A titrant for the SARS-CoV-2 main protease(m^(pro))was developed that enables,for the first time,the exact determination of the concentration of the enzymatically active m^(pro) by active-site *** covalent binding mode of the tetrapeptidic titrant was elucidated by the determination of the crystal structure of the enzyme–titrant *** fluorogenic substrates of m^(pro),including a prototypical,internally quenched Dabcyl-eDANS peptide,were compared in terms of solubility under typical assay *** exploiting the new titrant,key kinetic parameters for the m^(pro)-catalyzed cleavage of these substrates were determined.

The ability to learn incrementally is critical to the long-term operation of AI systems. Benefiting from the power of few-shot class-incremental learning(FSCIL), deep learning models can continuously recognize new classes with only a few samples. The difficulty is that limited instances of new classes will lead to overfitting and exacerbate the catastrophic forgetting of the old classes. most previous works alleviate the above problems by imposing strong constraints on the model structure or parameters, but ignoring embedding network transferability and classifier adaptation(CA), failing to guarantee the efficient utilization of visual features and establishing relationships between old and new classes. In this paper, we propose a simple and novel approach from two perspectives: embedding bias and classifier bias. The method learns an embedding augmented(eA) network with cross-class transfer and class-specific discriminative abilities based on self-supervised learning and modulated attention to alleviate embedding bias. Based on the adaptive incremental classifier learning scheme to realize incremental learning capability,guiding the adaptive update of prototypes and feature embeddings to alleviate classifier bias. We conduct extensive experiments on two popular natural image datasets and two medical datasets. The experiments show that our method is significantly better than the baseline and achieves state-of-the-art results.

Background In beef cattle, more than 50% of the energy input to produce a unit of beef is consumed by the female that produced the calf. Development of genomic tools to identify females with high genetic merit for reproductive function could increase the profitability and sustainability of beef *** Genome-wide association studies(GWAS) were performed using a single-step genomic best linear unbiased prediction approach on pregnancy outcome traits from a population of Angus–Brahman crossbred heifers. Furthermore, a validation GWAS was performed using data from another farm. Heifers were genotyped with the Bovine GGP F250 array that contains 221,077 SNPs. In the discovery population, heifers were bred in winter breeding seasons involving a single round of timed artificial insemination(AI) followed by natural mating for 3 months. Two phenotypes were analyzed: pregnancy outcome to first-service AI(PAI;n = 1,481) and pregnancy status at the end of the breeding season(PeBS;n = 1,725). The heritability was estimated as 0.149 and 0.122 for PAI and PeBS, respectively. In the PAI model, one quantitative trait locus(QTL), located between 52.3 and 52.5 mb on BTA7, explained about 3% of the genetic variation, in a region containing a cluster of γ-protocadherin genes and SLC25A2. Other QTLs explaining between 0.5% and 1% of the genetic variation were found on BTA12 and 25. In the PeBS model, a large QTL on BTA7 was synonymous with the QTL for PAI, with minor QTLs located on BTA5, 9, 10, 11, 19, and 20. The validation population for pregnancy status at the end of the breeding season were Angus–Brahman crossbred heifers bred by natural mating. The high-temperature oxidation resistance of the nickel superalloy grown by the Laser Powder Bed Fusion (LPBF) has been significantly increased as a result of in-situ formation of a thermal barrier layer (α-Al2O3+ CamoO4) during oxidative annealing of surface layers modified by electric spark treatment (eST). The reactive eST of the LPBF-built items based on nickel eP741NP alloy was carried out with low-melting Al-12%Si， Al-6%Ca-0.6%Si and Al-7%Ca-1%mn electrodes. It was found that at eST done by Al-7%Ca-1%mn electrode an intermetallic (β-NiAl + γ’-Ni3Al) 15 microns thick layer reinforced by spherical oxide (Came)O nanoparticles was formed. Formation of that structure increases the wear resistance of LPBF nickel superalloy by 4.5 times. Further oxidative annealing at 1000 °C lead to a formation of continuous two-layered coating with an inner layer of α-Al2O3and an outer layer of CamoO4， which together act as an effective barrier preventing the diffusion of oxygen into the volume of the superalloy.

The main objective of this paper is to investigate the influence of inertia of nonlinear springs on the dispersion behavior of discrete monoatomic chains with lumped and distributed *** developed model can represent the wave propagation problem in a non-homogeneous material consisting of heavy inclusions embedded in a *** inclusions are idealized by lumped masses,and the matrix between adjacent inclusions is modeled by a nonlinear spring with distributed ***,the model is capable of depicting the wave propagation in bi-material bars,wherein the first material is represented by a rigid particle and the second one is represented by a nonlinear spring with distributed *** discrete model of the nonlinear monoatomic chain with lumped and distributed masses is first considered,and a closed-form expression of the dispersion relation is obtained by the second-order Lindstedt-Poincare method(LPm).Next,a continuum model for the nonlinear monoatomic chain is derived directly from its discrete lattice model by a suitable continualization *** subsequent use of the second-order method of multiple scales(mmS)facilitates the derivation of the corresponding nonlinear dispersion relation in a closed *** novelties of the present study consist of(i)considering the inertia of nonlinear springs on the dispersion behavior of the discrete mass-spring chains;(ii)developing the second-order LPm for the wave propagation in the discrete chains;and(iii)deriving a continuum model for the nonlinear monoatomic chains with lumped and distributed ***,a parametric study is conducted to examine the effects of the design parameters and the distributed spring mass on the nonlinear dispersion relations and phase velocities obtained from both the discrete and continuum *** parameters include the ratio of the spring mass to the lumped mass,the nonlinear stiffness coefficient of the spring,and the wave amplitude.

