Osteoblastic glucocorticoid signaling exacerbates high-fat-diet-induced bone loss and obesity

Chronic high-fat diet(HFD)consumption not only promotes obesity and insulin resistance,but also causes bone loss through mechanisms that are not well ***,we fed wild-type CD-1 mice either chow or a HFD(43%of energy from fat)for 18 weeks;HFD-fed mice exhibited decreased trabecular volume(-28%)and cortical thickness(-14%)compared to chow-fed *** HFD-fed mice,bone loss was due to reduced bone formation and mineral apposition,without obvious effects on bone *** feeding also increased skeletal expression of sclerostin and caused deterioration of the osteocyte lacunocanalicular network(LCN).In mice fed HFD,skeletal glucocorticoid signaling was activated relative to chow-fed mice,independent of serum corticosterone *** therefore examined whether skeletal glucocorticoid signaling was necessary for HFD-induced bone loss,using transgenic mice lacking glucocorticoid signaling in osteoblasts and osteocytes(HSD2^(OB/OCY)-tg mice).In HSD2^(OB/OCY)-tg mice,bone formation and mineral apposition rates were not suppressed by HFD,and bone loss was significantly ***,in HSD2^(OB/OCY)-tg mice fed HFD,both Wnt signaling(less sclerostin induction,increased Pmatched *** osteocyte LCN remained intact in HFD-fed HSD2^(OB/OCY)-tg *** fed a HFD,HSD2^(OB/OCY)-tg mice also increased their energy expenditure and were protected against obesity,insulin resistance,and ***,glucocorticoid signaling in osteoblasts and osteocytes contributes to the suppression of bone formation in HFD-fed *** glucocorticoid signaling is also an important determinant of glucose uptake in bone,which influences the whole-body metabolic response to HFD.

Improved Quantile Convolutional and Recurrent Neural Networks for Electric Vehicle Battery Temperature Prediction

The battery thermal management of electric vehicles can be improved using neural networks predicting quantile sequences of the battery *** work extends a method for the development of Quantile Convolutional and Quantile Recurrent Neural Networks(namely Q*NN).Fleet data of 225629 drives are clustered and balanced,simulation data from 971 simulations are augmented before they are combined for training and *** Q*NN hyperparameters are optimized using an efficient Bayesian optimization,before the Q*NN models are compared with regression and quantile regression models for four *** analysis of point-forecast and quantile-related metrics shows the superior performance of the novel Q*NN *** median predictions of the best performing model achieve an average RmSE of 0.66°C and R^(2) of *** predicted 0.99 quantile covers 98.87%of the true values in the test *** conclusion,this work proposes an extended development and comparison of Q*NN models for accurate battery temperature prediction.

A review of control strategies used for morphing aircraft applications

This paper reviews the various control algorithms and strategies used for fixed-wing morphing aircraft applications. It is evident from the literature that the development of control algorithms for morphing aircraft technologies focused on three main areas. The first area is related to precise control of the shape of morphing concepts for various flight conditions. The second area is mainly related to the flight dynamics, stability, and control aspects of morphing aircraft. The third area deals mainly with aeroelastic control using morphing concepts either for load alleviation purposes and/or to control the instability boundaries. The design of controllers for morphing aircraft/wings is very challenging due to the large changes that can occur in the structural, aerodynamic, and inertial characteristics. In addition, the type of actuation system and actuation rate/speed can have a significant effect on the design of such controllers. The aerospace community is in strong need of such a critical review especially as morphing aircraft technologies move from fundamental research at a low Technology Readiness Level(TRL) to real-life applications. This critical review aims to identify research gaps and propose future directions. In this paper, research activities/papers are categorized according to the control strategy used. This ranges from simple Proportional Integral Derivative(PID) controllers at one end to complex robust adaptive controllers and deep learning algorithms at the other end. This includes analytical, computational, and experimental studies. In addition, the various dynamic models used and their fidelities are highlighted and discussed.

Evaluation of a hepatic biomarker of nutritional imbalance in juvenile red drum(Sciaenops ocellatus) fed 60% soybean meal-based diets using NmR-based metabolomics

A 12-week feeding trial with juvenile red drum(Sciaenops ocellatus) fed high-soybean meal(SBm) diets was conducted to investigate a putative biomarker of nutritional imbalance, N-formimino-L-glutamate(FIGLU). Three fishmeal-free, 60% SBm pelleted diets(named B12, Fol, and met, respectively) were tested to evaluate the effects on growth performance and tissue metabolite profiles of supplementation of vitamin B12(0.012 mg/kg), folate(10 mg/kg), methionine(1 g/kg) respectively, above basal supplementation levels. A fourth SBm-based diet(named B12/Fol/met) was formulated with a combination of B12, folate, and methionine to attain the above-mentioned target concentrations. A fifth 60% SBm diet(named FWS) with methionine supplementation(1 g/kg above basal supplementation levels), enriched with taurine, lysine and threonine as well as minerals, was also tested. This diet contained formulation targets and additives which have allowed for replacing fishmeal with plant proteins in rainbow trout feeds. Control diets included a fishmeal-based diet(named Fm), an unsupplemented basal 60% SBm diet(named SBm60), and a “natural” diet(named N) made up of equal parts of fish(cigar minnows), squid and shrimp as a positive reference for growth performance. Formulated feeds contained approximately 37% total crude protein, approximately 14% total crude lipid and were energetically balanced. Standard growth performance metrics were measured, and tissues(liver, muscle) were collected at week 12 to evaluate diet-induced metabolic changes using nuclear magnetic resonance(NmR)-based *** results show that the FWS diet outperformed all other SBm diets and the Fm diet under all performance metrics(Pm diets and the Fm diet. On Perron’s Formula and the Prime Numbers

The Riemann hypothesis is intimately connected to the counting functions for the primes. In particular, Perron’s explicit formula relates the prime counting function to fixed points of iterations of the explicit formula with particular relations involving the trivial and non-trivial roots of the Riemann Zeta function and the Primes. The aim of the paper is to demonstrate this relation at the fixed points of iterations of explicit formula, defined by functions of the form limT∈Ν→∞fT(zw)=zw,where, zwis a real number.

High quality micro mould tools are critical for ensuring defect-free production of micro injection moulded *** demoulding stage of the micro injection moulding can adversely affect the surface integrity due to friction,adhesion and thermal stresses between the metallic mould and polymeric replicated *** the present work,we propose the use of precision electropolishing(EP)as a shaping and polishing process to control the draft angle and fillet radius of micro features in order to ease *** defects that occur in replicated polymer parts include cracks,burrs and distorted features.A nickel mould having multiple linear ridges and star shape patterns was designed for the present investigation to have characteristic dimensions ranging from 10μm to 150μm and with various aspect ratios to study the effect of electropolishing on modifying the shape of micro features and surface morphology.A transient 2D computational analysis has been conducted to anticipate the effect of shaping on the Ni mould after electrochemical polishing with non-uniform material removal rates,based on the distribution of current *** experimental results indicate that after shaping using EP,the draft angle of star-patterns and linear patterns can be effectively increased by approximately 3.6°,while the fillet radius increases by up to 5.0μ*** controlling the electropolishing process,the surface roughness can be maintained under 50 *** work uses a green and environmental friendly nickel sulfamate electrolyte which can be effective for shaping of nickel micro features without causing any surface deposition.
