Adult bone structural integrity is maintained by remodeling via the coupling of osteoclastic bone resorption and osteoblastic bone *** or osteoblasts express receptor activator of nuclear factor k-B ligand(Rankl)or osteoprotegerin(Opg)to promote or inhibit osteoclastogenesis,*** morphogenetic protein(BMP)is a potent bone inducer,but its major role in adult bone is to induce osteocytes to upregulate sclerostin(Sost)and increase the Rankl/Opg expression ratio,resulting in promotion of ***,the precise effect of BMP-target gene(s)in osteoblasts on the Rankl/Opg expression ratio remains *** the present study,we identified atonal homolog 8(Atoh8),which is directly upregulated by the BMPSmadl axis in *** vivo,Atoh8 was detected in osteoblasts but not osteocytes in adult *** global Atoh8-knockout mice showed only a mild phenotype in the neonate skeleton,the bone volume was decreased and osteoclasts were increased in the adult ***8-null marrow stroma cells were more potent than wild-type cells in inducing osteoclastogenesis in marrow ***8 loss in osteoblasts increased Runx2 expression and the Rankl/Opg expression ratio,while Runx2 knockdown normalized the Rankl/Opg expression ***,Atoh8 formed a protein complex with Runx2 to inhibit Runx2 transcriptional activity and decrease the Rankl/Opg expression *** results suggest that bone remodeling is regulated elaborately by BMP signaling;while BMP primarily promotes bone resorption,it simultaneously induces Atoh8 to inhibit Runx2 and reduce the Rankl/Opg expression ratio in osteoblasts,suppressing osteoclastogenesis and preventing excessive BMP-mediated bone resorption.

Gradation representation method using a binary-weighted computer-generated hologram based on pulse-width modulation

We propose a simple gradation representation method using a binary-weighted computer-generated hologram（CGH） to be displayed on a high-speed spatial light modulator that can be controlled by the pulse-width modulation technique. The proposed method uses multiple bit planes comprising binary-weighted CGHs with various pulse widths. The object points of a three-dimensional（3D） object are assigned to multiple bit planes according to their gray levels. The bit planes are sequentially displayed in a time-division-multiplexed ***, the proposed method realizes a gradation representation of a reconstructed 3D object.

The ability of human deciduous tooth dental pulp cells（HDDPCs） to differentiate into odontoblasts that generate mineralized tissue holds immense potential for therapeutic use in the field of tooth regenerative medicine. Realization of this potential depends on efficient and optimized protocols for the genetic manipulation of HDDPCs. In this study, we demonstrate the use of a Piggy Bac（PB）-based gene transfer system as a method for introducing nonviral transposon DNA into HDDPCs and HDDPC-derived inducible pluripotent stem cells. The transfection efficiency of the PB-based system was significantly greater than previously reported for electroporation-based transfection of plasmid DNA. Using the neomycin resistance gene as a selection marker, HDDPCs were stably transfected at a rate nearly 40-fold higher than that achieved using conventional methods. Using this system, it was also possible to introduce two constructs simultaneously into a single cell. The resulting stable transfectants, expressing td Tomato and enhanced green fluorescent protein, exhibited both red and green fluorescence. The established cell line did not lose the acquired phenotype over three months of culture. Based on our results, we concluded that PB is superior to currently available methods for introducing plasmid DNA into HDDPCs. There may be significant challenges in the direct clinical application of this method for human dental tissue engineering due to safety risks and ethical concerns. However, the high level of transfection achieved with PB may have significant advantages in basic scientific research for dental tissue engineering applications, such as functional studies of genes and proteins. Furthermore, it is a useful tool for the isolation of genetically engineered HDDPC-derived stem cells for studies in tooth regenerative medicine.

Characteristic pathological findings and effects of ecabet sodium in rat reflux esophagitis

AIM:To explore the pathological flndings in the entire esophagus in rats with reflux esophagitis, and the effects of ecabet sodium(ES).METHODS:A rat model of chronic acid reflux esophagitis was *** the treatment group, ES was administered after surgery(n = 16).No drug was administered postoperatively to the esophagitis group(n = 9).Shamoperated rats were used as a control group(n = 5).Rats were sacriflced on day 7 after the *** epithelial thickness and leukocyte inflltration were examined in the upper, middle and lower areas of the *** survival rate, incidence of esophageal ulcer, and mean surface area and number of esophageal ulcers were determined in the esophagitis and ES *** histology was assessed in all three ***:Leukocyte infiltration in the esophagitis group was 26.3 ± 22.0 in the middle esophagus and 8.2 ± 4.9 in the upper esophagus, which was signiflcantly greater than that in the controls(1.3 ± 1.1 and 1.4 ± 1.0, respectively)(P < 0.05).The thickness of the epithelium in the esophagitis group was 210.8 ± 47.7 μm in the lower esophagus and 204.2 ± 60.1 μm in the middle esophagus, which was significantlygreater than that in the controls(26.0 ± 5.5 and 21.0 ± 6.5 μm, respectively)(P < 0.05).The mean number of ulcers per animal in the ES group in the entire esophagus was 5.4 ± 2.5, which was signiflcantly less than that in the esophagitis group(9.0 ± 3.5)(P < 0.05).The epithelial thickness in the ES group was 97.5 ± 32.2 μm in the lower esophagus, which was decreased compared with that in the esophagitis group(210.8 ± 47.7 μm)(P < 0.05).CONCLUSION:Mucosal inflammation extended to the upper esophagus close to the *** study suggested that ES may have a useful defensive role in reflux esophagitis.

