Cadmium isotope fractionation reveals genetic variation in Cd uptake and translocation by Theobroma cacao and role of natural resistanceassociated macrophage protein 5 and heavy metal ATPase-family transporters

In response to new european Union regulations,studies are underway to mitigate accumulation of toxic cadmium(Cd)in cacao(Theobroma cacao,Tc).This study advances such research with Cd isotope analyses of 19 genetically diverse cacao clones and yeast transformed to express cacao natural resistance-associated macrophage protein(NRAMP5)and heavy metal ATPases(HMAs).The plants were enriched in light Cd isotopes relative to the hydroponic solution withΔ^(114/110)Cd_(tot-sol)=−0.22±0.08‰.Leaves show a systematic enrichment of isotopically heavy Cd relative to total plants,in accord with closed-system isotope fractionation ofΔ^(114/110)C_(dseq-mob)=−0.13‰,by sequestering isotopically light Cd in roots/stems and mobilisation of remaining Cd to *** findings demonstrate that(i)transfer of Cd between roots and leaves is primarily unidirectional;(ii)different clones utilise similar pathways for Cd sequestration,which differ from those of other studied plants;(iii)clones differ in their efficiency of Cd *** yeast that expresses TcNRAMP5(*** natural resistance-associated macrophage gene)had isotopically lighter Cd than did *** suggests that NRAMP5 transporters constitute an important pathway for uptake of Cd by *** isotope signatures of transgenic yeast expressing HMA-family proteins suggest that they may contribute to Cd *** data are the first to record isotope fractionation induced by transporter proteins in vivo.

Statistical variability and fragility assessment of ballistic perforation of steel plates for 7.62 mm AP ammunition

The paper describes field test results of 7.62×51 mm M61 AP(armour piercing) ammunition fired into mild steel targets at an outdoor *** targets varied from 10 mm to 32 mm in *** tests recorded penetration depth,probability of perforation(i.e.,complete penetration),muzzle and impact velocities,bullet mass,and plate yield strength and *** measured penetration depth exhibited a variability of approximately±12%.The paper then compared ballistic test results with predictive models of steel penetration depth and thickness to prevent *** parameters were derived for muzzle and impact velocity,bullet mass,plate thickness,plate hardness,and model error.A Monte-Carlo probabilistic analysis was then developed to estimate the probability of plate perforation of 7.62 mm M61 AP ammunition for a range of impact velocities,and for mild steels,and High Hardness Armour(HHA) *** perforation fragility analysis considered the random variability of impact velocity,bullet mass,plate thickness,plate hardness,and model *** a probabilistic analysis allows for reliability-based design,where,for example,the plate thickness with 95% reliability(*** 1 in 20 shots will penetrate the wall) can be estimated knowing the probabilistic distribution of ***,it was found that the plate thickness to ensure a low 5% probability of perforation needs to be 11-15% thicker than required to have a 50/50 chance of perforation for mild steel *** would need to be 20-30% thicker if probability of perforation is reduced to zero.

Reactivity of Pyrogenic Carbonaceous Matter (PCM) in mediating environmental reactions: Current knowledge and future trends

Pyrogenic Carbonaceous Matter(PCM;e.g.,black carbon,biochar,and activated carbon)are solid residues from incomplete combustion of fussil fuel or *** are traditionally viewed as inert adsorbents for sequestering contaminants from the aqueous phase or providing surfaces for microbes to *** this account,we reviewed the recently discovered reactivity of PCM in promoting both chemical and microbial synergies that are important in pollutant transformation,biogeochemical processes of redox-active elements,and climate change mitigation with respect to the interaction between biochar and nitrous oxide(N2O).Moreover,we focused on our group's work in the PCM-enhanced abiotic transformation of nitrogenous and halogenated pollutants and conducted in-depth analysis of the reaction *** understand what properties of PCM confer its reactivity,our group pioneered the use of PCM-like polymers,namely conjugated microporous polymers(CMPs),to mimic the performance of *** approach allows for the controlled incorporation of specific surface properties(e.g.,quinones)into the polymer network during the polymer *** a result,the relationship between specific characteristics of PCM and its reactivity in facilitating the decay of a model pollutant was systematically studied in our group's *** findings summarized in this account help us to better understand an overlooked environmental process where PCM synergistically interacts with various environmental reagents such as hydrogen sulfide and ***,the knowledge gained in these studies could inform the design of a new generation of reactive carbonaceous materials with tailored properties that are highly efficient in contaminant removal.

The use of near infrared transmittance kernel sorting technology to salvage high quality grain from grain downgraded due to Fusarium damage

The mycotoxins associated with specific Fusarium fungal infections of grains are a threat to global food and feed security. These fungal infestations are referred to as Fusarium Head Blight(FHB) and lead to Fusarium Damaged Kernels(FDK). Incidence of FDK >0.25% will lower the grade, with a tolerance of 5%FDK for export feed grain. During infestation, the fungi can produce a variety of mycotoxins, the most common being deoxynivalenol(DON). Fusarium Damaged Kernels have been associated with reduced crude protein(CP), lowering nutritional, functional and grade value. New technology has been developed using Near Infrared Transmittance(NIT) spectra that estimate CP of individual kernels of wheat, barley and durum. Our objective is to evaluate the technology's capability to reduce FDK and DON of downgraded wheat and ability to salvage high quality safe kernels. In five FDK downgraded sources of wheat,the lowest 20% CP kernels had significantly increased FDK and DON with the high CP fractions having decreased FDK and DON, thousand kernel weights(TKW) and bushel weight(Bu). Strong positive correlations were observed between FDK and DON(r = 0.90); FDK and grade(r = 0.62) and DON and grade(r = 0.62). Negative correlations were observed between FDK and DON with CP(r =-0.27 and-0.32);TKW(r =-0.45 and-0.54) and Bu(r =-0.79 and-0.74). Results show improved quality and value of Fusarium downgraded grain using this technology.

