Zebrafish as a possible bioindicator of organic pollutants with effects on reproduction in drinking waters

Organic contaminants can be detected at low concentrations in drinking water, raising concerns for human health, particularly in reproduction. In this respect, we attempted to use the zebrafish as a bioindicator to detect the possible presence of these substances in drinking water, aiming to define the most relevant parameters to detect these substances, which particularly affect the development and reproduction of zebrafish. To this end, batches of30 embryos with the chorion intact were cultured in drinking waters from different sources,throughout their full life-cycle up to 5 months, in 20 L tanks. Six replicates were performed in all water groups, with a total of 24 aquariums. Two generations（F0 and F1） were studied and the following parameters were tested： in the F0 generation, survival and abnormality rates evaluated at 5 dpf（days post-fertilization） and at 5 mpf（months post-fertilization）,the onset of spawning and the fertility rate from 3 mpf to 5 mpf, and the sex ratio and underdeveloped specimens at 5 mpf. Furthermore, in the F0 offspring（F1）, survival and abnormality rates were evaluated at 5 dpf and the hatching rate at 72 hpf. These results revealed that the hatching rate is the most sensitive parameter to distinguish different levels of effects between waters during the early life stages, whereas the rate of underdeveloped specimens is more suitable at later life stages. Regarding adult reproduction, fertility rate was the most sensitive parameter. The possible reversibility or accumulative nature of such effects will be studied in future work.

Independent effects of diet and exercise training on fat oxidation in non-alcoholic fatty liver disease

AIm To investigate the independent effects of 6-mo of dietary energy restriction or exercise training on wholebody and hepatic fat oxidation of patients with nonalcoholic fatty liver disease(NAFLD).mETHODS Participants were randomised into either circuit exercise training(EX;n = 13;3 h/wk without changes in dietary habits),or dietary energy restriction(ER) without changes in structured physical activity(ER;n = 8).Respiratory quotient(RQ) and whole-body fat oxidation rates(Fatox) were determined by indirect calorimetry under basal,insulin-stimulated and exercise *** of disease and steatosis was determined by liver histology;hepatic Fatox was estimated from plasma β-hydroxybutyrate ***;cardiorespiratory fitness was expressed as VO2 ***-case analysis was performed(EX:n = 10;ER:n = 6).RESULTS Hepatic steatosis and NAFLD activity score decreased with ER but not with EX.β-hydroxybutyrate concentrations increased significantly in response to ER(0.08 ± 0.02 mmol/L vs 0.12 ± 0.04 mmol/L,P = 0.03) but remained unchanged in response to EX(0.10 ± 0.03 mmol/L vs 0.11 ± 0.07 mmol/L,P = 0.39).Basal RQ decreased(P = 0.05) in *** EX,while this change was not significant after ER(P = 0.38).VO_(2peak)(P m.xim.l Fa_(tox) during aerobic exercise(P = 0.03) improved with EX but not with ER(P > 0.05).The increase in β-hydroxybutyrate concentrations was correlated with the reduction in hepatic steatosis(r =-0.56,P = 0.04).CONCLUSION ER and EX lead to specific benefits on fat metabolism of patients with *** hepatic Fat_(ox) in response to ER could be one mechanism through which the ER group achieved reduction in steatosis.

AIm:To study the action of aminoguanidine on pancreatic cancer xenografts in relation to cell proliferation,apoptosis,redox status and ***:Xenografts of PANC-1 cells were developed in nude mice. The animals were separated into two groups:control and aminoguanidine treated. Tumor growth,survival and appearance of metastases were determined in vivo in both groups. Tumors were excised and ex vivo histochemical studies were performed. Cell growth was assessed by Ki-67 expression. Apoptosis was studied by intratumoral expression of B cell lymphoma-2 protein (Bcl-2) family proteins and Terminal deoxynucleotidyl transferase biotin-dUTP Nick End Labeling (Tunel). Redox status was evaluated by the expression of endothelial nitric oxide synthase (eNOS),catalase,copper-zinc superoxide dismutase (CuZnSOD),manganese superoxide dismutase (mnSOD) and glutathione peroxidase (GPx). Finally,vascularization was determined by massons trichromic staining,and by VEGF and CD34 ***:Tumor volumes after 32 d of treatment by aminoguanidine (AG) were significantly lower than in control mice (P median survival of AG mice was significantly greater than control animals (P molateral and contralateral palpable metastases was significantly delayed in AG group. Apoptotic cells,intratumoral vascularization (trichromic stain) and the expression of Ki-67,Bax,eNOS,CD34,VEGF,catalase,CuZnSOD and mnSOD were diminished in AG treated mice (P moral action of aminoguanidine is associated with decreased cell proliferation,reduced angiogenesis,and reduced expression of antioxidant enzymes.

