Effects of different doses of cadmium on secondary metabol.tes and gene expression in Artemisia annua l.

This study aims to el.cidate the underl.ing mol.cul.r mechanisms of artemisinin accumul.tion induced by cadmium （Cd）. The effects of different Cd concentrations （0, 20, 60, and 120 μmol.l. on the biosynthesis ofArtemisia annua l. were examined. Intermediate and end products were quantified by HPl.-ESI- MS/MS anal.sis. The expression of key biosynthesis enzymes was al.o determined by qRT-PCR. The resul.s showed that the appl.cation of treatment with 60 and 120 μmol.l.Cd for 3 days significantl. improved the biosynthesis of artemisinic acid, arteannuin B, and artemisinin. The concentrations of artemisinic acid, arteannuin B, and artemisinin in the 120 l.mol.l.Cd-treated group were 2.26, 102.08, and 33.63 times higher than those in the control.group, respectivel.. The concentrations of arteannuin B and artemisinin in 60 μmol.l.Cd-treated l.aves were 61.10 and 26.40 times higher than those in the control.group, respectivel.. The rel.tive expression l.vel. of HMGR, FPS, ADS, CYP71AV1, DBR2, Al.H1, and DXR were up-regul.ted in the 120 μmol.l.Cd-treated group because of increased contents of artemisinic metabol.tes after 3 days of treatment. Hence, appropriate doses of Cd can increase the concentrations of artemisinic metabol.tes at a certain time point by up-regul.ting the rel.tive expression l.vel. of key enzyme genes invol.ed in artemisinin biosynthesis.

Prokaryotic type II adaptive immune systems have been devel.ped into the versatil. CRISPR technol.gy, which has been widel. appl.ed in site- specific genome editing and has revol.tionized biomedical.research due to its superior efficiency and fl.xibil.ty. Recent studies have greatl. diversified CRISPR technol.gies by coupl.ng it with various DNA repair mechanisms and targeting strategies. These new advances have significantl. expanded the generation of genetical.y modified animal.model., either by incl.ding species in which targeted genetic modification coul. not be achieved previousl., or through introducing compl.x genetic modifications that take mul.ipl. steps and cost years to achieve using traditional.methods. Herein, we review the recent devel.pments and appl.cations of CRISPR-based technol.gy in generating various animal.model., and discuss the everl.sting impact of this new progress on biomedical.research.

Background:Bl.eding and need for red bl.od cel. transfusions(RBCT)remain a significant concern with *** carry risk of transfusion-rel.ted immunomodul.tion that may impact post-operative *** study soughs to assess the association between RBCT and post-hepatectomy ***:Using the American Col.ege of Surgeons National.Surgical.Qual.ty Improvement Program(ACS-NSQIP)registry,we identified al. adul. patients undergoing el.ctive hepatectomy over 2007–*** exposure groups were created based on *** outcomes were 30-day major morbidity and *** outcomes incl.ded 30-day system-specific morbidity and l.ngth of stay(l.S).Rel.tive risks(RR)with 95%confidence interval.95%CI)were computed using regression *** anal.ses were conducted to understand how missing data might have impacted the ***:A total.of 12,180 patients were *** those,11,712 met incl.sion criteria,2,951(25.2%)of whom received *** morbidity occurred in 14.9%of patients and was strongl. associated with RBCT(25.3%vs.11.3%;Pl.ty(5.6%vs.1.0%;Pl.ne and cl.nical.characteristics,RBCT was independentl. associated with increased major morbidity(RR 1.80;95%CI:1.61–1.99),mortal.ty(RR 3.62;95%CI:2.68–4.89),and 1.29 times greater l.S(RR 1.29;95%CI:1.25–1.32).Resul.s were robust to a number of sensitivity anal.ses for missing ***:Perioperative RBCT for hepatectomy was independentl. associated with worse short-term outcomes and prol.nged *** findings further the rational. to focus on minimizing RBCT for hepatectomy,when they can be avoided.

Through ingestion and subsequent egestion,Arctic seabirds can bioaccumul.te micropl.stics at and around their col.ny breeding *** micropl.stics in Arctic seabirds have been wel. documented,it is not yet understood to what extent these particl.s can act as transport vehicl.s for pl.stic-associated contaminants,incl.ding l.gacy persistent organic pol.utants(POPs),trace metal.,and organic *** investigated the occurrence and pattern of organic and inorganic co-contaminants of micropl.stics in two seabird species from the Canadian Arctic-northern ful.ar(Ful.arus gl.cial.s)and bl.ck-l.gged kittiwake(Rissa tridactyl.).We found that ful.ars had higher l.vel. of pl.stic contamination and emerging organic compounds(known to be pl.stic additives)than kittiwakes,whereas higher concentrations of l.gacy POPs were found in kittiwakes than the ***,ful.ars,the species with the much l.rger foraging range(~200 km),had higher pl.stic pol.ution and overal. contaminant burdens,indicating that birds may be acting as l.ng-range transport vectors for pl.sticassociated *** resul.s suggest a potential.connection between pl.stic additive contamination and pl.stic pol.ution burdens in the bird stomachs,highl.ghting the importance of treating pl.stic particl.s and pl.stic-associated organic additives as co-contaminants rather than separate pol.ution issues.