Red blood cells(RBCs)are the most abundant human blood *** aggregation and deformation strongly determine blood viscosity which impacts hemorheology and *** turn,RBC properties depend on di®erent endogenous and exogenous *** such factor is nitric oxide(NO),which is mainly produced by endothelial cells(eC)from L-arginine amino acid in the circulatory *** the mechanisms of the RBC-endothelium interplay are not clear up to date and considering its possible clinical importance,the aims of this study are to investigate in vitro:(1)The e®ect of L-arginine induced NO on RBC aggregation and adhesion to endothelium;(2)the NO e®ect on RBC aggregation and deformation induced by L-arginine and sodium nitroprusside without the presence of endothelium in the *** RBC aggregation and adhesion to a monolayer of eC were studied using optical tweezers(OT).The RBC deformability and aggregation without endothelium in the samples were studied using the°ow chamber method and myrenne *** con¯rmed that NO increases deformability and decreases aggregation of *** showed that the soluble guanylate cyclase pathway appears to be the only NO signaling pathway *** the samples with the endothelium,the\bell-shaped"dependence of RBC aggregation force on L-arginine concentration was observed,which improves our knowledge about the process of NO production by ***,data related to L-arginine accumulation by endothelium were obtained:Necessity of the presence of extracellular L-arginine stated by other authors was put under *** our study,NO decreased the RBC-endothelium adhesion,however,the tendency appeared to be weak and was not con¯rmed in another set of *** our knowledge,this is the¯rst attempt to measure the forces of RBC adhesion to endothelium monolayer with OT.

In this editorial,we comment on the article by Lei et al,with a specific focus on the timing of the initiation of the antifibrotic agent pirfenidone(PFD)in the management of idiopathic pulmonary fibrosis(IPF)and its impact on lung function of IPF *** is an antifibrotic agent that is widely used in the management of IPF in both early and advanced *** inhibits various pathways and has antifibrotic,anti-inflammatory,and antioxidant *** dosage lowering,PFD slowed IPF progression and maintained functional *** 6-min walk distance test indicated that patients tolerated adverse events well,and PFD significantly reduced the incidence of progression episodes and *** when a single disease-progression event occurred,continuing PFD treatment had benefits.

Alzheimer’s disease (AD) and associated dementia patient numbers continue to increase globally with associated economic costs to healthcare systems. Of note is the increase in numbers in lower and middle-income countries (LmICs) including Sub-Saharan African (SSA) countries, which already face challenges with their health budgets from communicable and non-communicable diseases. Ghana, an SSA country, faces the problem of healthcare budgetary difficulties and the additional impact of AD as a consequence of increasing population strata of old aged persons (OAPs) due to the demographic transition effect. This article uses examples of known patients’ illness courses to give a perspective on the lived experience of patients with dementia (PWD) in Ghana, living amongst a populace with a culture of stigmatization of PWD, and a relatively fragile public mental health system (PmHS) for those with mental illness, including AD. The lived experience of AD patients is characterised by stigmatisation, discrimination, non-inclusiveness, diminished dignity and human rights abuses in the face of their mental disability, and eventually death. This article is an advocacy article giving voice to the voiceless and all persons suffering from AD and other dementias in Ghana, whilst pleading for a call to action from healthcare professionals and responsible state agencies.

Frequency-dependent viscoelasticity effects on the wave attenuation performance of multi-layered periodic foundations

In this paper,layered periodic foundations(LPFs)are numerically examined for their responses to longitudinal and transverse modes in the time and frequency *** different unit-cells,i.e.,2-layer,4-layer,and 6-layer unit-cells,comprising concrete/rubber,concrete/rubber/steel/rubber,and concrete/rubber/steel/rubber/lead/rubber materials,respectively,are taken into ***,the viscoelasticity behavior of the rubber is modeled with two factors,i.e.,a frequency-independent(FI)loss factor and a linear frequency-dependent(FD)loss *** the extraction of the complex dispersion curves and the identification of the band gaps(BGs),the simulations of wave transmission in the time and frequency domains are performed using the COmSOL *** parametric studies evaluate the effects of the rubber viscoelasticity models on the dispersion curves and the wave transmission for the longitudinal and transverse *** results show that considering the rubber viscoelasticity enhances the wave attenuation ***,the transverse-mode damping is more sensitive to the viscoelasticity model than its longitudinal *** 6-layer unit-cell LPF exhibits the lowest BG,ranging from 4.8 Hz to 6.5 Hz.
