Identification of Quantitative Trait Loci (QTL) Underlying Protein, Oil, and Five Major Fatty Acids’ Contents in Soybean

Improved seed composition in soybean [Glycine max (L.) Merr.] for protein and oil quality is one of the major goals of soybean breeders. A group of genes that act as quantitative traits with their effects can alter protein, oil, palmitic, stearic, oleic, linoleic, and linolenic acids percentage in soybean seeds. The objective of this study was to identify Quantitative Trait Loci (QTL) controlling protein, oil, and fatty acids content in a set of F5:8 rILs derived from a cross between lines, ‘MD 96-5722’ and ‘Spencer’ using 5376 Single nucleotide Polymorphism (SnP) markers from the Illumina Infinium SoySnP6K BeadChip array. QTL analysis used WinQTL Cart 2.5 software for composite interval mapping (CIM). Identified, were;one protein content QTL on linkage group (LG-) B2 or chromosome (Chr_) 14;11 QTL associated with oil content on six linkage groups LG-n (Chr_3), LG-A1 (Chr_5), LG-K (Chr_9), LG-F (Chr_13), LG-B2 (Chr_14), and LG-J (Chr_16);and sixteen QTL for five major fatty acids (palmitic, stearic, oleic, linoleic, and linolenic acids) on LG-n (Chr_3), LG-F (Chr_13), LG-B2 (Chr_14), LG-E (Chr_15), LG-J (Chr_16), and LG-G (Chr_18). The SnP markers closely linked to the QTL reported here will be useful for development of cultivars with altered oil and fatty acid compositions in soybean breeding programs.

Limited information is available on the genetic analysis of amino acid composition in soybean seeds. Previously, quantitative trait loci (QTLs) for seed isoflavones, protein, oil, and fatty acids were identified in the “MD96-5722” by “Spencer” and other rIL populations. There were wide variations for these seed constituents among the rIL populations. Therefore, the objective of this study was to identify QTLs controlling different amino acids content in soybean seeds. To achieve this objective, ninety-two F5:7 recombinant inbred lines (rIL), developed from a cross of MD96-5722 and Spencer, using a total 5376 Single nucleotide Polymorphism (SnPs) markers, were used. The rILs were genotyped by using 537 polymorphic, reliably segregating SnP markers, developed from the Illumina Infinium SoySnP6K BeadChip array. A total of 13 QTLs were identified with three QTLs for threonine on the linkage group (LG) A1, C2, and B2. Two QTLs were identified for each of the amino acids proline on LG D1a and B2, serine on LG A1 and C2, tryptophan on LG K and G, and cysteine on LG A1 and K. One QTL was identified for arginine on LG n and histidine on LG J. The new QTLs findings for seed amino acid will facilitate the development of soybean cultivars with higher protein and amino acid quality to help meet the industry and consumer needs.

通过土壤培育试验,运用定量PCr技术,研究了添加生物炭和有机肥对土壤氮营养及氨氧化古菌(AOA)和氨氧化细菌(AOB)丰度的影响。结果表明,土壤培育时间、有机肥显著增加硝态氮的含量(P<0.001),促进硝化作用;生物炭通过抑制有机氮矿化作用降低硝化作用,106d时生物炭处理的硝态氮、可溶性总氮含量低于没有生物炭的处理(P<0.05),并且中和了有机肥对矿化作用的促进效应。随着处理时间的延长,生物炭促进氨氧化菌群的丰度,AOA丰度的提高较AOB显著,且AOA丰度的变化与硝态氮含量的变化呈正相关。因此认为AOA是土壤中氨氧化作用的主要驱动力,且生物炭与有机肥主要对AOA的丰度产生影响。

The Effect of Media on Biomass and Oil Production in Botryococcus braunii Strains Kossou-4 and Overjuyo-3

The green algae Botryococcus braunii is widely recognized as a source of oil, including hydrocarbons. However, the slow rate of growth B. braunii hampers its commercial development. This stu- dy addresses this by examining the effects of three growth media on biomass and oil production in two B. braunii race B strains, Kossou-4 and Overjuyo-3. Growth of B. braunii strains in BG11 medium resulted in significantly higher growth (2.3 - 4.2 and 2.9 - 6.0 fold increases in Kossou-4 and Overjuyo-3 respectively) compared to the JM and BBM-3n media after 15 days. A similar trend was obtained when biomass was measured indirectly using optical density (OD) and chlorophyll fluo-rescence. Oil production was also significantly higher in BG11 whether measured as oil weight or absorbance (ODs at 680 and 750 nm). However, the presence of extracellular oil was shown to in-crease absorbance values making OD measurements less reliable than dry weight assays. Maximum recovery of oil was recorded when hexane was used as solvent compared to hexane-isopro- panol and heptane. These results suggest that BG11 is the best growth medium for these two strains under the conditions of this experiment.

