Background: The concomitant presence of intestinal parasite infections (IP) and tuberculosis (TB) has relevance. M. tuberculosis immune response is associated with type 1 T helper cell (Th1) while IP is associated with type Th2 cell. However, there are several contradictory reports on cytokine production under coinfection and this could be in association to the mycobacterial antigens used in the studies. Aim: To get insight into the effects of different M. tuberculosis-specific antigens (ESAT-6/cFP-10 and 38 kDa/cFP-10) in generating of appropriate cytokines on peripheral blood mononuclear cells of IPTB co infected patients. Method: ELISA assessed IFN-γ and other 16 cytokines production and plasm IgE. In 18 months, we documented demographic, economic, clinical characteristics and IP frequency in individuals from Brazil. Results: An overall 10/35 (28.5%) were IPTB co infected and 40/76 (52.6%;p = 0.024) asymptomatic intestinal parasite infected community controls (IPcc). Endo-limax nana (40%) and Entamoeba coli (22%), were the most nonpathogenic protozoan identified and Entamoeba histolytica, Giardia intestinalis, Ascaris lumbricoides and Strongyloides stercoralis were the pathogenic species (40%). IgE was higher in IPcc (p = 0.036). cytokine profiles were significantly biased toward a Th2 type IL-5 (p = 0.001) and IL-13 (p = 0.033), pro-inflammatory GM-cSF (p = 0.019) and borderline lower IL-1β in IPTB, all associated with ESAT-6/cFP-10, while IL-7 was borderline lower, but 38 kDa/cFP-10 associated;as well as IL-8 higher (p cc/IPcc. The TB/IPTB IFN-γ levels were similar to both antigens stimuli (p ≥ 0.208). conclusion: Therefore, coin-cident IPTB coinfection did not exert a significant inhibitory effect in IFN-γ production in response to either of the two antigens, but the partial discrepancy in Th1/Th2 response, is associated with the antigen priming cells.

Subcutaneous Model for the Study of dengue Virus Infection in Immune competent Mice

Various mouse models to study dengue have been described by different authors, some of them using immunodeficient or some using humanized mice. Our group reported previously a deadly murine model, which used the intracranial inoculum of highly virulent dengue virus (deNV) in immune competent mouse. Here we present a model of immune competent mouse (c57BL/6), infected subcutaneously by the same highly virulent deNV (deNV3 genotype I). In this immunocompetent systemic mice model, the cytokine levels and hematological parameters such as total and differential leukocyte and platelets counts, together with weight loss, were considered important monitoring parameters, allowing a better understanding of the systemic human disease. Mice were inoculated subcutaneously and evaluated by the percentage weight variation as well as the clinical signs. Hematological parameters and cytokines levels were measured and viral titration in brain tissue or serum neutralization was performed to confirm mice infection. The subcutaneously deNV inoculated mice showed weight loss after infection, but they did not show any other clinical signs. The leukocytes and platelets decreased after subcutaneous inoculation. The cytokines TNF alpha and IFN gamma increased after infection in mice. The subcutaneous model provided scope for improved understanding of the dengue pathogenesis, as well as possible mechanism for protection to subsequent mouse infected by intracranial route in mice. This model could be used to study the vertebrate immune response and evaluation of drugs or vaccine against dengue virus.

The treatment of infections caused by fungi and trypanosomatids is difficult due to the eukaryotic nature of these microbial cells,which are similar in several biochemical and genetic aspects to host *** this scenario,very few antifungal and anti-trypanosomatidal agents are in clinical use and,therefore,therapy is limited by drug safety considerations and their narrow spectrum of activity,efficacy and resistance. What Do Seedlings Like? The Relationship between Seedling Richness and Abundance with Abiotic Factors

