Type 2 diabetes mellitus and metabolic disorders have become an epidemic globally.However,the pathogenesis remains largely unclear and the prevention and treatment are still limited.In addition to environmental factors during adulthood,early life is the critical developmental window with high tissue plasticity,which might be modified by external environmental cues.Substantial evidence has demonstrated the vital role of early-life nutrition in programming the metabolic disorders in later life.In this review,we aim to overview the concepts of fetal programming and investigate the effects of early-life nutrition on energy metabolism in later life and the potential epigenetic mechanism.The related studies published on PubMed database up to March 2020 were included.The results showed that both maternal overnutrition and undernutrition increased the riskes of metabolic disorders in offspring and epigenetic modifications,including DNA methylation,miRNAs,and histone modification,might be the vital mediators.The beneficial effects of early-life lifestyle modifications as well as dietary and nutritional interventions on these deleterious metabolic remolding were initially observed.Overall,characterizing the early-life malnutrition that reshapes metabolic disease trajectories may yield novel targets for early prevention and intervention and provide a new point of view to the energy metabolism.

Quorum sensing(QS)plays an essential role in virulence factor production,biofilm formation,and antimicrobial resistance.As a potent QS inhibitor,hordenine can inhibit both QS and biofilm formation in Pseudomonas aeruginosa and Serratia marcescens.In this work,we tested the QS inhibitory potential of 27 hordenine analogs against QS and biofilm formation in P.aeruginosa and S.marcescens.Among the tested analogs,seven(12,28,27,26,2,23,and 7)exhibited strong QS inhibitory activity against P.aeruginosa,five of which(12,28,27,26,and 2)showed better inhibitory activity than hordenine.In addition,seven analogs(28,12,23,7,26,2,and 27)exhibited better biofilm inhibition against P.aeruginosa than hordenine.Four analogs(7,28,2,and 12)showed QS inhibitory activity against S.marcescens,two of which(7 and 28)demonstrated better inhibitory activity than hordenine.Furthermore,analog 7 showed similar biofilm inhibition against S.marcescens as hordenine.Structure-activity relationship(SAR)analysis indicated that the inhibitory activities of the analogs were related to four factors,i.e.,carbon chain length,presence or absence of anα,β-C=C bond,amino group with/without lipophilic group,such as methyl group,and hydroxyl group in benzene ring.

Influence Analysis of Machining and Installation Errors on the Radial Stiffness of a Non-Pneumatic Mechanical Elastic Wheel

Machining and installation errors are unavoidable in mechanical structures. However, the effect of errors on radial stiffness of the mechanical elastic wheel(ME-Wheel) is not considered in previous studies. To this end, the interval mathematical model and interval finite element model of the ME-Wheel were both established and compared with bench test results. The intercomparison of the influence of the machining and installation errors on the ME-Wheel radial stiffness revealed good consistency among the interval mathematical analysis, interval finite element simulation,and bench test results. Within the interval range of the ME-Wheel machining and installation errors, parametric analysis of the combined elastic rings was performed at different initial radial rigidity values. The results showed that the initial radial stiffness of the flexible tire body significantly influenced the ME-Wheel radial stiffness, and the inverse relationship between the hinge unit length or suspension hub and the radial stiffness was nonlinear. The radial stiffness of the ME-Wheel is predicted by using the interval algorithm for the first time, and the regularity of the radial stiffness between the error and the load on the ME-Wheel is studied, which will lay the foundation for the exact study of the ME-Wheel dynamic characteristics in the future.

This study explored the potential of using probabilistic neural networks (PNN) to predict shrinkage of thermal insulation mortar.Probabilistic results were obtained from the PNN model with the aid of Parzen non-parametric estimator of the probability density functions (PDF).Five variables,water-cementitious materials ratio,content of cement,fly ash,aggregate and plasticizer,were employed for input variables,while a category of 56-d shrinkage of mortar was used for the output variable.A total of 192 groups of experimental data from 64 mixtures designed using JMP7.0 software were collected,of which 120 groups of data were used for training the model and the other 72 groups of data for testing.The simulation results showed that the PNN model with an optimal smoothing parameter determined by the curves of the mean square error (MSE) and the number of unrecognized probability densities (UPDs) exhibited a promising capability of predicting shrinkage of mortar.

