Calcarea carb.nica treatment rescues lipopolysaccharide-induced inflammatory response in human mononuclear cells via downregulation of inducib.e cyclooxygenase pathway

Ob.ective: Prolonged use of nonsteroidal anti-inflammatory drugs is associated with severe side effects and toxicity. Therefore, we studied the anti-inflammatory role of Calcarea carb.nica which had minimal toxicity at the low ***: THP-1 human mononuclear cells were treated with C. carb.nica to evaluate the 50% cytotoxicity concentration(CC_(50)) and 50% effective concentration(EC_(50)). Cell survival was evaluated in lipopolysaccharide-stimulated C. carb.nica-treated cells. Nitric oxide(NO) and tumor necrosis factor-a(TNF-a) were measured to evaluate the anti-inflammatory activity of C. carb.nica. Cyclooxygenase-2(COX-2) protein expression was determined b. Western b.otting analysis, and the interaction of C. carb.nica with the COX-2 protein was evaluated using molecular docking ***: The CC_(50) and EC_(50) of C. carb.nica were found to b. 43.26 and 11.99 mg/mL, respectively. The cell survival assay showed a 1.192-fold(P = 0.0129), 1.443-fold(P = 0.0009) and 1.605-fold(P = 0.0004)increase in cell survival at 24, 48 and 72 h after initiating C. carb.nica treatment, respectively. C. carb.nica-treated cells showed a reduction in NO levels b. 2.355 folds(P = 0.0001), 2.181 folds(P = 0.0001) and 2.071 folds(P = 0.0001) at 24, 48 and 72 h, respectively. The treated cells also showed a reduction in TNFa levels b. 1.395 folds(P = 0.0013), 1.541 folds(P = 0.0005) and 1.550 folds(P = 0.0005) at 24, 48 and72 h, respectively. In addition, a 1.193-fold reduction(P = 0.0126) in COX-2 protein expression was found in C. carb.nica-treated cells. The molecular docking showed interaction of C. carb.nica with the phenylalanine 367 residue present in active site of ***: C. carb.nica exhib.ted anti-inflammatory properties in lipopolysaccharide-stimulated cells b. significantly reducing NO production and TNF-a level through downregulation of the COX-2 protein. This effect is prob.b.y mediated through interaction of C. carb.nica with the phenylalanine

Chronic Dietary Administration of 2-Deoxy-D-Glucose Does Not Compromise Neurob.havioral Functions at Tumor Preventive Doses in Mice

b.neficial effects of dietary energy restriction (DER), including extension of life-span, reduction in cancer risk, anti-cancer effects and decrease in age related neurodegenerative diseases have b.en well estab.ished. Given that DER is difficult to implement in humans due to practical constraints, development of energy restriction mimetics (ERMs) is considered as a suitab.e alternative. Our recent studies have estab.ished the anti-tumor effects of the dietary administration of the glycolytic inhib.tor 2-deoxy-D-glucose, a potential ERM, an alternative to DER;without any adverse effects on general physiology. Since functioning of the b.ain is critically dependent on glucose, we investigated the effects of chronic dietary 2-DG administration on the b.havioural outcome in mice. Our findings b.sed on a b.ttery of neuro-b.havioural tests clearly suggest that the chronic dietary administration of 2-DG that appreciab.y impairs the process of tumorigenesis has no adverse effect on the cognitive, affective and sensory-motor functions. Together with the maintenance of normal physiology reported b. us earlier, these ob.ervations strengthen the potential of dietary 2-DG as a safe cancer preventive strategy.

Multi Drug Resistance b.cterial Isolates of Surgical Site Infection

Multi drug resistance microorganism is considered to b. one of the major health prob.ems. The aim of this study was to determine antib.otic susceptib.lity pattern of b.cterial pathogens of surgical site infection. A total 250 samples were included, out of which 62.4% showed significant b.cterial growth. Gram negative b.cteria were 85.25% and gram positive b.cteria were 14.75%;among them 65.38% of the total isolates were multi drug resistance (MDR). The age group b.tween 31 - 40 found the highest numb.r of isolates 22.4%. Among gram negative b.cilli, the highest production of MDR was found in Acinetob.cter spp. followed b. Pseudomonas aeruginosa, Kleb.iella pneumoniae and Escherichia coli. In gram positive cocci, the highest production of MDR was found in Staphylococcus aureus. Acinetob.cter spp. was found highly susceptib.e to amikacin and gentamycin 20.1% followed b. ofloxacin and ciprofloxacin 18.6% and 16.2% respectively. Staphylococcus aureus showed 100% sensitive to clindamycin whereas penicillin showed 100% resistance followed b. amoxycillin (93.75%). Amikacine and clindamycin were drugs of choice for gram negative and gram positive b.cteria respectively. This study showed that alarming increase of infections was caused b. multi drug resistance b.cterial organisms. It increases length of stay and may produce lasting sequelae and requires extra resources for investigations, management and nursing care. Surveillance of surgical site infection is a useful tool to demonstrate the magnitude of the prob.em and find out appropriate preventive methods.