In systems biology, the dynamics of biological networks are often modeled with ordinary differential equations (ODes) that encode interacting components in the systems, resulting in highly complex models. In contrast, the amount of experimentally available data is almost always limited, and insufficient to constrain the parameters. In this situation, parameter estimation is a very challenging problem. To address this challenge, two intuitive approaches are to perform experimental design to generate more data, and to perform model reduction to simplify the model. experimental design and model reduction have been traditionally viewed as two distinct areas, and an extensive literature and excellent reviews exist on each of the two areas. Intriguingly, however, the intrinsic connections between the two areas have not been recognized. experimental design and model reduction are deeply related, and can be considered as one unified framework. There are two recent methods that can tackle both areas, one based on model manifold and the other based on profile likelihood. We use a simple sum-of-two-exponentials example to discuss the concepts and algorithmic details of both methods, and provide Matlab-based code and implementation which are useful resources for the dissemination and adoption of experimental design and model reduction in the biology community. From a geometric perspective, we consider the experimental data as a point in a high-dimensional data space and the mathematical model as a manifold living in this space. Parameter estimation can be viewed as a projection of the data point onto the manifold. By examining the singularity around the projected point on the manifold, we can perform both experimental design and model reduction. experimental design identifies new experiments that expand the manifold and remove the singularity, whereas model reduction identifies the nearest boundary, which is the nearest singularity that suggests an appropriate form of a reduced

Heart healthy equals prostate healthy and statins, aspirin, and/or metformin （S.A.M.） are the ideal recommendations for prostate cancer prevention

Cardiovascular disease （CVD） has been the number one cause of death in the U.S. for 114 of the last 115 years. Lifestyle factors that promote CVD also appear to increase prostate cancer risk and those that reduce CVD risk also appear to reduce the risk of prostate cancer. The largest randomized trials utilizing dietary supplements or pharmacologic agents for prostate cancer prevention （Selenium and Vitamin e Cancer Prevention Trial [SeLeCT]） have also shed light on the problems and future solutions in this area. Dietary supplements that have not been found to be CVD protective, such as selenium and Vitamin e have not been found to be prostate protective. In addition, over exposure to specific anti-oxidants in nutritionally replete populations may be encouraging cancer growth. Futuretrials of dietary supplements to prevent prostate cancer could be problematic because by the time a definitive trial is initiated the participants will no longer be ＂deficient＂ in the nutrient being tested, which arguably occurred in the SeLeCT trial. It is also interesting that statins, aspirin, and/or metformin （S.A.M.） are 3 generic, low-cost, heart healthy agents derived from natural sources with separate mechanism of actions, which all appear to have the best benefit to risk ratio compared to any other agent available for prostate cancer prevention, especially aggressive disease, or as an ancillary agent （s） to conventional cancer treatment. It is time to focus on the forest over the trees and recommend proven CVD protective measures for men concerned about their risk of prostate cancer.

Dietary supplementation of sulfur amino acids improves intestinal immunity to eimeria in broilers treated with anti-interleukin-10 antibody

Oral antibody to interleukin-10(anti-IL-10)enhances the intestinal immune defense against *** sulfur amino acids methionine and cysteine(MtC)play essential roles in inducing and maintaining protective immune responses during intestinal ***,increased dietary MtC may support the anti-IL-10-induced intestinal immunity to ***(n=640)were arranged in a 2×2×2 factorial design with 2 levels of each of the 3 main factors:dietary standardized ileal digestible(SID)MtC levels(0.6%or 0.8%),dietary anti-IL-10 supplementation(with or without),and coccidiosis challenge(control or challenge).Briefly,the broilers were supplied with either 0.6%or 0.8%SID MtC,each with or without anti-IL-10(300 mg/kg),from d 10 to *** d 14,broilers from each diet were gavaged with either PBS or *** resulting eimeria infection induced fecal oocyst shedding and intestinal *** fed 0.8%SID MtC(main effects,P≤0.05)had decreased feed-to-gain ratio,increased duodenum and cecum luminal anti-eimeria IgA titers,and decreased fecal oocyst counts,when compared to 0.6%SID *** supplementation of anti-IL-10(main effects,P≤0.05)increased cecum luminal total IgA concentration and decreased cecum ***(P≤0.05)were detected for growth performance and cecum luminal IFN-γ.Briefly,the highest body weight gain and feed intake were reached in PBS-gavaged broilers fed 0.8%SID MtC with no anti-IL-10 and in eimeria-challenged broilers fed 0.8%SID MtC with *** eimeria-infected broilers,anti-IL-10 increased intestinal luminal IFN-γ and body weight gain only at 0.8%SID ***,anti-IL-10 increased intestinal luminal IFN-γ levels,decreased cecum lesions and restored growth only when fed with adequate amounts of sulfur amino *** findings underscore the importance of providing sufficient essential nutrients to support the anti-IL-10 induced immunity against coccidiosis.

Folate binding protein—Outlook for drug delivery applications

Serum proteins represent an important class of drug and imaging agent delivery vectors. In this mini- review, key advantages of using serum proteins are discussed, followed by the particular advantages and challenges associated with employing soluble folate binding protein. In particular, approaches employing drugs that target folate metabolism are reviewed. Additionally, the slow-onset, tight- binding interaction of folate with folate binding protein and the relationship to a natural oligomerization mechanism is discussed. These unique aspects of folate binding protein suggest interesting applications for the protein as a vector for further drug and imaging agent development.
