The safety and efficacy of palbociclib in older patients with advanced breast cancer in a real-world setting

Aims:Palbociclib has been approved in combination with endocrine therapy(ET)for hormone receptor-positive(HR+)/human epidermal growth factor receptor 2-negative(HER2-)advanced breast cancer(ABC),regardless of *** though ABC is one of the most prevalent cancers in older patients,very few patients≥65 years old were included in pivotal ***,the current study evaluated the safety and efficacy of palbociclib in“real-world”routine treatment of unselected older patients with HR+/***:Data were collected on patients>70 years old who were treated with palbociclib plus ET for HR+/HER2-ABC in our *** analyzed safety data(CTCAE v4.0 criteria)and outcomes,such as progression-free survival(PFS)and overall survival(OS),as well as any associations between main geriatric characteristics and our ***,we assessed safety at a national level by analyzing all palbociclib-related adverse events(AEs)reported in the French Pharmacovigilance Database(FPVD)during the same ***:From February 2016 to July 2019,52 patients were identified with a median age of 80.9 years,of whom 88%presented an *** most common grade 3-4 AE was neutropenia(64%).Median PFS and OS were nine months and not reached,*** FPVD reports 227 cases of palbociclib-related AEs,with older and younger patients sharing similar ***:Palbociclib is well tolerated in older patients with efficacy comparable to that in younger ***,the addition of palbociclib to ET should be evaluated individually in this older and frailer subgroup.

Spatio-temporal variability and dynamics in Sahelian agro-pastoral zones make each local situation a special case. These specificities must be considered to guide the dissemination of agricultural options with a view to sustainable development. The territorial scale of municipalities is not sufficient for this necessary contextualization;the scale of the “village terroir” seems to be a better option. This is the hypothesis we put forward in the framework of the Global Collaboration for Resilient Food Systems program (CRFS), i.e. local context is spatially defined by village terroir. The study is based on data collected through participatory mapping and surveys in “village terroirs” in three regions of Niger (Maradi, Dosso and Tillabéri). Then the links between farm managers and their cultivated land, as well as the spatio-temporal dynamics of local context are analyzed. This study provides evidence of the existence and functional usefulness of the village terroir for farmers, their land management and their activities. It demonstrates the usefulness of contextualizing agricultural options at this scale. Their analysis elucidates the links between “terroirs village” and the specific functioning of the agrosocio-ecosystems acting on each of them, thus laying the systemic and geographical foundations for a model of the spatio- temporal dynamics of “village terroirs”. This initial work has opened up new perspectives in modeling and sustainable development.

Adsorption Behavior of Reducing End-Modified Cellulose Nanocrystals:A Kinetic Study Using Quartz Crystal Microbalance

In this work,we studied the adsorption of modified cellulose nanocrystals onto solid surfaces by quartz crystal microbalance with dissipation monitoring(QCM-D).Cellulose nanocrystals obtained from tunicate(CNC)were modified at reducing end by amidation *** different functionalities were investigated:a polyamine dendrimer(CNC-NH_(2)),which interacts with gold surface by the amine groups;and a biotin moiety(CNC-Biot),which has a strong affinity for the protein streptavidin(SAV).QCM-D results revealed different adsorption behaviors between modified and unmodified ***,unmodified CNCs covered almost all the surface forming a rigid and flat layer whereas reducing end modified CNCs remained rather upright forming a hydrated and viscoelastic layer with lower surface *** analysis of adsorption kinetics allowed the calculation of an apparent collision rate factor,which resulted 10-fold higher for unmodified CNCs compared to reducing end modified CNCs,therefore,demonstrating the different adsorption behavior.

BACKGROUND Patients with paroxysmal nocturnal hemoglobinuria(PNH)have a clonal population of blood cells deficient in glycosylphosphatidylinositol-anchored(GPIanchored)proteins,most of the time resulting from a mutation in the X-linked gene *** report a patient with PNH resulting from a rare biallelic PIGT mutation on chromosome *** SUMMARY A 47-year-old man was referred to our hospital for febrile *** patient reported a history of recurrent urticaria and arthralgia and he presented during 3 mo recurrent acute dermo-hypodermitis and aseptic *** on clinical cases published with PIGT-PNH,with clinically typical PNH and autoinflammatory symptoms,we treated our patients with repeated infusions of eculizumab to decrease autoinflammatory symptoms and then we performed an allogeneic stem cell transplantation(allo-SCT)with a mismatched unrelated *** patient experienced no acute Graft vs Host disease(GvHD)and a moderate chronic GvHD and is now considered cured at 24 mo after *** This case report suggests that allo-SCT should be considered to cure PIGT-PNH patients.