L-Arginine Supplementation Alters Maternal Blood Biochemical Attributes and Milk Composition relative to neonatal Traits of <i>najdi</i>Ewes

In a 2 × 3 factorial arrangement, 3 levels of L-arginine (0, 75 and 150 mg/kg BW per day) were tested at two stages of pregnancy (Early 60 days vs. Late 60 days) on the maternal blood metabolites and postnatal milk composition relative to the newborn traits of najdi ewes. Thirty adult pregnant najdi ewes were randomly assigned into 6 treatments (n = 5). Control early (CE) ewes were orally administered 50 mL normal saline (0.9% naCl) daily during the first 60 days of pregnancy;control late (CL) ewes were similar to CE except saline was given at the last 60 days of pregnancy;low dose of L-arginine at early pregnancy (LE);low dose of L-arginine at late pregnancy (LL);high dose of L-arginine at early (HE) and high dose of L-arginine at late pregnancy (HL). Blood and milk samples were collected on a weekly basis. Data of lambing and newborn traits were considered. There were non-significant differences in litter size due to L-arginine;however lamb birth and weaning weights increased by 35% in LE-ewes. Arginine did not change blood protein and albumin. Low arginine decreased lipid fractions, creatinine, urea, and milk fat;however, it increased AST, ALT, milk lactose and milk ash. Administration of L-arginine at 75 mg/Kg BW per day for 60 days at an early gestation in ewes improved the maternal body health and milk composition for the welfare of the neonates.

rong>Introduction:rong> Keratoconus (KCn) is one of the degenerative corneal disorders, which causes impairment in normal visual acuity with a higher prevalence in Saudi Arabia. The study aimed to assess the awareness and knowledge regarding KCn, its risk factors, and management among the Saudi population. rong>Materials and Methods:rong> A survey was conducted using a pre-tested and validated questionnaire distributed through online platforms. A mixture of convenience and snowball sampling techniques was adopted for sample collection. Our analysis included a final sample size of 837 participant responses. The questionnaire had items related to sociodemographic characteristics (5 items), knowledge (10 items), and source of information (1 item). The mean knowledge scores were calculated and categorized (good, fair, and poor) to assess each participant’s knowledge. Pearson’s chi-square test was used to determine the statistical association between categorical variables. rong>results:rong> The mean knowledge score was found to be 4.12 ± 2.6, and 67.5% of the participants showed “poor” scores. There was no statistically significant association of knowledge scores observed with any of the sociodemographic details. It was agreed by 76% and 42.5% that genetic predisposition and chronic eye inflammation could predispose to KCn. Only 38.1% (n = 157) agreed that KCn could be treated with optical glasses or contact lenses in the initial stages. rong>Conclusion:rong> The knowledge regarding KCn was not satisfactory among our study population. There is a need to create an increased level of awareness for KCn. Health awareness programs and community campaigns should be organized to raise awareness regarding KCn and better eye health care utilization.

Convective Heat and Mass Transfer in Magneto Jeffrey Fluid Flow on a rotating Cone with Heat Source and Chemical reaction

The present paper addresses the megnetohydrodynamic Jeffrey fluid flow with heat and mass transfer on an infinitely rotating upright cone. Inquiry is carried out with heat source/sink and chemical reaction ***, constant thermal and concentration flux situations are imposed. Optimal homotopy analysis method (OHAM) is employed to achieve series solutions of the concerned differential equations. Important results of the flow phenomena are explored and deliberated by means of graphs and numerical tables. It is perceived that thermal boundary layer thickness possess contrast variations for the heat source and heat sink, respectively. The chemical reaction enhances the heat transfer rate but decline the mass transfer rate. Moreover, the precision of the existing findings is verified by associating them with the previously available work.
