An immunochemical sol-gel-based immunoaffinity purification (IAP) method for purification and detection of the progestin drug medroxyprogesterone acetate (MPA) was developed. A polyclonal antibody (Ab) for MPA was generated, and two competitive (indirect and direct) sensitive enzyme-linked immunosorbent assays (ElISAs) for its detection were developed and implemented to determine the recovery and efficiency of the sol-gel based IAP method. The detection limits of the assays were 1.4 ± 0.2 ng·ml-1 (n = 4) and 4.0 ± 0.4 ng·ml-1 (n = 25) for the indirect and direct ElISAs, respectively. The Abs did not exhibit cross-reactivity with any other progestin or steroid hormone, with the exception of megestrol acetate, with which the Ab exhibited 76% cross-reactivity. The sol-gel IAP method successfully eliminated serum interference to a degree that enabled ElISA analysis of spiked serum samples. This method was also found fully compatible with subsequent chemical analytical methods, such as liquid chromatography followed by mass spectrometry (lC-MS/MS). The approaches developed in this study form a basis for analysis of MPA in biological samples and may be further used to study population exposure to MPA and to monitor MPA contamination in water samples.

Cryptosporidium infection is estimated to cause 2.9 million diarrheal cases yearly among children aged under 24 months in sub-Saharan Africa. Studies have shown long-term climatic variations can affect infectious diseases. The burden of cryptosporidiosis in rural areas of sub-Saharan Africa is well characterized. However, the trend of Cryptosporidium infection is not known, especially in informal urban settings. This study therefore sought to determine cryptosporidiosis trends, and further explore the association between year and Cryptosporidium infection among children below 24 months in Kibera urban informal settlement in Kenya. Data collected by the Kenya Medical Research Institute longitudinal study in Tabitha clinic in Kibera from 2009 to 2015 were used. At least 3000 children aged , children presenting with diarrhea were eligible for stool sample collection (n = 477), out of which 421 stool samples were tested using TaqMan™ Array Card (TAC) polymerase chain reaction panel that included a target for Cryptosporidiumle="font-family:Verdana;"> genus. BACKGROUND Current guidelines do not address the post–sustained virological response management of patients with baseline hepatitis C virus (HCV) cirrhosis and oesophageal varices taking betablockers as primary or secondary prophylaxis of variceal bleeding. We hypothesized that in some of these patients portal hypertension drops below the bleeding threshold after sustained virological response, making definitive discontinuation of the betablockers a safe option. AIM To assess the evolution of portal hypertension, associated factors, non-invasive assessment, and risk of stopping betablockers in this population. METHODS Inclusion criteria were age > 18 years, HCV cirrhosis (diagnosed by liver biopsy or transient elastography > 14 kPa), sustained virological response after directacting antivirals, and baseline oesophageal varices under stable, long-term treatment with betablockers as primary or secondary bleeding prophylaxis. Main exclusion criteria were prehepatic portal hypertension, isolated gastric varices, and concomitant liver disease. Blood tests, transient elastography, and upper gastrointestinal endoscopy were performed. Hepatic venous pressure gradient (HVPG) was measured five days after stopping betablockers. Betablockers could be stopped permanently if gradient was lTS Sample comprised 33 patients under treatment with propranolol or carvedilol: median age 64 years, men 54.5%, median Model for End-Stage liver Disease (MElD) score 9, Child-Pugh score A 77%, median platelets 77.000 × 103/μl, median albumin 3.9 g/dl, median baseline transient elastography 24.8 kPa, 88% of patients received primary prophylaxis. 目的　探索用等位基因缺失检测的方法预测口腔黏膜早期不典型性增生病变的癌变。方法　用组织显微切割法提取组织并获得DNA ,以聚合酶链反应和多态性微卫星标记物分析检测等位基因缺失。 2 1个微卫星标记物对 8条染色体臂 ( 3p、4 q、8p、9p、11q、13q、14q和 17p)进行了检测。比较轻度和中度不典型性增生 ( 5 5例 )与高危不典型性增生 (重度不典型性增生和原位癌 2 3例 ;与癌组织同时存在的轻度和中度不典型增生 4 8例 )的等位基因缺失情况。增生病变 ( 34例 )和癌组织 ( 34例 )为对照。结果　等位基因缺失发生率随着病变恶性程度的增高而增高 ;增生病变仅见单个等位基因缺失 ,而不典型增生和癌组织可见多个等位基因缺失 ;轻度和中度不典型增生标本中 3个或 3个以上臂的等位基因缺失少见 ,而重度不典型增生、原位癌和癌组织 3个或 3个以上臂的等位基因缺失多见 ;轻度和中度不典型增生的等位基因缺失多发生在 3p、9p和 14 q ,而高危不典型增生的等位基因缺失可发生在除此以外的 4 q、8p、11q、13q、17p染色体臂上。结论　早期的不典型增生没有等位基因缺失时癌变危险可能较小 ,而出现多于 3个或 3个以上臂 ,特别是在 8p、11q、13q和 17p上的等位基因缺失。

