Expression of Cyclooxygenase-2 and Ki-67 in Actinic Keratosis and Cutaneous Squamous Cell Carcinoma in Dogs

Actinic keratosis is a common disease in humans, which also affects dogs. Lesions occur in chronically sun exposed areas, such as flank, ventral and lateral abdomen. It has been reported that actinic keratosis is a pre-neoplastic disease which may evolve into squamous cell carcinoma (SCC), which is one of the most frequent malignant neoplasm in dogs. The aim of this research was to investigate the relationship between COX-2 and cell proliferation on the outcome of dogs with actinic keratosis and cutaneous squamous cell carcinoma. This study included 10 skin sections of actinic keratosis (G1) and 10 cutaneous SCC (G2). Data including age, breed, gender and histopathological findings were documented. Paraffin-embedded tissues were retrieved for COX-2 and Ki-67 staining. American Pit Bull Terrier dogs were the most affected ones in both G1 and G2, the mean age was 4.3 (±0.8) years and 5.6 (±1.7) years, respectively. mean score of COX-2 immunostaining in G1 and G2 was 8.16 (±3.51) and 8.56 (±1.03), respectively. mean percentage of immunopositive cells for Ki-67 in G1 and G2 was 15.77 (±8.81) and 17.71 (±12.21), respectively. There was no association between COX-2 expression and Ki-67, recurrence, survival and metastasis rate (p > 0.05). These findings highlight the role of COX-2 and Ki-67 in carcinogenesis, but do not confirm the relationship between COX-2 expression and increased cell proliferation in dogs. COX-2 may play a role in carcinogenesis, but this pathway is not responsible for cellular proliferation in actinic keratosis and cutaneous SCC in dogs. This study investigated the action of proteolytic enzymes in the production of goat viscera hydrolysates,and their flavoring *** study focus the production of flavor precursor molecules and the formation of volatile compounds resulting from four hydrolysis treatments:HA-Alcalase®,HF-Flavourzyme®,HN-Neutrase®,and HFN-Flavourzyme® and Neutrase®.The highest degree of hydrolysis (26.74%),number (23 volatiles) and concentration of volatile compounds (274.0 μg/L) were obtained with the action of *** main classes of volatile compounds identified in hydrolysates included acids,aldehydes,pyrazines,terpenes and Streacker *** acids,sugars and fatty acids were identified with higher concentrations in HF and HFN hydrolysates due to the low participation of these precursors in the formation of volatile ***,the use of Alcalase in the proteolysis of goat viscera stood out compared to other enzymes,providing impacting aroma compounds,such as pyrazines,for meat flavor.

Gastric cancer in a Caucasian population: Role of pepsinogen C genetic variants

AIm： To study the role of an insertion/deletion polymorphism in the pepsinogen C （PGC） gene, an effective marker for terminal differentiation of the stomach mucosa, in the susceptibility to the development of gastric lesions. mETHODS： The study was performed with 99 samples of known gastric lesions and 127 samples without evidence of neoplastic disease. PCR was employed and the 6 polymorphic alleles were amplified： Allele 1 （510 bp）, Allele 2 （480 bp）, Allele 3/4 （450/460 bp）, Allele 5 （400 bp） and Allele 6 （310 bp）. RESULTS： Our results revealed that Allele 6 carriers seemed to have protection against the development of any gastric lesion （OR = 0.34; P 〈 0.001）, non-dysplastic lesions associated with gastric adenocarcinoma such as atrophy or intestinal metaplasia （OR = 0.28; P 〈 0.001） or invasive GC （OR = 0.39; P = 0.004）. CONCLUSION： Our study reveals that the Allele 6 carrier status has a protective role in the development of gastric lesions, probably due to its association with higher expression of PGC. moreover, the frequency of Allele 6 carriers in the control group is far higher than that obtained in Asian populations, which might represent a genetic gap between Caucasian and Asian populations.

The loss of biodiversity and ecosystem services disturbance will result in serious ecological and socioeconomic consequences. How can people be prepared to be more efficient in preserving ecosystems, its services and resources? The messalo Wilderness Area (mWA) is an area lacking a bottom-up approach for the design of a management plan. This research intended to develop a strategy to gather people’s views to begin a human wellbeing assessment for the area, for which a Digital Questionnaire (DQ) was developed and tested. The specific objectives were to 1) verify the dispersion/distribution of people and elephants;2) study the natural resources used by the inhabitants;3) locate the areas from where the most important natural resources are extracted and where conflict between humans and animals occur;4) propose a management Plan (mP) capable of mitigating the human-animals conflict, promoting the conservation of elephants and, consequently, of other types of biodiversity, thus working towards a better socio-economical development of the area. The elements gathered in the consulted references were used to design several maps and cartographic figures shown in this article, using the map Window software. These maps describe: fresh water distribution, habitat distribution, population dispersal and main road connections, crop fields distribution and main exploitation spots of the different resources, orography and topography. The results from the DQ analysis helped to gather indicators to the human wellbeing assessment. The information gathered in the literature and by inquiry was effective in confirming the high dependency of local people on land and sea resources, as well as the conflicts between people and wild animals, such as elephants and African wild dogs, and the reasons for those conflicts. The DQ is effective in gathering people’s opinions and it constitutes an important tool in a bottom-up approach to the design of a management plan as to the design of the mP for

Lack of correlation between p53 codon 72 polymorphism and anal cancer risk

AIm:To investigate the potential role of p53 codon 72 polymorphism as a risk factor for development of anal cancer. mETHODS:Thirty-two patients with invasive anal carcinoma and 103 healthy blood donors were included in the study.p53 codon 72 polymorphism was analyzed in blood samples through polymerase chain reaction-restriction fragment length polymorphism and DNA sequencing. RESULTS:The relative frequency of each allele was 0.60 for Arg and 0.40 for Pro in patients with anal cancer, and 0.61 for Arg and 0.39 for Pro in normal controls. No significant differences in distribution of the codon 72 genotypes between patients and controls were found. CONCLUSION:These results do not support a role for the p53 codon 72 polymorphism in anal carcinogenesis.