Natural regeneration is the interaction of natural processes to restore the forest ecosystem. Its dynamics are influenced by the intensity and extent of a series of abiotic and biotic factors, which may be intrinsic or extrinsic. Knowing the importance of establishing natural regeneration within forest ecosystems, this work aimed to evaluate whether litter depth and trail distance influenced seedling abundance and richness in a forest fragment undergoing natural restoration. The hypothesis tested in this research was that abiotic factors influence the natural regeneration of this forest since they are factors that are directly linked to seed germination and seedling establishment. 30 plots of 1 m2 were randomly analyzed within the forest located on the brown trail. A millimeter ruler was used to diagnose the litter depth and a measuring tape to measure the distance from the plot to the edge. In each plot the seedlings were morpho-specified and each morphospecies had the number of individuals counted. Linear regression tests were performed to assess the relationships between species richness and trail distance and litter depth. The same was done for species abundance. All results showed that there is no relationship pattern between any of the variables. Other factors also influence the regeneration of a forest, such as luminosity and seed bank. Furthermore, litter depth is related to the successional stage of the forest. It was concluded that the abiotic factors tested do not influence the regeneration of the study area.

Each Epiphyte on Its Branch: A comparative Study between Different Phorophytes

Epiphytes are plants that use the mechanical support provided by a host tree, called phorophytes, without emitting haustorial structures. constituting an important component of the flora, playing an important role in the maintenance of ecosystems, such as water and nutrient cycling, in addition to providing resources, sometimes unique, such as food and shelter, for the canopy fauna. However, the epiphytic flora may show high beta diversity, even when compared with different phorophytes, possibly as a response to the dendromorphological characteristics of the phorophyte. Thus, this work aims to: 1) compare whether the species richness of epiphytes differs in trees with different morphological characteristics, and 2) compare species composition among three tree species. For this, 10 individuals of three tree species were selected: Guaria guidonia, Ficus sp., and Roystonea oleracea. In each individual, the number of species (richness) of vascular epiphytes was recorded and obtained from the species composition. To test differences in richness values between the three trees, we used analysis of variance (ANOVA) followed by Tukey’s test. To understand whether the epiphyte community formed separate groups according to the tree species used, we used the available non-metric multidimensional scaling method (NMDS). Thirty-four species of epiphytes were found in the 30 phorophytes surveyed. The three tree species showed significantly different richness, with the highest found in Guarea guidonea and the lowest in Roystonea oleraceae. The NMDS showed that epiphytes group the three tree species into distinct groups, although Ficus sp individuals present a transitional community between the two other species. Thus, the results suggest that the differences in the richness and composition of the epiphytic flora of the different phorophytes are influenced by their distinct morphological characteristics.

Rapid Differentiation of closely Related citrus Genotypes by Fluorescence Spectroscopy

The differentiation of closely related citrus genotypes is a meticulous, laborious, and time-consuming task that involves the assessment of complex traits such as growth, tolerance to stress, photosynthetic efficiency, yield, and many others. Such a task is generally accomplished either by analyzing specific features of adult plants or by applying molecular markers to young trees. On one hand, only after plants start yielding can distinct genotypes be differentiated by comparing their fruit sizes, shapes, taste, and the number of seeds. On the other hand, molecular markers are expensive, and demand expertise and time for the analysis of a larger number of plants. For these reasons, the development of techniques that could assist in an early, quick and accurate differen-tiation of closely related citrus varieties is of utmost importance. In this context, laser-induced fluorescence spectroscopy (LIFS) is a promising technique, since it is rapid, highly sensitive, and inexpensive. Previous studies showed that LIFS can differentiate a variety of sweet orange. However, this new study aimed to determine LIFS accuracy in the differentiation and grouping of very closely varieties of four Sunki mandarin selections: comum, Florida, Tropical, and Maravilha. Furthermore we compared the results with ISSR and SSR molecular markers for the same varieties. LIFS technique distinguished the four selections with accuracy greater than 70%. Only with molecular markers was possible distinguishing clearly Tropical from Maravilha, but not comum from Florida selections. In this way the results suggest that LIFS may be a sound tool for helping the identification of closely citrus varieties.

