rANK-ligand and osteoprotegerin as biomarkers in the differentiation between periprosthetic joint infection and aseptic prosthesis loosening

AIM To assess serum levels of rANK-ligand(rANKL) and osteoprotegerin(OPG) as biomarkers for periprosthetic joint infection(PJI) and compare their accuracy with standard *** One hundred and twenty patients presenting with a painful total knee or hip arthroplasty with indication for surgical revision were included in this prospective clinical trial. Based on standard diagnostics(joint aspirate, microbiological, and histological samples) and Musculoskeletal Infection Society consensus classification,patients were categorized into PJI, aseptic loosening,and control groups. Implant loosening was assessed radiographically and intraoperatively. Preoperative serum samples were collected and analyzed for rANKL, OPG, calcium, phosphate, alkaline phosphatase(AP), and the bone-specific subform of AP(b AP). Statistical analysis was carried out, testing for significant differences between the three groups and between stable and loose implants. reSULTS All three groups were identical in regards to age, gender, and joint distribution. No statistically significant differences in the serum concentration of rANKL(P = 0.16) and OPG(P = 0.45) were found between aseptic loosening and PJI, with a trend towards lower rANKL concentrations and higher OPG concentrations in the PJI group. The rANKL/OPG ratio was significant for the comparison between PJI and non-PJI(P = 0.005). A ratio > 60 ruled out PJI in all cases(specificity: 100%, 95%CI: 89, 11% to 100.0%) but only 30% of non-PJI patients crossed this threshold. The positive predictive value remained poor at any cut-off. In the differentiation between stable and loose implants, none of the parameters measured(calcium, phosphate, AP, and b AP) showed a significant difference, and only AP and b AP measurements showed a tendency towards higher values in the loosened group(with P = 0.09 for AP and P = 0.19 for b AP). CONCLUSION Lower rANKL and higher OPG concentrations could be detected in PJI, without statistical significance.

Slow strain rate testing （SSrT） was employed to study the stress corrosion cracking （SCC） behavior of Ze41 magnesium alloy in 0.01 M NaCl solution. Smooth tensile specimens with different thicknesses were strained dynamically in both longitudinal and transverse direction under permanent immersions at a strain rate of 10-6 s-1. It is found that Ze41 magnesium alloy is susceptible to SCC in 0.01 M NaCl solution. The SCC susceptibility of the thinner specimen is lower than that of the thicker specimen. Also, the longitudinal specimens are slightly more susceptible to SCC than the transverse specimens. The SCC mechanism of magnesium alloy is attributed to the combination of anodic dissolution with hydrogen embrittlement.

OI 630.0 nm airglow observations,from a new observatory at Bom Jesus de Lapa,were used to study the interaction between ePBs(equatorial Plasma Bubbles)and the MSTID(Medium-Scale Traveling Ionospheric Disturbance)over the Northeast region in *** the night of September 16 to 17,2020,an ePB was observed propagating eastward,in an apparent fossil stage,until it interacted with a dark band electrified MSTID(e MSTID).After the interaction,four ePBs merged,followed by an abrupt southward development and *** of the data suggests that an eastward polarization electric field,induced by the dark band e MSTID,forced the ePB into an upward drift,growing latitudinally along the magnetic field lines and then bifurcating.

A partial rice (Oryza sativa L.) cDNA clone. OsPMK1c, was isolated through screening of a cDNA library constructed from tillering materials. OsPI4Klc encoded a peptide of 608 amino acids with a calculated molecular mass of 68.4 kDa. The OsPI4Klc peptide shared high homology and possessed the highly conserved domains present in most isolated cloned PI4-kinases, i.e. a lipid kinase unique (LKU) domain and a catalytic (CAT) domain. A region with similarity to pleckstrin homology (PH) domain was present in OsPI4K1c as well. Further comparison with genomic sequences in databases revealed that OsPI4K1c is located at the 3'-end of a putative rice PI 4-kinase coding gene OsPI4K1, and its coding region corresponded to the C-terminal half of OsPI4Kl protein. Twelve exons (49-562 bp in size) and 11 introns (77-974 bp in size) were identified in OsPI4K1c. Nutrient deficiency,especially bioavailable nitrogen deficiency,often impedes the bioremediation efforts of mining generated *** nitrogen-fixation is a critical process necessary for the initial nitrogen buildup in *** knowledge regarding the diazotrophs that inhabit tailings is still in its ***,in this study,a comprehensive investigation combining geochemical characterization,sequence analyses,molecular techniques,and activity measurements was conducted to characterize the diazotrophic community residing in tailing *** differences between tailings and their adjacent soils in prokaryotic and diazotrophic communities were ***,strong and significant correlations between the absolute abundance of the nitrogen fixation(nif H),carbon fixation(cbb L),sulfur oxidation(sox B),and arsenite oxidation(aio A) genes were observed in the tailings,but not in the *** reconstructed nif-containing metagenome-assembled genomes(MAGs) suggest that the carbon fixation and sulfur oxidation pathways were important for potential diazotrophs inhabiting the *** measurements further confirmed that diazotrophs inhabiting tailings preferentially to use the inorganic electron donors(e.g.,elemental sulfur) compared to organic electron donors(e.g.,sucrose),while diazotrophs inhabiting soils preferred organic carbon ***,these findings suggest that chemolithoautotrophic diazotrophs may play essential roles in acquiring nutrients and facilitating ecological succession in tailings.

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous *** assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous *** species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding *** a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid *** a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic *** StCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of StCDF1 are correlated with maturity in a dosage-dependent *** of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.
