Caused by four serotypes,dengue fever is a major public health concern *** modeling efforts have mostly focused on primary and heterologous secondary infections,assuming that lifelong immunity prevents reinfections by the same ***,recent findings challenge this assumption,prompting a reevaluation of dengue immunity *** this study,we develop a within-host modeling framework to explore different scenarios of dengue *** previous studies,we go beyond a deterministic framework,considering individual immunological *** deterministic and stochastic models are calibrated using empirical data on viral load and antibody(Igm and IgG)concentrations for all dengue serotypes,incorporating confidence intervals derived from stochastic *** good agreement between the mean of the stochastic realizations and the mean field solution for each model,our approach not only successfully captures primary and heterologous secondary infection dynamics facilitated by antibody-dependent enhancement(AdE)but also provides,for the first time,insights into homotypic reinfection *** study discusses the relevance of homotypic reinfections in dengue transmission at the population level,highlighting potential implications for disease prevention and control strategies.

Background The utility of routine extensive molecular profiling of pediatric tumors is a matter of debate due to the high number of genetic alterations of unknown significance or low evidence and the lack of standardized and personalized decision support *** drug assignment(ddA)is a novel computational method to prioritize treatment options by aggregating numerous evidence-based associations between multiple drivers,targets,and targeted *** has been validated to improve personalized treatment decisions based on the outcome data of adult patients treated in the SHIVA01 clinical *** aim of this study was to evaluate the utility of ddA in pediatric *** Between 2017 and 2020,103 high-risk pediatric cancer patients(nvolved in our precision oncology program,and samples from 100 patients were eligible for further *** or blood samples were analyzed by whole-exome(WES)or targeted panel sequencing and other molecular diagnostic modalities and processed by a software system using the ddA algorithm for therapeutic decision ***,a molecular tumor board(mTB)evaluated the results to provide therapy *** Of the 100 cases with comprehensive molecular diagnostic data,88 yielded WES and 12 panel sequencing *** identified matching off-label targeted treatment options(actionability)in 72/100 cases(72%),while 57/100(57%)showed potential drug *** reached 88%(29/33)by 2020 due to the continuous updates of the evidence *** approved the clinical use of a ddA-top-listed treatment in 56 of 72 actionable cases(78%).The approved therapies had significantly higher aggregated evidence levels(AELs)than dismissed *** of WES results for targeted panels missed important mutations affecting therapy *** ddA is a promising approach to overcome challenges associated with the interpretation of extensive molecular profiling in the routine car

With high water content,excellent biocompatibility and lubricating properties,and a microstructure similar to that of the extracellular matrix,hydrogel is becoming one of the most promising materials as a substitute for articular ***,it is a challenge for hydrogel materials to simultaneously satisfy high loading and low *** hydrogels are brittle,with fracture energies of around 10 J·m^(-2),as compared with∼1000 J·m^(-2) for cartilage.A great deal of effort has been devoted to the synthesis of hydrogels with improved mechanical properties,such as increasing the compactness of the polymer network,introducing dynamic non-covalent bonds,and increasing the hydrophobicity of the polymer,all at the expense of the lubricating properties of the ***,we develop a hydrogel material with anisotropic tubular structures where the compactness gradually decreases and eventually disappears from the surface to the subsurface,achieving a balance between lubrication and *** porous layer with hydrophilic carboxyl groups on the surface exhibits extremely low friction(coefficient of friction(COF)∼0.003,1 n;COF∼0.08,20 n)against the hard steel ball,while the bottom layer acts as an excellent load-bearing *** is more,the gradual transition of the tubular structures between the surface and the subsurface ensures the uniform distribution of friction stress between a lubricating and bearing layers,which endows the material with long-lasting and smooth friction *** extraordinary lubricious performance of the hydrogels with anisotropic tubular structure has potential applications in tissue engineering and medical devices.

As a substitute for synthetic ammonia under mild condition, electrocatalytic nitrogen reduction reaction(nRR) provides a hopeful approach for the development of ammonia. nevertheless, the current development of nRR electrocatalysts is far from enough and a systematic research is needed to gain a better improvement. This article presents that 2 d C_(3)n_(4)-nV with a large specific surface area and abundant nitrogen vacancies is prepared by a simple and feasible method, and used as a metal-free catalyst for electrocatalytic nRR. Experiment result and density functional theory(dFT) calculation reveal that nitrogen vacancies in 2 d C_(3)n_(4)-nV can act as an efficient active site for catalytic nRR, which is conducive to capturing and activating n_(2), lowering Gibbs free energy(dG) in reaction and inhibiting hydrogen evolution reaction(HER) at the same time. In addition, the larger specific surface area also makes more active site exposed, which is good for the contact between the electrolyte and the active site, thus enhancing its nRR activity. The electrocatalyst shows an excellent catalytic activity for nRR in 0.1 m HCl, including Faradaic efficiency of 10.96%, nH_(3) yields of 17.85 lg h^(-1) mg_(cat)^(-1)., and good stability(over 20 h).

