l-glutamic acid(lA)is a bio-based,non-toxic,environmentally friendly material derived from *** present study reports the application of Passerini three-component polymerization(P-3CP)for the straightforward preparation of lA-based light-responsive polyesters(PlTDs)under mild *** with molar masses up to 8500 g/mol and high yields exceeding 90%are *** chemical structures and light-responsive self-immolative behavior of PlTDs are comprehensively characterized by employing ultraviolet-visible(UV-Vis)spectroscopy,size exclusion chromatography(SEC),nuclear magnetic resonance(NMR)spectroscopy,and liquid chromatography mass spectrometry(lC-MS).Meanwhile,monodisperse PlTD-based doxorubicin-loaded nanoparticles(PlTD-DOX-NP)(size=193 nm,PDI=0.018)are formulated by nanoprecipitation *** light-induced depolymerization,the PlTD-DOX-NP undergoes rapid decomposition,resulting in a burst release of 80%cargo within 13 ***,according to biological toxicity tests,the PlTD-NP possesses adequate biosafety,both before and after ***,the incorporation of P-3CP with biorenewable lA-based monomer adheres to the principles of green chemistry,significantly simplifying the synthetic pathway of light-responsive polymers.

The seed storage materials accumulate during seed development,and are essential for seed germination and seedling *** we employed two bi-parental populations of an F2:3 population developed from a cross of improved 220(I220,small seeds with low starch)and PH4CV(large seeds with high starch),as well as recombinant-inbred lines(RIls)of X178(high starch)and its improved introgression line I178(low starch),to identify the genes that control seed storage *** identified a total of 12 QTls for starch,protein and oil,which explained 3.44-10.79%of the phenotypic *** them,qSTA2-1 identified in F2:3 and qSTA2-2 identified in the RIls partially overlapped at an interval of 7.314-9.554 Mb,and they explained 3.44-10.21%of the starch content variation,so they were selected for further *** mapping of qSTA2-2 with the backcrossed populations of ^(I220)/PH4CV in each generation narrowed it down to a 199.7 kb interval that contains 14 open reading frames(ORFs).Transcriptomic analysis of developing seeds from the near-isogenic lines(NIls)of ^(I220)/PH4CV(BC_(5)F_(2))showed that only 11 ORFs were expressed in 20 days after pollination(DAP)*** of them were upregulated and six of them were downregulated in NIl^(I220),and the differentially expressed genes(DEGs)between NIl^(I220) and NIl^(PH4CV) were enriched in starch metabolism,hormone signal transduction and glycosaminoglycan *** the eleven NIl^(I220) differential expressed ORFs,ORF4(Zm00001d002260)and ORF5(Zm00001d002261)carry 75%protein sequence similarity,both encodes an glycolate oxidase,were the possible candidates of *** analysis and validation indicated that mutation of the qSTA2-2 locus resulted in the dysfunction of ABA accumulation,the embryo/endosperm ratio and the starch and hormone levels.

Nitrogen(N)priming is a microbially mediated biochemical process as affected by different incorporation ***,little information is known about the microbial mechanisms driving the response of N priming to co-operation of Chinese milk vetch(CMV,Astragalus sinicus l.)and different rates of chemical fertilizers in paddy soils in South ***,an anaerobic incubation experiment was conducted to study N priming effects(PE)and their relationships with soil microbial functional genes after CMV incorporation alone(M),co-incorporation of CMV with 100%(normal dosage)chemical fertilizers(MC100),and co-incorporation of CMV with 80% chemical fertilizers(MC80).Co-incorporation of CMV and chemical fertilizers enhanced the short-time scale(the first 20 d of incubation)positive PE of N,while no significant differences existed among the three treatments on day 60 or 90 of incubation(P>0.05).Compared with the M treatment,gross priming effect(GPE)in the MC100 and MC80 treatments significantly increased by 34.0% and 31.3%,respectively,and net priming effect(NPE)increased by 47.7% and 47.8%,respectively,during the first 20 d of incubation(Plikely attributed to soil nutrient availability and added substrate *** MC100 and MC80 treatments increased the gdhA gene abundance by 5.0% and 9.8%,increased the gdh2 gene abundance by 12.7% and 45.7%,and increased the nasB gene abundance by 9.5% and 41.4%,respectively,in comparison with the M treatment on day 20 of *** analyses indicated that soil microbial functional genes involved in N mineralization(gdhA and gdh2),assimilatory nitrate reduction(nasB),and nitrification(amoB)were significantly correlated with N priming under different incorporation practices during the incubation period(Pl fertilizers can regulate soil microbial community functional gene structure,which may accelerate mineralization and assimilatory nitrate reduction and inhibit

