The formation of axonal spheroid is a common feature following spinal cord *** further understand the source of Ca^(2+)that mediates axonal spheroid formation,we used our previously characterized ex vivo mouse spinal cord model that allows precise perturbation of extracellular Ca^(2+).We performed twophoton excitation imaging of spinal cords isolated from Thy1YFP+transgenic mice and applied the lipophilic dye,Nile red,to record dynamic changes in dorsal column axons and their myelin sheaths *** selectively released Ca^(2+)from internal stores using the Ca^(2+)ionophore ionomycin in the presence or absence of external Ca^(2+).We reported that ionomycin dose-dependently induces pathological changes in myelin and pronounced axonal spheroid formation in the presence of normal 2 m M Ca^(2+)artificial cerebrospinal *** contrast,removal of external Ca^(2+)significantly decreased ionomycin-induced myelin and axonal spheroid formation at 2 hours but not at 1 hour after *** mice that express a neuron-specific Ca^(2+)indicator in spinal cord axons,we confirmed that ionomycin induced significant increases in intra-axonal Ca^(2+),but not in the absence of external Ca^(2+).Periaxonal swelling and the resultant disruption in the axo-myelinic interface often precedes and is negatively correlated with axonal spheroid *** with YM58483(500 n M),a well-established blocker of store-operated Ca^(2+)entry,significantly decreased myelin injury and axonal spheroid ***,these data reveal that ionomycin-induced depletion of internal Ca^(2+)stores and subsequent external Ca^(2+)entry through store-operated Ca^(2+)entry contributes to pathological changes in myelin and axonal spheroid formation,providing new targets to protect central myelinated fibers.

随着化石燃料燃烧导致的二氧化碳排放不断加剧气候变化,且化石燃料储量日益减少,寻求可再生能源已成为一项紧迫的任务.其中,藻类衍生可持续燃料因具有成本优势和可运输性,在解决全球能源危机方面展现出广阔前景,备受关注.利用化学转化技术从微藻中提取脂质,并通过酯交换反应可以将其转化为脂肪酸甲酯,是生产绿色生物燃料的有效途径.这一过程涉及游离脂肪酸、磷脂和甘油三酯的提取,并且生产过程能耗低,成为满足日益增长的能源需求的一种理想解决方案.本文综述了微藻脂质提取和酯交换制备生物燃料的相关研究进展.首先,介绍了微藻脂质的提取方法,包括溶剂提取法、索氏提取法、布利格和戴耶法、超临界二氧化碳提取以及离子液体溶剂法等,并分析了各方法的优缺点.随后,重点阐述了酯交换技术在微藻脂质转化中的应用,包括酸碱催化、酶催化以及原位酯交换反应等,并探讨了这些技术的反应机理、催化剂选择、反应器设计以及生物油生产工艺等方面的研究进展.通过综述上述研究进展,为微藻脂质的生产和应用提供了理论指导.研究表明,通过优化催化剂种类、反应条件以及提取方法,可以有效提高微藻脂质的转化效率和生物油品质.同时,本文也指出了当前微藻脂质生产中面临的挑战,如微藻栽培和生长条件优化、高效转化技术的开发等.随着可持续能源日益受到重视,微藻脂质作为一种可再生能源具有巨大的发展潜力.未来研究应进一步关注微藻的规模化栽培、生长条件优化以及高效转化技术的研发,以提高微藻脂质的产量和品质.同时,应进一步推动和实现微藻生物燃料的实际应用,从而为应对气候变化和能源危机提供有效的解决方案.

Transcriptional regulation of human polo-like kinases and early mitotic inhibitors

Human polo-like kinases （PLK1-PLK4） have been implicated in mitotic regulation and carcinogenesis. PLK1 phosphorylates early mitotic inhibitor 1 （emil） to ensure mitosis entry, whereas emi2 plays a key role during the meiotic cell cycle. Transcription factor e2F is primarily considered to regulate the G1/S transition of the cell cycle but its involvement in the regulation of mitosis has also been recently suggested. A gap still exists between the molecular basis of e2F and mitotic regulation. The present study was designed to characterize the transcriptional regulation of human PLK and emi genes. Adenoviral overexpression of e2F1 increased PLK1 and PLK3 mRNA levels in A549 cells. A reporter gene assay revealed that the putative promoter regions of PLK1, PLK3, and PLK4 genes were responsive to activators e2F, e2F1-e2F3. We further characterized the putative promoter regions of emil and emi2 genes, and these could be regulated by activators e2F and e2F1-e2F4, respectively. Finally, PLK1-PLK4, emil, and emi2 mRNA expression levels in human adult, fetal tissues, and several cell lines indicated that each gene has a unique expression pattern but is uniquely expressed in common tissues and cells such as the testes and thymus. Collectively, these results indicate that e2F can integrate G1/S and G2/M to oscillate the cell cycle by regulating mitotic genes PLK and emi, leading to determination of the cell fate.