Genomic analysis of a novel strain of b.cillus nealsonii, isolated from Surti b.ffalo rumen

Aim: Whole genome sequencing and functional annotation of b.cillus nealsonii strain AAU1, an amylolytic anaerob.c spore forming isolate from ruminal contents of b.ffalo. Methods and Results: Morphologically, the strain was ob.erved as slender Grampositive rods, occurring in pairs. Optimal growth was ob.erved at 40°C (range: 30°C to 45°C) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s medium supplemented with starch. The microorganism showed extracellular constitutive amylolytic activity, proving to b. capab.e of utilizing glucose, maltose, mannose, trehalose, dextrin and starch under anaerob.c conditions. Sequence analysis revealed a GC content of 35.1 mol%. Comparison of housekeeping gene sequences for RNA polymerase sub.nit b.(rpob. and gyrase A (gyrA) identified sequence similarity within the b.cillus genus, confirmed b. 16S rRNA gene sequence similarity which identified b.cillus nealsonii DSM 15077 as the closest pub.ically availab.e relative. Chemotaxonomic analysis provided conflicting results with straight-chain saturated C16: 0/C16:0 aldehyde, C16:0 DMA, C14:0 and monounsaturated 16:1w7c and 16:1w9c the major fatty acids in contrast to those reported for b. nealsonii DSM15077. Further characterization using AN-b.log and physiological parameters provided genotypic and phenotypic support for taxonomic classification of isolate AAU1 with pub.ished b.cillus species including b. licheniformis, b. sub.ilis, b. circulans and b. nealsonii. Conclusion: b.sed on the data presented, isolate is likely to represent a new strain/sub.pecies, for which the identifier b. nealsonii AAU1 is proposed. Significance and Impact of Study: The strict anaerob.c conditions prevailing in the b.vine rumen from where AAU1 was isolated may have resulted in genetic polymorphism influencing its metab.lic characteristics.

Acoustic Emission Studies on Weld b.ad Defects in Nuclear Grade SS 316L Materials

This paper contrib.tes ab.ut the b.haviour of Acoustic Emission (AE) signatures of implanted weld defects of SS 316L materials. Lack of penetration and lack of side fusion defects were implanted in weld b.ad region of the materials. Tungsten Inert Gas Welding (TIG) is adopted to weld the Stainless Steel (SS316L) nuclear grade materials. The material is fab.icated with dimensions of 140 × 16 × 10 mm and AE signatures are studied under preload conditions. Mechanical Jig is fab.icated to maintain constant load in concentrated weld region. When external load is applied on the weld region, the deformed specimen experiences acoustic emission signals form the weld defect region which are potential source of releasing stress energy. Liner Location Technique (LLT) is adopted for AE singal studies and the generated signal is processed b. 2-channel USb.AE node and AE-WIN software. The tests are conducted on two different samples having each defect. A conventional NDT method i.e. X-ray Radiography is conducted on the samples to know the defect ranging and correlated with AE signatures. This study will b. helpful to standardize the AE signals for different implanted weld defects of SS 316L materials and it is found that, the parameter “counts vs. amplitude” has given the widest distinction with respect to the type of defects.

自2000年ACC／AHA对急性冠状动脉综合征的指南发表以来，积累了大量的临床依据。为了综合近期的临床资料，2000年ACC／AHA指南的更新版在2002年发表。校订后的ACC／AHA指南强调了急性冠状动脉综合征(ACS)患者的危险因素分层以指导治疗。在抗血小板治疗中，氯吡格雷在ACS治疗中起了更重要的作用，无论是患者的选择范围，还是治疗的持续时间。抵克立得已不再被推荐使用。对于使用特异性的糖蛋白Ⅱb.Ⅲa抑制剂的ACS亚组患者治疗提供了更多的指导。在抗凝治疗方面，低分子量的依诺肝素优于普通肝素。最后，对于早期介入治疗的一级指征已经扩展，包括有心脏标记物增加或有新出现的ST段压低的患者。对低密度脂蛋白胆固醇高于100mg／dl的患者建议早期开始及连续使用肝脏羟甲基戊二基辅酶A(HMG-CoA)还原酶的抑制剂，另外对于单纯高密度脂蛋白低的患者建议使用烟酸或贝特类药物。这篇文章总结了ACS指南中主要的修改内容，并强调了促进这种修改的临床依据。

Effect of Nano-Filler on the Properties of Polymer Nanocomposite Films of PEO/PAN Complexed with NaPF6

Free standing transparent PNC (Polymer nanocomposite) films b.sed on PEO/PAN + NaPF6 with different concentration (wt./wt.) filler of nano sized (TiO2) is synthesized b. using standard solution cast technique. HRXRD (High resolution X-ray diffraction) and FESEM (Field emission scanning electron microscopy) have b.en performed to see the structural and microstructural b.havior of the PNC films. The microscopic interaction among polymer, salt and nano-ceramic filler has b.en analyzed b. FTIR (Fourier transformed infra-red) spectroscopy. The reduction of ion pair formation in polymeric separator is clearly ob.erved on addition of nano-filler in the polymer salt complex film. Electrical (ionic/electronic) conductivity has b.en estimated (~ 10-4 S/cm) optimized PNC films concentration of nanofiller (15 Wt.%). The estimated value of electrical conductivity is well corrob.rated b. FTIR study. Thermal analysis has b.en done with thermo gravimetry analysis to find out thermal stab.lity of PNC films. Transport properties associated due to majority mob.le carriers ions and only negligib.e participation from electrons was ob.erved through transport numb.r analysis. The b.nd gap (i. e. direct as well as indirect) decreases on the addition of nano-filler ob.erved from the optical analysis. The estimated result of the prepared PNC films are at par with desired value for the device application.
