Salinity Tolerance in Argentinean Population of <i>Bromus catharticus</i>. Variability and Direct and Indirect Effects on Seedling Characters

The aim of this research was to evaluate the salinity tolerance in prairie grass populations at the seedling stage quantifying the variability and the influence of physiological traits related to it. Salinity tolerance, in Bromus catharticus Vahl (prairie grass) populations collected in different environments of the Pampean Phytogeography region (Argentine) was evaluated at the seedling stage, using controlled condition of temperature and light. It was adopted a completely randomized design using 3 plots with three plants each one per population and two levels of treatment: 0 mM and 100 mM NaCl. Morphological, biomass and membrane stability root and shoot traits were studied. A factorial ANOVA with interaction was estimated. Then one way ANOVA for all seedling traits in both treatments allowed estimating variance components, coefficient of genotypic determination (CGD) and variation index (VI). Comparisons between populations were made using Tukey test (at 5% of probability). Phenotypic correlations among traits were calculated and then a path coefficient analysis separated direct and indirect effects at 100 and 0 mM NaCl. No significant interactions “Population × Treatment” were found for any character. The saline stress caused a pairing in the population means for the most traits. Coefficients of variation were mainly higher when the seedlings grew without stress (0 mM) because it allowed a greater potential genotypic expression. The absence of significant interactions denotes a good homeostatic capacity of the prairie grass facing that abiotic stress. Leaf length, shoot length and root dry matter were the variables with the largest direct and indirect effects. Renin-Angiotensin System (RAS) is involved with hypertension and other cardiovascular diseases. However, the association of RAS components to cancer still causes suspicion. To try to clarify this, here we aimed to show this association for three important components: Angiotensin Converting Enzyme 1 (ACE1), Angiotensin Type 1 Receptor (AGTR1) and Angiotensin Type 2 Receptor (AGTR2). The first articles show that association of RAS components with cancer dates back to the 70’s. ECA1 and AGTR1 have close association with cancer and ACE1 inhibitors or AGTR1 blockers are candidates to treatment of some tumors. Moreover, the action of AGTR2 is still controversial, but most studies show that the increased expression of AGTR2 can attack the cancer cells. In breast cancer, these components have also been widely studied and many works have shown that the correlation exists. Therefore specific target using these RAS components could be a beneficial, novel therapy to various tumors.

Background: Breast cancer is the most common type of cancer among women. Diagnosed and treated timely, patients may have good prognostics. In Brazil, in 2012, the estimate of new cases was 52,680 and the number of registered deaths in 2012 was 12,852. The Renin-Angiotensin System (RAS) is known for its role in arterial hypertension and in other cardiovascular diseases. Angiotensin-Converting Enzyme 2 (ACE2) is the key to Ang-(1-7) formation, and counterbalances the ACE1/AngII/AGTR1 axis actions. RAS components have complex interactions with different tissues and their actions are not restricted to the cardiovascular system. Recently, the RAS has been associated with different types of cancers and in particular with gynecological cancers. Objectives: Our aim is to investigate possible associations between allelic distribution of two genetic polymorphisms in the AGTR2 receptor with ACEs 1 and 2 plasma levels among women with breast cancer. Patients and Methods: Patients with breast cancer were genotyped for two polymorphisms of the AGTR2 (T1247G and A5235G). Genotyping assays (TaqMan) were performed with genomic DNA extracted from blood cells. ACEs plasma level measurements were conducted in women from the breast-cancer group (N = 53). ACEs were measured in the plasma of these patients using ELISA kits. Results: SNPs genotype distribution is correlated with ACEs plasma levels. ACEs plasma levels are also correlated with clinical variables and ACE2 high levels are associated with better prognostics. Conclusions: Changes in circulating levels of ECA1/AngII ECA2/ Ang-(1-7) determine the magnitude of the inflammatory response that an individual can trigger and the variation in ACE 1 and 2 plasma level measurements in the blood of breast cancer patients suggests an association with the process of mammary carcinogenesis. Thus, the RAS may be associated with the process of mammary carcinogenesis by both genotypic variations of RAS components and by circulating levels of ACEs