Antimicrobial resistance(AMR)poses a critical threat to global health and development,with environmental factors—particularly in urban areas—contributing significantly to the spread of antibiotic resistance genes(ARGs).However,most research to date has been conducted at a local level,leaving significant gaps in our understanding of the global status of antibiotic resistance in urban *** address this issue,we thoroughly analyzed a total of 86,213 ARGs detected within 4,728 metagenome samples,which were collected by the Meta SUB International Consortium involving diverse urban environments in 60 cities of 27 countries,utilizing a deep-learning based *** findings demonstrated the strong geographical specificity of urban environmental resistome,and their correlation with various local socioeconomic and medical *** also identified distinctive evolutionary patterns of ARG-related biosynthetic gene clusters(BGCs)across different countries,and discovered that the urban environment represents a rich source of novel *** study provides a comprehensive overview of the global urban environmental resistome,and fills a significant gap in our knowledge of large-scale urban antibiotic resistome analysis.

GINS, a heterotetramer of SLD5, PSF1, PSF2, and PSF3 proteins, is an emerging chromatin factor recognized to be involved in the initiation and elongation step of DNA replication. Although the yeast and Xenopus GINS genes are well documented, their orthologous genes in higher eukaryotes are not fully characterized. In this study, we report the genomic structure and transcriptional regulation of mammalian GINS genes. Serum stimulation increased the GINS mRNA levels in human cells. Reporter gene assay using putative GINS promoter sequences revealed that the expression of mammalian GINS is regulated by 17β-estradiolstimulated estrogen receptor a, and human PSF3 acts as a gene responsive to transcription factor e2F1. The goal of this study is to present the current data so as to encourage further work in the field of GINS gene regulation and functions in mammalian cells.

为探究土壤重金属无机钝化剂及其有机改良化钝化剂对稻田镉钝化和温室气体排放的综合影响,本研究通过水稻盆栽实验,实施了空白对照组、施加1%无机钝化剂(石灰∶海泡石=1∶1)组、施加1%无机钝化剂和0.5%生物炭组以及施加1%无机钝化剂和0.5%发酵菌糠组4个处理,监测不同处理对镉钝化、温室气体排放、作物产量和土壤质量的影响.结果表明:无机钝化剂显著降低了糙米镉含量43.4%(p<0.05),对稻田温室气体排放无显著影响(p<0.05).生物炭改良和发酵菌糠改良的钝化剂分别降低糙米镉含量的60.3%和46.7%(p<0.05),但同时显著增加了温室气体甲烷的累积排放通量(分别增加30.4%和113.7%,p<0.05).相比于发酵菌糠改良钝化剂,生物炭改良能更显著地提升镉钝化效率并且较低地增加稻田甲烷排放风险.在作物增产和土壤提质方面,生物炭改良钝化剂还具有显著提升水稻籽粒产量、土壤有机碳和速效养分含量的优点.本研究表明,稻田施用无机钝化剂是一种保守且无碳排影响的修复方式,施用生物炭改良钝化剂是一种高效、多效但具低甲烷排放风险的修复方式.

Reactive oxygen species (ROS) have been known to play an important role in the pathogenesis of atherosclerosis and several other cardiovascular diseases. It is now apparent that ROS induce endothelial cell damage and vascular smooth muscle cell (VSMC) growth and cardiac remodeling, which are associated with hypertension, atherosclerosis, heart failure, and restenosis. Several lines of evidence have indicated that ROS and mitogen activated protein (MAP) kinases were involved in vascular remodeling under various pathological conditions. Recently, it was also reported that MAP kinases were sensitive to oxidative stress. MAP kinases play an important role in cell differentiation, growth, apoptosis, and the regulation of a variety of transcription factors and gene expressions. Bioflavonoids and polyphenolic compounds are believed to be beneficial for the prevention and treatment of atherosclerosis and cardiovascular diseases.