Background: SIRT1 histone deacetylase acts on many epigenetic and non-epigenetic targets. It is thought that SIRT1 is involved in oocyte maturation;therefore, the importance of the ooplasmic SIRT1 pool for the further fate of mature oocytes has been strongly suggested. We hypothesised that SIRT1 plays the role of a signalling molecule in mature oocytes through selected epigenetic and non-epigenetic ***: We observed SIRT1 re-localisation in mature oocytes and its association with spindle *** mature oocytes, SIRT1 distribution shows a spindle-like pattern, and spindle-specific SIRT1 action decreasesα-tubulin acetylation. Based on the observation of the histone code in immature and mature oocytes, we suggest that SIRT1 is mostly predestined for an epigenetic mode of action in the germinal vesicles(GVs) of immature oocytes. Accordingly, BMl-278-driven trimethylation of lysine K9 in histone H3 in mature oocytes is considered to be a result of GV epigenetic ***: Taken together, our observations point out the dual spatiotemporal SIRT1 action in oocytes,which can be readily switched from the epigenetic to non-epigenetic mode of action depending on the progress of meiosis.

Habanero pepper(Capsicum chinense Jacq.)is a crop of economic relevance in the Peninsula of *** fruits have a high level of capsaicinoids compared to peppers grown in other regions of the world,which gives them industrial *** is an important factor that affects pepper development,nutritional quality,and capsaicinoid ***,the effect of soil type on fruit development and capsaicinoid metabolism has been little *** work aimed to compare the effect of soils with contrasting characteristics,black soil(BS)and red soil(RS),on the expression of genes related to the development of fruits,and capsaicinoid synthesis using a transcriptomic analysis of the habanero pepper *** growing in RS had bigger fruits and higher expression of genes related to floral development,fruit abscission,and softening which suggests that RS stimulates fruit development from early stages until maturation *** from plants growing in BS had enrichment in metabolic pathways related to growth,sugars,and ***,these fruits had higher capsaicinoid accumulation at 25 days post-anthesis,and higher expression of genes related to the branched-chain amino acids metabolism(ketol-acid reductisomerase KARI),pentose phosphate pathway and production of NADPH(glucose-6-phosphate-1-dehydrogenase G6PDH),and proteasome and vesicular traffic in cells(26S proteasome regulatory subunit T4 RPT4),which suggest that BS is better in the early stimulation of pathways related to the nutritional quality and capsaicinoid metabolism in the fruits.

Introduction: Neisseria is a large genus of bacteria that colonize mucosal surfaces. Of the 11 species that colonize humans, only two are pathogens, N. meningitidis and N. gonorrhoeae. Both are obligate human pathogens;the last is causal agent of gonorrhea disease. Although curable with timely antibiotic treatment, an annual incidence of more than 62 million cases is estimated by the World Health Organization and there are no successful vaccines available. In contrast, several prophylactic vaccine options for Neisseria meningitidis meningitis do exist. Of note, there is trace of cross parenteral response induced between N. meningitidis and N. gonorrhoeae, and Proteoliposome (Pl, also named outer membrane vesicles, OMV) vaccine has shown high impact on this response. Objective: To determine effect of VAMENGOC-BC? and its derivates (AFPl1 and AFCo1) at mucosal and systemic level and possible cross response against Neisseria gonhorroeae in Balb/c and C57Bl/6 mice. Methods: We evaluated cross response against N. gonorrhoeae in mouse serum IgG and saliva IgA after mucosal immunization with VA-MENGOC-BC or its derivatives (AF, Adjuvant Finlay Pl1 or AFCo (cochleate) 1). Results: Immunizations with AFPl1 or AFCo1 induce anti N. gonorrhoeae at saliva IgA and serum IgG responses similar to VA-MENGOC-BC? vaccine. Conclusions: Such data confirms a new possible window of prime-boost vaccination strategy against gonorrhea and extends our knowledge regarding the effect of Pl vaccines on cross responses.