为了提高农业用水效率,解决用水矛盾,2013-2016年在陕西泾惠渠灌区连续进行了4年的冬小麦非充分灌溉制度试验,试验共设置8个非充分灌水处理,每个处理重复2次。通过分析不同生育期灌水量对冬小麦株高、产量、水分利用效率及经济效益在不同降雨频率年型下的影响,确定了泾惠渠灌区合理的冬小麦非充分灌溉制度及模式。试验结果表明:不同生育期灌水量对小麦生长发育和产量有不同的影响。返青灌、拔节灌、抽穗灌均有利于冬小麦的生长发育,其中返青灌的影响尤为显著;返青灌和灌浆灌是影响冬小麦产量的重要时期,返青灌也是提高水分利用效率和经济效益的敏感期。平水年和枯水年降雨年型下,追求高产的最优灌溉模式是冬灌+拔节灌+灌浆灌,灌溉定额为3 600 m3/hm2;然而综合考虑产量、灌溉水利用系数及节水经济效益的最优灌溉模式是冬灌+返青灌,灌溉定额为2 550 m3/hm2;推荐泾惠渠灌区非充分灌溉模式为冬灌+返青灌。研究成果为泾惠渠灌区提供了具体可行的灌溉模式,为非充分灌溉制度的进一步完善奠定了基础,对灌区农业用水的合理配置具有一定的指导意义。

Background and aims: Diverticulosis is a common disease of not completely defined pathogenesis. motor abnormalities of the intestinal wall have been frequently described but very little is known about their mechanisms. We investigated in vitro the neural response of colonic longitudinal muscle strips from patients undergoing surgery for complicated diverticular disease (diverticulitis). methods: The neural contractile response to electrical field stimulation of longitudinal muscle strips from the colon of patients undergoing surgery for colonic cancer or diverticulitis was challenged by different receptor agonists and antagonists. Results: Contractions of colonic strips from healthy controls and diverticulitis specimens were abolished by atropine. The βadrenergic agonist (-) isoprenaline and the tachykinin NK1 receptor antagonist SR140333 had similar potency in reducing the electrical twitch response in controls and diseased tissues, while the cannabinoid receptor agonist (+)WIN 55,212-2 was 100 times more potent in inhibiting contractions in controls (IC50 42 nmol/l) than in diverticulitis strips. SR141716, a selective antagonist of the cannabinoid CB1 receptor, had no intrinsic activity in control preparations but potentiated the neural twitch in diseased tissues by up to 196%in a concentration dependent manner. SR141716 inhibited (+)WIN 55,212-2 induced relaxation in control strips but had no efficacy on (+)WIN 55,212-2 responses in strips from diverticular disease patients. Colonic levels of the endogenous ligand of cannabinoid and vanilloid TRPV1 receptors anandamide were more than twice those of control tissues (54 v 127 pmol/g tissue). The axonal conduction blocker tetrodotoxin had opposite effects in the two preparations, completely inhibiting the contractions of control strips but potentiating those in diverticular preparations, an effect selectively inhibited by SR140333. 为了建立一种快速、特异、敏感的检测牛支原体血清抗体的方法,对牛支原体(***)P48膜蛋白基因进行密码子优化,并在编码基因两端加入酶切位点。利用生物学软件对P48蛋白进行抗原位点和亲水性分析,选择P48蛋白的主要抗原表位区和亲水性区域以及全基因进行原核表达。采用SDS-PAGE和Western blot对重组蛋白进行鉴定及反应原性分析。结果显示,表达的P48全蛋白、28～185、221～455位氨基酸区域蛋白均能与牛支原体阳性血清发生特异性反应,P48(221～455)效果最好。采用Ni-NTA对目的蛋白进行纯化,并基于纯化的P48(221～455)蛋白建立了一种间接ELISA检测方法。该方法组内及组间变异系数均小于7%,重复性较好。临床样本检测结果表明,建立的检测方法符合率较高。