AIM: To analyze the level of apoptosis in different mucosal compartments and the differential expression of Fas/Fas-ligand and perforin in H pylori-associated gastric ulcer. METHODS: Antral specimens from patients with H pylori-related active gastric ulcer (GU), H pylori-related gastritis, and non-infected controls were analysed for densities and distribution of apoptotic cells determined by the TdT-mediated dUDP-biotin nick-end-labelling method. GU patients were submitted to eradication therapy with follow-up biopsy after 60 d. Fas, FasL, and perforin-expressing cells were assessed by immunoper- oxidase, and with anti-cD3, anti-cD20 and anti-cD68 by double immunofluorescence and confocal microscopy. Quantitative analysis was performed using a computer- assisted image analyser. RESULTS: H pylori-infected antrum showed greater surface epithelial apoptosis which decreased after eradi- cation therapy. In the lamina propria, higher rates of mononuclear cell apoptosis were observed in H pylori- gastritis. co-expression of Fas with T-cell and macro- phage markers was reduced in GU. FasL- and perforin-expressing cells were increased in H pylori-infection and correlated with epithelial apoptosis. Perforin-expressing cells were also increased in GU compared with H pylori- gastritis. cONcLUSION: Epithelial apoptosis is increased in H pylori-infection and correlates to FasL- and perforin- expression by T cells. Expression of perforin is correlated with the tissue damage, and may represent the enhance- ment of a distinct cytotoxic pathway in GU. Increased expression of FasL not paralleled by Fas on T-cells and macrophages may indicate a reduced susceptibility to the Fas/FasL-mediated apoptosis of lymphoid cells in H pylori-infection.

Bromeliaceae are important in ecological restoration, showing easy adaptation to different environments, and constantly interacting with each other through ecological relationships, whose effects may or may not benefit the species involved. The hypothesis tested whether the population density of Hohenbergia augusta is higher in areas at a more advanced successional stage (brown trail), since the species is typical of more structured forest environments, occurring especially in the lower strata of the forest. Six plots of 10 meters (6 × 10 m) were installed, parallel to the brown and yellow trails, and with a distance of 5 m from the edge. Plots were allocated alternately between the two sides of the trail, 10 meters apart from each other when on the same side. In each plot, all H. augusta (Vell.) E. Morren bromeliads were sampled, without a minimum inclusion criterion, provided they were epiphytic phorophytes. The logarithm of the value was used for data normalization and later, the data were tested in a student’s t-test. The study pointed out that the highest population density of H. augusta occurred in the brown trail because the area is in a more advanced successional stage.

Bee venom acupuncture reduces neuroinflammation modulating microglia/macrophage phenotype polarization in spinal cord injury compression model

Aim: The present study aimed to examine whether apipuncture (stimulation of acupuncture points with bee venom)at ST36 and GV3 acupoints promotes neuroprotection and reduces neuroinflammation by modulating M1 and M2 phenotype ***: Wistar rats were treated with bee venom (BV) (0.08 mg/kg) injection at acupoints ST36 and GV3 [BV (ST36 + GV3)-spinal cord injury (ScI)] or BV injection at non-acupoints [BV (NP)-ScI] or no treatment (cTL-ScI)after ScI by compression. The spinal cord mRNA expression of iNOS, Arg-1 and TGF-β was measured by real time PcR and the levels of IBA-1;BcL-2;NeuN e cNPase was measured by western blotting. Locomotor performance was measured by Basso, Beattie, and Bresnahan (BBB) and grid-walking ***: Apipuncture treatment was able to (1) ameliorate locomotor performance;(2) reduce inflammatory markers (cox-2 levels) and activation of microglia and macrophages;(3) reduce the polarization of the M1 phenotype marker (iNOS) and increase M2 (Arg-1 and TGF-β) phenotypic markers;(4) promote neuroprotection by reducing the death of neurons and oligodendrocytes;and (5) increase the expression of the anti-apoptotic factor ***: Apipuncture treatment induces locomotor recovery and neuroprotection after the compression model of spinal cord injury. Further, it reduces neuroinflammation by decreasing M1 polarization and increasing M2 phenotype.
