Effect of Environmental Factors on Rock-inhabiting Fungal Communities from Brazilian Soapstone Samples

The present study aims to evaluate the environmental effect on fungal community composition associated with biodeterioration occurring in stones （soapstone） at two distinct locations in Minas Gerais State, Brazil： Congonhas city and Sanctuary of Cara^a. Four collections of fungal communities over one year were obtained from both research sites from the soapstone block surfaces exposed for over two decades. The molecular diversity profile of the fungal community at the two localities was obtained by DGGE （Denaturing Gradient Gel Electrophoresis）, and the genomes of the most representative population were sequenced. DGGE showed the formation of two clusters with filamentous fungal communities. Sequencing of the most representative bands revealed the presence of fungi associated with the biodeterioration of soapstone. In addition, many of the identified species were associated with photobionts that could generate lichens, indicating that environmental characteristics affect the occurrence of filamentous fungi, which leads to biodeterioration of stones. Authors＇ study focused on an environmental variation of an extreme habitat for fungi associated with soapstone in the state of Minas Gerais, Brazil and identified the presence of interesting rock-inhabiting fungal communities including species related to lichens, which can accelerate the deterioration of stones by the production of organic acids.

Evaluation of Visible Losses and Damage to the Ratoon Cane in the Mechanized Harvesting of Sugarcane for Different Displacement Speeds

This study aimed at quantitatively evaluating the sugarcane losses and the damage caused to the ratoon cane while using the Case IH A8800 harvester in different displacement speeds;it was conducted in the municipality of Campos dos Goytacazes, in Rio de Janeiro, Brazil. Three speeds were used as treatment (2 km·h-1, 3 km·h-1, and 4.5 km·h-1), and each treatment was composed of six rows of harvested ratoon, each with a length of 290 m. In order to evaluate the quantitative losses in t·ha-1 and the percentage of losses, the remaining sugarcane, left on the field after harvesting, was collected. The sampling frame was set to every 50 m, dividing the borders by 40 m;the measurement for each sampling area was of 20 m2, with five repetitions. In order to evaluate the damage caused to the ratoon canes, we chose a visual methodology to classify the damage degrees, ranging from one to four. There was no significant difference in losses when comparing different speeds. Therefore, it is more advantageous and economically viable to use the speed of 4.5 km·h-1, which collects more in less time.

Production of Aflatoxins from Aspergillus flavus in Liquid Medium

Aflatoxins are produced mainly by Aspergillus flavus and Aspergillus parasiticus, and can be found in many grains such as peanuts, soybeans and com. This study aimed to qualitatively and quantitatively evaluate the production of aflatoxin in liquid media using strains of Aspergillus flavus obtained from peanuts marketed in the city of Fortaleza, CEo Strains of Aspergillus flavus were inoculated into a liquid medium malt extract and after 2 days inoculated into a second medium containing sucrose 5%, MgSo4·7H20 0.1%, KH2Po4 1%, ZnSo4·7H2o 0.0176 g, and cultured for 3 more days. The media were kept at room temperature ranging from 24°C to 32 °C with agitation of 130 rpm and aeration of 4.17 Llmin. Qualitative analysis was performed by thin layer chromatography and quantitatively by high performance liquid chromatography with fluorescence detection, demonstrating the production of aflatoxin B I （588 mg/L） and B2 （929 mg/L）.

AIM To develop metabonomic models(MMs), using 1 H nuclear magnetic resonance(NMR) spectra of serum, to predict significant liver fibrosis(SF: Metavir ≥ F2), advanced liver fibrosis(AF: METAVIR ≥ F3) and cirrhosis(C: METAVIR = F4 or clinical cirrhosis) in chronic hepatitis C(CHC) patients. Additionally, to compare the accuracy of the MMs with the aspartate aminotransferase to platelet ratio index(APRI) and fibrosis index based on four factors(FIB-4). METHoDS Sixty-nine patients who had undergone biopsy in the previous 12 mo or had clinical cirrhosis were included. The presence of any other liver disease was a criterion for exclusion. The MMs, constructed using partial least squares discriminant analysis and linear discriminant analysis formalisms, were tested by cross-validation, considering SF, AF and C. RESULTS Results showed that forty-two patients(61%) presented SF, 28(40%) AF and 18(26%) C. The MMs showed sensitivity and specificity of 97.6% and 92.6% to predict SF; 96.4% and 95.1% to predict AF; and 100% and 98.0% to predict C. Besides that, the MMs correctly classified all 27(39.7%) and 25(38.8%) patients with intermediate values of APRI and FIB-4, respectively. CoNCLUSIoN The metabonomic strategy performed excellently in predicting significant and advanced liver fibrosis in CHC patients, including those in the gray zone of APRI and FIB-4, which may contribute to reducing the need for these patients to undergo liver biopsy.