Peanut(Arachis hypogaea l.)is a globally important oil *** blotch is one of the most important foliar diseases affecting peanut,which results in serious yield losses *** web blotch-resistant peanut varieties is the most effective and economically viable method for minimizing yield losses due to web *** the current study,a bulked segregant analysis with next-generation sequencing was used to analyze an F2:3 segregating population and identify candidate loci related to web blotch *** on the fine-mapping of the candidate genomic interval using kompetitive allele-specific PCR(KASP)markers,we identified a novel web blotch resistance-related locus spanning approximately 169 kb on chromosome *** region included four annotated genes,of which only Arahy.35VVQ3 had a non-synonymous single nucleotide polymorphism in the coding region between the two *** markers(Chr.16.12872635 and Chr.16.12966357)linked to this gene were shown to be co-segregated with the resistance of peanut web blotch by 72 randomly selected recombinant inbred lines(RIl),which could be used in marker-assisted breeding of resistant peanut varieties.

Immunosuppressive microenvironments present critical problems in clinical *** regulate the tumor immune microenvironment for enhancing antitumor effect,a combination of immune checkpoint inhibitors(ICIs)with chemotherapeutics has been applied *** this study,miriplatin(MiPt),the lipidic derivative of 5-fluorouracil(Fu-OA),as well as the programmed death ligand 1(PD-l1)target si RNA(siPD-l1)were integrated into lip-Pt/Fu@siPD-l1 nanoparticles(NPs)for *** vitro results showed that lip-Pt/Fu@siPD-l1 NPs could exhibit effective siRNA gene silencing and promote the phagocytosis of tumor cells by ***,in vivo results revealed that lipPt/Fu@siPD-l1 NPs showed significantly higher anti-tumor efficiency than that of the physical mixing of Mi Pt,5-fluorouracil,and lip@siPD-l1 NPs(delivery of siPD-l1 by liposomes).The best anti-tumor efficiency of lip-Pt/Fu@siPD-l1 NPs resulted from the synergistic immunotherapeutic effects of Mi Pt and siPD-l1 based on the inhibition of CD47 expression and the downregulation of PD-l1 in tumor cells,which elicited a robust anti-tumor immune response through the activation of macrophage phagocytosis and immune checkpoint *** lip-Pt/Fu@siPD-l1 NPs provide a potential strategy for tumor chemo-immunotherapy.

The aim of this study was to investigate the oxidation reactivity and behavior of exhaust particulate matter(PM)from diesel *** samples from two diesel engines(1K,CY4102)with different emission levels were collected by a thermophoretic system and a quartz *** oxidation reactivity,oxidation behaviors,and physicochemical properties of the PM samples were analyzed using thermogravimetric analysis(TGA),high-resolution transmission electron microscopy(HRTEM),Fourier-transform infrared spectrometry(FTIR),and Raman *** results showed that there was a great difference in the oxidation reactivity of soot particles emitted by the two different diesel engines.A qualitative analysis of the factors influencing oxidation reactivity showed that the nanostructure,degree of graphitization,and relative concentration of aliphatic C—H functional groups were the most important factors,whereas no significant correlation was found between the primary particle size and activation energy of the diesel *** on the oxidation behavior analysis,the diesel soot particles exhibited both internal and surface oxidation modes during the oxidation *** oxidation was dominant during the initial stage,and as oxidation progressed,the mode gradually changed to internal *** oxidation mode of soot particles from the 1K engine was significantly higher than that of CY4102.