Maturity-onset diabetes of the young 10 caused by the c.4G>A (p.Ala2Thr)mutation is extremely rare, with only two reported studies to date. Herein, wereport another case that differs from previous cases in phenotype.CASE SUMMARYThe proband developed diabetes at the age of 27 years, despite having a normalbody mass index (BMI). She exhibited partial impairment of islet function, testedpositive for islet antibodies, and required high doses of insulin. Her sister alsocarried the c.4G>A (p.Ala2Thr) mutation, and their mother was stronglysuspected to carry the mutated gene. Her sister developed diabetes around 40years of age and required high doses of insulin, while the mother was diagnosedin her 20s and was managed with oral hypoglycemic agents;neither of them wereobese.CONCLUSION p.Ala2Thr mutation carriers often experience relatively later onset and normalBMI. Treatment regimens vary between individuals.

During the Ordovician-Silurian transition period,the Yangtze area experienced significant environmental changes,which were recorded by the widely distributed black shale of the Upper Ordovician Wufeng and Lower Silurian Longmaxi formations.However,the petrological and geochemical analyses of the Upper Ordovician-Lower Silurian black shale in Well JY1(eastern Sichuan Basin)and Well YY2(western Hubei Province)drill cores show that there are some significant differences with respect to lithology,thickness,and geochemical characteristics despite the long-term similar variation trend in petrology and geochemistry.This may suggest some major environmental changes and their differential influence on black shale deposition in the Yangtze area.Based on the analyses of lithology,total organic carbon(TOC),Mo_(EF),and U_(EF),four long-term sedimentary cycles(one for the Wufeng Formation and three for the Longmaxi Formation)were identified in Well JY1.In Well YY2,only three long-term sedimentary cycles can be recognized(one for the Wufeng Formation and two for the Longmaxi Formation).Different from the gradual increase of the geochemical proxies(such as TOC,MoEF,and NiEF)followed by an abrupt decrease in the Wufeng Formation,these proxies of the Silurian sedimentary cycles in both wells exhibit an abrupt increase at the base followed by a gradual decrease,showing that controlling mechanisms for the Lower Silurian black shale deposition in the study areas were different from those of the Upper Ordovician black shale.However,the black shale of each Silurian cycle in Well JY1 is much thicker than that in Well YY2.Compared with the thick silty mudstone developed in Well JY1 during the Rhuddanian-Aeronian transition,there is a stratigraphic gap in Well YY2 except for a thin calcareous dolostone formed in a strongly restricted environment.These differences indicate that tectonism and glacial eustasy should be the key factors for black shale development,but their influences on the deposition of the Lower Silurian black shale varied