FoxP3^(+)regulatory T cells(Tregs)play crucial roles in peripheral immune *** addition,Tregs that reside or accumulate in nonlymphoid tissues,called tissue Tregs,exhibit tissue-specific functions and contribute to the maintenance of tissue homeostasis and *** an experimental mouse model of crescentic glomerulonephritis induced by an anti-glomerular basement membrane antibody,Tregs started to accumulate in the kidney on day 10 of disease onset and remained at high levels(~30–35%of CD4^(+)T cells)during the late stage(days 21–90),which correlated with stable disease *** depletion on day 21 resulted in the relapse of renal dysfunction and an increase in Th1 cells,suggesting that Tregs are essential for disease control during the convalescence *** Tregs that accumulated in the kidney showed tissue Treg phenotypes,including high expression of GATA3,ST2(the IL33 receptor subunit),amphiregulin(Areg),and PPARγ.Although T-bet^(+) Tregs and RORγt^(+) Tregs were observed in the kidney,GATA3^(+) Tregs were predominant during the convalescence stage,and a PPARγagonist enhanced the accumulation of GATA3^(+) Tregs in the *** understand the function of specific genes in kidney Tregs,we developed a novel T cell transfer system to T cell-deficient *** experiment demonstrates that ST2,Areg,and CCR4 in Tregs play important roles in the accumulation of GATA3^(+) Tregs in the kidney and in the amelioration of renal *** data suggest that GATA3 is important for the recruitment of Tregs into the kidney,which is necessary for convalescence after renal tissue destruction.

Effect of Parameters on Potential Induced Degradation of Solar Cell

Solar cells are widely used to generate electric energy even at homes. It surely has a great advantage of sustainability. However, the potential induced degradation has been found to be an obstacle problem for practical use. It was reported that the main cause is the dielectric breakdown in the glass covered over the solar cells triggered by the thunderstroke. In this paper, the effects of the parameters such as the position of thunderstroke, the wave form, the peak value and the front duration of the lightning current, were examined by means of numerical calculation. For the lightning current, a step-like waveform and an impulse waveform were examined. The effect of the induced voltage was found to be independent of the waveform. The peak value, the front duration of the lightning current greatly affects the induced voltage.

Response to Applied Voltage to Polyurethane Soft Actuators with Loaded Weights

We are investigating a polymer soft actuator using a poly urethane elastomer film that bends by an applied voltage, which is based on the electrostriction. In this article, the bending magnitude was studied in a weight-loaded condition. It was found that the thinner film can bend the more without any load, but that the film of 200-μm thickness gave the largest bending when weights were loaded. The thickness of some degree is critical for large bending torque.

A selective ensemble mean technique for tropical cyclone(TC)track forecasts,which excludes from the ensemble those models that have large position errors at short lead times,was applied to a set of TC track forecasts produced by 11 operational global deterministic *** position errors of the resulting selective ensemble mean TC track forecasts were verifi ed for 91 TCs in the western North Pacifi c from 2010 to 2013 that reached an intensity classifi cation of'tropical storm'or *** TC position errors of the selective ensemble mean were smaller than those of a simple 11-member ensemble mean by 14.4%,7.4%and 4.7%at forecast times of24,48 and 72 hours,***,the errors were larger than those of the best single-model-based deterministic forecasts,which were ECMWF *** correlation between TC position errors at short and long lead times was weak,which partially explains why the selective ensemble mean technique in this study had lesser skill than ECMWF *** operational forecasting,simple ensemble mean forecasts by ECMWF and NCEP generally provide the best forecast performance for verifi cation samples from 2010 to 2013.

Acetylacetone-Based Electrolyte in Dye Sensitized Solar Cell

Dye sensitized solar cells attract much attention for a clean energy generation device. Among several solvents for the electrolyte, we investigated here the cell characteristics with acetylacetone as a solvent. The electric conductivity of the electrolyte increases as the concentration of polyethylene glycol (PEG) decreases or that of ionic liquid increases. The addition of pyridine into the electrolyte improves both the open voltage and the short current density. On the other hand, the replacement of PEG with fluorinated oligomer in the gel electrolyte highly increases the short current density where the open voltage is not varied. As the concentration of ionic liquid increase, the open voltage and the short current density gradually increase. When more than 20 wt.% of the ionic liquid was mixed, the gelation was not obtained. As a result, acetylacetone is a practical solvent for a gel electrolyte with the fluorinated oligomer and ionic liquid.

Electric Conductivity of Fluorinated Oligomer Gel Electrolyte Including Ionic Liquid and Performance of Dye Sensitized Solar Cell

Fluorinated oligomer gel is suitable to the electrolyte of dye sensitized solar cell. This article studied mainly in the scope of electric conductivity, including ionic liquid in the electrolyte. It was found that the ratio of mixing with dimetyl sulfoxide and the concentration of LiI affect the conductivity. The behavior is different depending on the type of ionic liquid. Although the mixing ionic liquid enhances the conductivity, the short circuit current density of assembled solar cell with it was suppressed so much.