Due to the complicated metabolic and regulatory networks of l-serine biosynthesis and degradation,microbial cell factories for l-serine production using non-model microorganisms have not been *** this study,a combination of synthetic biology and process optimization were applied in an ethanologenic bacterium Zymomonas mobilis for l-serine *** blocking the degradation pathway while introducing an exporter EceamA from ***,l-serine titer in recombinant *** was increased from 15.30 mg/l to 62.67 mg/*** was further increased to 260.33 mg/l after enhancing the l-serine biosynthesis ***,536.70 mg/l l-serine was achieved by removing feedback inhibition with a SerA mutant,and an elevated titer of 687.67 mg/l was further obtained through increasing serB copies while enhancing the ***,855.66 mg/l l-serine can be accumulated with the supplementation of the glutamate *** work thus not only constructed an l-serine producer to help understand the bottlenecks limiting l-serine production in *** for further improvement,but also provides guidance on engineering non-model microbes to produce biochemicals with complicated pathways such as amino acids or terpenoids.

Major volcanic eruptions(MVEs)have attracted increasing attention from the scientific *** studies have explored the climatic impact of MVEs over the past two ***,proxy-based reconstructions and climate model simulations indicate divergent responses of global and China’s regional climates to ***,we used multiple data from observations,reconstructions,simulations,and assimilations to summarize the historical facts of MVEs,the characteristics and mechanisms of their climatic impact,and directions for future *** reviewed volcanic datasets and determined intensive MVE periods;these periods corresponded to the years 530–700,1200‒1460,and 1600‒1840 *** tropical MVEs,a substantial cooling effect is observed throughout the globe and China on the interannual-interdecadal time scales but an inconsistent cooling magnitude is detected between reconstructions and *** the first summer after tropical MVEs,a decrease in global and monsoonal precipitation is *** reconstructions and simulations,an increased precipitation is seen for the Yangtze River Basin,while large uncertainties in precipitation changes are present for other regions of *** drought can be induced by frequent eruptions and volcanism superimposed on low solar irradiation and internal *** affect climate directly through the radiative effect and indirectly by modulating internal variability,such as the El Niño‒Southern Oscillation(ENSO)and Atlantic Multidecadal Oscillation(AMO).However,changes in the phase,amplitude,and periodicity of ENSO and AMO after MVEs and the associated mechanisms remain controversial,which could account for model-reconstruction ***,other internal variability,uncertainties in reconstruction methods and aerosol‒climate models,and climate background may also induce model-reconstruction *** gaps and directions for future research are also discussed.

Adeno-associated virus(AAV)has emerged as a pivotal delivery tool in clinical gene therapy owing to its minimal pathogenicity and ability to establish long-term gene expression in different *** AAV(rAAV)has been engineered for enhanced specificity and developed as a tool for treating various ***,as rAAV is being more widely used as a therapy,the increased demand has created challenges for the existing manufacturing *** rAAV-based gene therapy products have received regulatory approval,but there continue to be concerns about safely using high-dose viral therapies in humans,including immune responses and adverse effects such as genotoxicity,hepatotoxicity,thrombotic microangiopathy,and *** this review,we explore AAV biology with an emphasis on current vector engineering strategies and manufacturing *** discuss how rAAVs are being employed in ongoing clinical trials for ocular,neurological,metabolic,hematological,neuromuscular,and cardiovascular diseases as well as *** outline immune responses triggered by rAAV,address associated side effects,and discuss strategies to mitigate these *** hope that discussing recent advancements and current challenges in the field will be a helpful guide for researchers and clinicians navigating the ever-evolving landscape of rAAV-based gene therapy.