Based on reports that variables related to photosynthesis(net assimilatory ratio and chloroplast pigments)are more plastic than the morphological and biochemical variables in a climax tropical species,Cariniana estrellensis,under contrasting irradiance,we hypothesised that plasticity of gas exchange variables is higher in relation to growth,biochemical and anatomical *** that were 14 months old were subjected to200 up to 2000 lmol m^(-2)s^(-1) of photo synthetically active radiation(PAR)for 140 *** increased up to1400 lmol m^(-2)s^(-1)PAR and then decreased at 2000 lmol m^(-2)s^(-1) PAR(full sunlight).This was related to the rate of photosynthesis(A)which showed the same tendency as photosystem Ⅱ performance index(PI_(ABS)).The greater thickness of the secondary xylem at 1400 lmol m^(-2)s^(-1) PAR indicated a higher water conductance *** decrease in specific leaf area,elongation of mesophyll cells,and increase in stomatal density observed at higher irradiances were associated with higher leaf fructose and sucrose *** concluded that the better growth in partial shade was due to higher photosynthetic *** the other hand,photosynthesis was inhibited equally in full sun and deep *** high adjustment capacity of *** to wide amplitudes of irradiance was provided by greater plasticity of gas exchange variables in relation to growth,anatomical,and biochemical variables.

Early dynamic transcriptomic changes during preoperative radiotherapy in patients with rectal cancer: A feasibility study

AIm: To develop novel biomarkers of rectal radiotherapy, we measured gene expression profiles on biopsies taken before and during preoperative radiotherapy. mETHODS: Six patients presenting with a locally advanced rectal cancer (T>T2, N0/Nx, m0) eligible for preoperative radiotherapy (45 Gy in 25 fractions) were selected in a pilot study. Six tumor and 3 normal tissues biopsies were taken before and during radiotherapy,after a dose of 7.2 Gy at a median time of 1 h following irradiation (0:27-2:12). Tumor or normal tissue purity was assessed by a pathologist prior to RNA extraction. mean RNA content was 23 μg/biopsy (14-37) before radiotherapy and 22.7 μg/biopsy (12-35) during radiotherapy. After RNA amplification, biopsies were analysed with 54K HG-U133A Plus 2.0 Affymetrix expression micro-arrays. Data were normalized according to mAS5 algorithm. A gene expression ratio was calculated as: (gene expression during radiotherapy-gene expression before radiotherapy)/gene expression before radiotherapy. Were selected genes that showed a ratio higher than ± 0.5 in all 6 patients. RESULTS: microarray analysis showed that preoperative radiotherapy significantly up-regulated 31 genes and down-regulated 6 genes. According to the Gene Ontology project classification, these genes are involved in protein metabolism (ADAmDEC1 ; AKAP7 ; CAPN5 ; CLIC5 ; CPE ; CREB3L1 ; NEDD4L ; RAB27A), ion transport (AKAP7 ; ATP2A3 ; CCL28 ; CLIC5 ; F2RL2 ; NEDD4L ; SLC6A8), transcription (AKAP7 ; CREB3L1 ; ISX ; PAB-PC1L ; TXNIP), signal transduction (CAPN5 ; F2RL2 ; RA- B27A ; TNFRSF11A), cell adhesion (ADAmDEC1 ; PXDN ; SPON1 ; S100A2), immune response (CCL28 ; PXDN ; TNFRSF11A) and apoptosis (ITm2C ; PDCD4 ; PVT1). Chronic Diseases of Childhood and the International Classification of Functioning, Disability, and Health: A Systematic Review

The increase in chronic diseases in childhood highlights the need for a biopsychosocial approach to deal with the complexity of these health conditions. The International Classification of Functioning, Disability, and Health (ICF) results from the need to implement new explanatory evaluative and therapeutic models. Thus, the present systematic review aims to identify published studies on the use of the ICF in chronic childhood diseases. As a secondary objective, to map the themes that have already been studied in the area to support the discussion on the expansion of the use of this classification in health services. The systematic review followed the PRISmA protocol, and the model was the PICO acronym, where Population was children and adolescents with chronic diseases, Intervention/Exposure was evaluation based on ICF concepts, Comparator was any tool, instrument, or intervention, and outcome was direct or indirect use of the ICF. We selected eight articles, five of which used the ICF as a conceptual tool and three as a classification system, divided into the following themes: quality of life, evaluation of patients (without using coding) and mapping the inclusion of the activity’s results and participation in clinical trials. Thus, use of the ICF in clinical practice is still incipient, although it has been recommended in guidelines. Further studies are necessary to expand the number of contributions by the ICF and to develop the necessary approaches for understanding the classification’s use.