Breast cancer is the most common cancer among women. Angiotensin-(1 - 7) [Ang-(1 - 7)] has been correlated with cancer antiproliferative and apoptotic effects, similar properties of the human Chorionic Gonadotrofin (hCG). The aims of this work are to evaluate the role of Ang-(1 - 7) and of hCG in modulating the expression of Nuclear Receptors and Coregulators related genes in the tumorigenic breast cell line SK-BR3. Three experimental groups were created: control, hCG and hCG + Ang-(1 - 7). Cells were treated for 11 days and then had their RNA extracted. Samples were loaded into PCR Array plates containing 84 genes relate to Nuclear Receptors and Coregulators pathways. Gene expression data were used to construct canonical pathways (MetacoreTM). hCG and hCG + Ang-(1 - 7) treatments markedly modulate the expression of Nuclear Receptors and Coregulators related genes. hCG differentially expressed 17% of the genes, being 29% upregulated and 71% downregulated. Meanwhile, hCG + Ang-(1 - 7) changed the expression of 30% of the genes on the plate, among these genes 56% were upregulated and 44% downregulated. Among these differentially expressed genes, we highlight Esr1, Nr2f2, and Nr2f1, Esr1, Hdac5, and Nr4A1 (>4 fold). Finally MetaCore analysis based on Gene Ontology (GO) generated six networks for hCG and ten networks for the combined treatment. All generated networks are related to regulation of apoptosis or to Programmed Cell Death processes. In summary, our results herein demonstrate that the modulation of sexual hormones and of other nuclear factor genes expression might underlie the tumorigenic protection effect and the induction of cell differentiation caused by the hormones hCG and Ang-(1 - 7), especially in Cancer Stem Cells.

Selective biomass conversion over novel designed tandem catalyst

Selective conversion of biomass into targeted molecules like polyols, especially, from cellulosic compounds, is being widely investigated as a sustainable process to produce biodiesel and bio- additives. The known process involves two steps, namely hydrolysis and hydrogenation. Thus, it requires two different catalytic materials or bifunctional catalysts. In this context, the present work reports a new catalytic approach based on the use of tandem catalysts, consisting of the combination of an acid solid catalyst (active for hydrolysis) and a supported metal catalyst (active for hydrogenation). Two different functionalized activated carbons and the resin Amberlyst 15 have been tested as solid acid catalysts, and Ru nanoparticles supported on the original activated carbon (SA) are the metal catalyst part of the tandem. All the tested tandem catalysts exhibited higher activity than the supported Ru catalyst did. The highest cellulose conversion and selectivity to sorbitol (70% and 86%, respectively) have been obtained over a novel tandem catalyst, which resulted from a physical mixture between a sulfuric acid modified SA carbon (SASu) and Ru loaded SA (Ru/SA), leading to a tandem catalyst (Ru/SA + SASu). This novel-designed tandem catalyst is reusable. Based on tandem catalysts with a solid-solid system combination, the adopted novel- designed catalytic approach is cost-efficient and sustainable, and can be considered promising for the green production of high-added-value chemicals.

In recent years,the incidence of inflammatory bowel disease(IBD) has been on the rise,extending to countries where it was infrequent in the past. As a result,the gap between high and low incidence countries is decreasing. The disease,therefore,has an important economic impact on the healthcare system. Advances in recent years in pharmacogenetics and clinical pharmacology have allowed for the development of treatment strategies adjusted to the patient profile. Concurrently,new drugs aimed at inflammatory targets have been developed that may expand future treatment options. This review examines advances in the optimization of existing drug treatments and the development of novel treatment options for IBD.

Background: Pancreaticoduodenectomy is the standard treatment for resectable periampullary cancer. Surgical site infections(SSI) are common complications with increased morbidity. The study aimed to describe the prevalence, risk factors, microbiology, and outcomes of SSI among patients undergoing pancreaticoduodenectomy. Methods: We conducted a retrospective study in a referral cancer center between January 2015 and June 2021. We analyzed baseline patient characteristics and SSI occurrence. Culture results and susceptibility patterns were described. Multivariate logistic regression was used to determine risk factors, proportional hazards model to evaluate mortality, and Kaplan-Meier analysis to assess long-term survival. Results: A total of 219 patients were enrolled in the study;101(46%) developed SSI. Independent factors for SSI were diabetes mellitus, preoperative albumin level, biliary drainage, biliary prostheses, and clinically relevant postoperative pancreatic fistula. The main pathogens were Enterobacteria and Enterococci. Multidrug-resistance rate in SSI was high but not associated with increased mortality. Infected patients had higher odds of sepsis, longer hospital stay and intensive care unit stay, and readmission rate. Neither 30-day mortality nor long-term survival was significantly different between infected and non-infected patients. Conclusions: SSI prevalence among patients undergoing pancreaticoduodenectomy was high and largely caused by resistant microorganisms. Most risk factors were related to preoperative instrumentation of the biliary tree. SSI was associated with greater risk of unfavorable outcomes;however, survival was unaffected.
