A m2 kg-1)and low coercivity(Hc=0.056 A m^(-1))was obtained.

MOF-derived ni3Fe/ni/niFe2O4@C for enhanced hydrogen storage performance of MgH2

Magnesium hydride（MgH2） is an important material for hydrogen（H2） storage and transportation owing to its high capacity and ***,its intrinsic properties have considerably limited its industrial *** this study,the niFe-800 catalyst as metal-organic framework（MOF） derivative was first utilized to promote the intrinsic properties of *** to pure MgH2,which releases1.24 wt% H2in 60 min at 275℃,the MgH2-10 niFe-800 composite releases 5.85 wt% H2in the same *** at a lower temperature of 250℃,the MgH2-10 niFe-800 composite releases 3.57 wt% H2,surpassing the performance of pure MgH2at 275℃.Correspondingly,while pure MgH2absorbs 2.08 wt% H2in60 min at 125℃,the MgH2-10 niFe-800 composite absorbs 5.35 wt% H2in just 1 min,Remarkably,the MgH2-10 niFe-800 composite absorbs 2.27 wt% H2in 60 min at 50℃ and 4.64 wt% H2at 75℃.This indicates that MgH2-10 niFe-800 exhibits optimum performance with excellent kinetics at low ***,the capacity of the MgH2-10 niFe-800 composite remains largely stable after ***,the Mg2ni/Mg2niH4acts as a "hydrogen pump",providing effective diffusion channels that enhance the kinetic process of the composite during ***,Fe0facilitates electron transfer and creates hydrogen diffusion channels and catalytic ***,carbon（C） effectively prevents particle agglomeration and maintains the cyclic stability of the ***,the synergistic effects of Mg2ni/Mg2niH4,Fe0,and C considerably improve the kinetic properties and cycling stability of *** work offers an effective and valuable approach to improving the hydrogen storage efficiency in the commercial application of MgH2.