Enteroparasitoses in Patients with Rheumatoid Arthritis

Background: Patients with rheumatic diseases have profound alterations in the immune system as a result of underlying diseases and the treatments used, which increases the risk of occurrence and severity of infections, including enteroparasitosis. The current treatment for rheumatoid arthritis involves immunosuppressive therapies powerfully needed for screening infectious processes. The prevalence of parasitic infections in rheumatoid arthritis patients is not currently documented in literature. The objective of the current study was to determine the prevalence of parasitic diseases in a population with RA. Methods: We collected demographic and socioeconomic data from 67 patients at the Hospital Universitário de Brasília from July 2015 to April 2016. All patients underwent a parasitological examination of their stool and multiple variables were analyzed using Poisson regression method. Results: The mean age of patients was 53.9 years. They were predominantly in women (94%) and caucasian (47.8%). The mean disease duration was 9.2 years and most patients had the disease in remission or light activity. The prevalence of parasitic infections in these patients was 11.9%, all cases being that of protozoa of the following species: Endolimax nana, Entamoeba histolytica and Entamoeba coli. The final multivariate analysis indicated that the presence of disease had a significant statistical relationship between the presence of enteroparasitosis and rheumatoid arthritis with no fatigue by VAS (p = 0.0488) and best current health index by VAS (p = 0.0012). Conclusion: This study indicates that the prevalence of enteroparasitoses in a population with Rheumatoid Arthritis was 11.9% and all cases were found of protozoa.

Visible Losses to the Mechanical Harvesting of Ratoon Sugarcane Using the Harvester John Deere 3520

The aim of this study was to evaluate the visible loss of sugar cane and the damage to the knuckles, using the John Deere 3520 harvester on three different travel speeds (3.0;4.0 and 5.0 km·h-1), in DIC with five repetitions in Campos dos Goytacazes, Rio de Janeiro/Brazil. Each treatment was composed by six lines of harvested cane stumps, with a length of 290 m each. In these six lines, the remains of sugar cane left in the field were collected by placing the sampling frame in two central lines every 50 m, and separating 40 m of edge. The sampling area was surrounded by 2 m wide and 10 m long, totaling 20 m2. To calculate the damage to stumps, a visual methodology by [1], was used, which classifies the damage grade, ranging from 1 to 4. Fifty stumps were assessed randomly for each speed, in a sampling area of 1800 m2. The data were submitted to ANoVA and Tukey test at 5%, in order to compare the effect of different speeds on the losses and damages. There was no significant difference regarding the loss or damage by comparing the different speeds. Therefore, it is more advantageous to use the speed of 5.0 km·h-1, harvesting more in less time and causing the same level of damage.

Synthesis and Characterization of LaNixCo1-xo3 Perovskites via Complex Precursor Methods

This work presents a study on the synthesis of LaNixCo1-xo3 perovskites via complex precursor methods. Perovskite oxides with the composition LaNixCo1-xo3 were synthesized by chelating precursor and polymeric precursor methods using nickel and/or cobalt nitrates, lanthanum nitrate, ethylene glycol, citric acid, and EDTA as starting source. The obtained perovskite were characterized by thermogravimetric analysis, infrared spectroscopy, X-ray diffraction and the morphology of the samples were investigated by N2 adsorption experiments and average medium particle size. TG curves and FTIR spectra were particularly useful in establishing of the optimal calcination temperature of the precursor powders. X-ray diffraction patterns revealed the formation of the perovskite structure in all samples prepared by both synthesis method and after calcinations at 700℃. The results showed that the preparation method resulted in oxides with the intended structure. The specific surface area values were influenced by preparation method.

Comparison of Two Protocols of Agar Gel Immunodiffusion(AGID)Used to Diagnose of Equine Infectious Anemia(EIA)

The aim of the present study was to compare two protocols of Agar Gel Immunodiffusion (AGID) that can be used to diagnose Equine Infectious Anemia (EIA). Different volumes of reagents and support for agar gel were used, as recommended by the Ministry of Agriculture, Livestock and Food Supply (MAPA) and the World organization for Animal Health (oIE). Equidae sera samples (n = 1000) were tested using a national commercial kit and different volumes (i.e. 25 μL—MAPA and 50 μL—oIE) of reagents and standard sera. Antigen was used as recommended by MAPA. The results were assessed by two operators, 48 hours after beginning the test. At the end of the experiment, a Kappa index of 1 was obtained by the operators, for the interpretation of the results and for the different volumes of reagents used. Similar results of agreement (Kappa index = 1) were obtained when using slides or Petri dishes as support for the agar gel. In both cases, an almost perfect agreement was achieved, suggesting that the use of different supports does not affect the results. These findings indicate that both protocols (MAPA and oIE) can be used to diagnose EIA.