Background Diabetic nephropathy is a common complication of diabetes mellitus. This study aimed to explore whether mesenchymal stem cells （MSCs） transplantation could attenuate diabetic nephropathy in experimental diabetic rats. Methods Sprague-Dawley rats received a single intraperitoneal injection of streptozotocin （STZ）（60 mg/kg）. Diabetic rats were randomized to four groups： diabetes control group （DC）, ciclosporin A group （CsA）, MSC group, and MSC＋CsA group （MSCA）. Bone marrow mesenchymal stem cells were cultured, identified and labeled by 5-bromo-2＇-deoxyuridine （BrdU） in vitro. Then they were transplanted to diabetic rats via introcardiac infusion. Ciclosporin A was administered daily at 5 mg/kg. At 1,2, 4, 8 weeks after transplantation, random blood glucose, urine albumin/creatinine ratio （AIb/Cr）, endogenous creatinine clearance rate and renal mass index were tested. Renal morphology and labeled cells were examined. Results Cultured MSCs expressed mesenchymal cell phenotype, and could be multidifferentiated to osteogenic and adipogenic cells. Labeled MSCs could be detected in the kidney of nephropathic rats, mainly in renal interstitium, but they did not propagate after engrafting in kidney. Over the course of the experiment, MSCA group showed a significant decrease in blood glucose compared with MSC group, CsA group and DC group （P 〈0.05, respectively）. The AIb/Cr in MSCA group and MSC group were significantly lower than CsA group and DC group （P〈0.05）. And the AIb/Cr in MSCA group showed a significant decrease compared with MSC group （0.74 vs 0.84, P 〈0.05）. There was a significant difference in renal mass index between the MSCA group and DC group （5.66 vs 6.37, P 〈0.05）. No significant difference was found in creatinine clearance rate among 4 groups （P 〉0.05）. Treatment with MSC＋CsA significantly ameliorated the morphology of diabetic kidney. Conclusion MSC could mildly ameliorate diabetic nephropathy by decreasing blood glucose, AIb/Cr ratio and renal mass index.

Plasmonic optical manipulation has emerged as an affordable alternative to manipulate single chemical and biological molecules in nanoscience.Although the theoretical models of sub-5 nm single-molecule trapping have been considered promising,the experimental strategies remain a challenge due to the Brownian motions and weak optical gradient forces with significantly reduced molecular polarizability.Herein,we address direct trapping and in situ sensing of single molecules with unprecedented size,down to∼5Åin solution,by employing an adjustable plasmonic optical nanogap and single-molecule conductance measurement.The theoretical simulations demonstrate that local fields with a high enhancement factor,over 103,were generated at such small nanogaps,resulting in optical forces as large as several piconewtons to suppress the Brownian motion and trap a molecule of length sub-1 nm.This work demonstrates a strategy for directly manipulating the small molecule units,promising a vast multitude of applications in chemical,biological,and materials sciences at the single-molecule level.

A facial strategy to efficiently improve catalytic performance of CoFe_(2)O_(4) to peroxymonosulfate

Cobalt iron spinel(CoFe_(2)O_(4)) has been considered as a good heterogeneous catalysis to peroxymonosulfate(PMS) in the degradation of persistent organic pollutants due to its magnetic properties and good chemical stability. However, its catalytic activity needs to be further improved. Here, a facial strategy, “in-situ substitution”, was adopted to modify CoFe_(2)O_(4) to improve its catalytic performance just by suitably increasing the Co/Fe ratio in synthesis process. Compared with CoFe_(2)O_(4), the newly synthesized Co_(1.5)Fe_(1.5)O_(4), could not only significantly improve the degradation efficiency of phenol, from 50.69 to 93.6%, but also exhibited more effective mineralization ability and higher PMS utilization. The activation energy advantage for phenol degradation using Co_(1.5)Fe_(1.5)O_(4)was only 44.2 k J/mol, much lower than that with CoFe_(2)O_(4)(127.3 k J/mol). A series of related representations of CoFe_(2)O_(4) and Co_(1.5)Fe_(1.5)O_(4) were compared to explore the possible reasons for the outstanding catalytic activity of Co_(1.5)Fe_(1.5)O_(4). Results showed that Co_(1.5)Fe_(1.5)O_(4)as well represented spinel crystal as CoFe_(2)O_(4)and the excess cobalt just partially replaced the position of iron without changing the original structure. Co_(1.5)Fe_(1.5)O_(4) had smaller particle size(8.7 nm), larger specific surface area(126.3 m^(2)/g), which was more favorable for exposure of active sites. Apart from the superior physical properties, more importantly, more reactive centers Co(Ⅱ) and surface hydroxyl compounds generated on Co_(1.5)Fe_(1.5)O_(4), which might be the major reason. Furthermore, Co_(1.5)Fe_(1.5)O_(4) behaved good paramagnetism, wide range of pH suitability and strong resistance to salt interference, making it a new prospect in environmental application.