management of mSW(municipal Solid Waste)is a major downside in most of the biggest *** composting of the organic fraction of mSW is one of the oldest and simplest ways of organic waste *** is a self-heating biological conversion that generates appropriate finished merchandise such as soil conditioner or *** City generates about 12,500 ton/day of mSW,44%of it is food scraps and yard trimmings which are the OFmSW(Organic Fraction of municipal Solid Waste),2,500 ton/day of it is composted at the BPCP(Bordo Poniente Composting Plant)yielding 500 ton/day of *** purpose of this study was to evaluate three treatments to accelerate the composting process,so eventually the city could increase the amount of the OFmSW at *** compared three different treatments;one of them showed a significant reduction in time of the composting process(p≤0.05)i.e.,it took less time to reach the thermophilic stage,maturity and stabilization *** was achieved at 35 days and 60 days to *** conclude that shredding the feedstock plus leachate inoculum addition at the beginning of the process,reduces the composting time in about 61%with respect to the time it takes at the BPCP,where lasted 90 days.

Interlayer and doping engineering in partially graphitic hollow carbon nanospheres for fast sodium and potassium storage

Constructing anodes with fast ions/electrons transfer paths is an effective strategy to achieve high-performance sodium ion batteries(SIBs)/potassium ion batteries(PIBs). Amorphous carbon is a promising candidate anode for SIBs/PIBs owing to its disordered carbon layers, abundant defects/pores, and lowcost resources. However, the larger radius of na^(+)/K^(+) leading to depressed kinetics and poor cycling performance, impeding their further applications. Herein, we propose an efficient strategy to construct of nitrogen, sulfur co-doped hollow carbon nanospheres(nS-HCS) involving an in situ growth of polydopamine on nano-ni(OH)2template with subsequent sulfur doping process. during the formation process, the produced ni nanospheres play as the hard template and catalyst for the formation of hollow carbon nanosphere with partially graphite microcrystalline structure, while the sulfur doping process can enlarge the interlayer space and create more defects on the surface of carbon nanospheres, thus synchronous improve the na^(+)/K^(+) insertion and adsorption ability in nS-HCS. With the synergistic control of the enlarged interlayer spacing, high content of pyridinic n/pyrrolic n and graphitization, a hybrid storage mechanism facilitates the transport kinetics and endows the nS-HCS electrode with high capacities and good cycling stability in SIBs and PIB. Benefit from the multiple effects, nS-HCS exhibits the improved capacity of 274.8 m Ah/g at 0.1 A/g and excellent cycling stability of 149.5 m Ah/g after 5000 cycles at2.5A/g in SIBs, as well as good potassium ion storage behavior with a high capacity retention of 76.5%after 700 cycles at 1.0 A/g, demonstrating the potential applications of nS-HCS for high-performance SIBs and PIBs.

AIm To investigate the role of regulatory T cell(Treg) subsets in the balance between Treg and T helper 17(Th17) cells in various tissues from mice with dextran sulfate sodium-induced *** T r e g c e l l s, T r e g c e l l s u b s e t s, T h 1 7 c e l l s, a n d Cd4+Cd25+FoxP 3+IL-17+ cells from the lamina propria of colon(LPC) and other ulcerative colitis(UC) mouse tissues were evaluated by flow cytometry. Forkhead box protein 3(FoxP 3), interleukin 17A(IL-17A), and RORC m RnA levels were assessed by real-time PCR, while interleukin-10(IL-10) and IL-17 A levels were detected with a Cytometric Beads *** In peripheral blood monocytes(PBmC), mesenteric lymphnode(mLn), lamina propria of jejunum(LPJ) and LPC from UC mice, Treg cell numbers were increased(P nd FoxP 3 and IL-10 mR nA levels were decreased. Th17 cell numbers were also increased in PBmC and LPC, as were IL-17 A levels in PBmC, LPJ, and serum. The number of FrI subset cells(Cd4+Cd45ra+FoxP 3low) was increased in the spleen, mLn, LPJ, and LPC. FrI I subset cells(Cd4+Cd45ra-Fox P3high) were decreased among PBmC, mLn, LPJ, and LPC, but the number of Fr III cells(Cd4+Cd45ra-FoxP 3low) and Cd4+Cd25+FoxP 3+IL-17A+ cells was increased. Fox P3 m RnA levels in Cd4+Cd45ra-Fox P3 low cells decreased in PBmC, mLn, LPJ, and LPC in UC mice, while IL-17 A and RORC mR nA increased. In UC mice the distribution of Treg, Th17 cells, Cd4+Cd45ra-FoxP 3high, and Cd4+Cd45ra-FoxP 3low cells was higher in LPC relative to other *** Increased numbers of Cd4+Cd45ra-FoxP 3low cells may cause an imbalance between Treg and Th17 cells that is mainly localized to the LPC rather than secondary lymphoid tissues.
