The effects of contemporary climate,habitat heterogeneity and long-term climate change on species richness are well studied for woody plants in forest ecosystems,but poorly understood for herbaceous plants,especially in alpine–arctic ***,we aim to test if the previously proposed hypothesis based on the richness–environment relationship could explain the variation in richness patterns of the typical alpine–arctic herbaceous genus *** a newly compiled distribution database of 437 Saxifraga species,we estimated the species richness patterns for all species,narrow-and wide-ranged *** used generalized linear models and simultaneous autoregressive models to evaluate the effects of contemporary climate,habitat heterogeneity and historical climate on species richness *** regressions were used to determine the independent and shared effects of different *** widely used models were tested to identify their predictive power in explaining patterns of species *** found that temperature was negatively correlated with the richness patterns of all and wide-ranged species,and that was the most important environmental factor,indicating a strong conservatism of its ancestral temperate *** heterogeneity and long-term climate change were the best predictors of the spatial variation of narrow-ranged species ***,the combined model containing five predictors can explain ca.40%–50%of the variation in species *** further argued that additional evolutionary and biogeographical processes might have also played an essential role in shaping the Saxifraga diversity patterns and should be considered in future studies.

Replication of natural surface topographies to generate advanced cell culture substrates

Surface topographies of cell culture substrates can be used to generate in vitro cell culture environments similar to the in vivo cell niches. In vivo, the physical properties of the extracellular matrix (ECm), such as its topography, provide physical cues that play an important role in modulating cell function. mimicking these properties remains a challenge to provide in vitro realistic environments for cells. Artificially generated substrates’ topographies were used extensively to explore this important surface cue. more recently, the replication of natural surface topographies has been enabling to exploration of characteristics such as hierarchy and size scales relevant for cells as advanced biomimetic substrates. These substrates offer more realistic and mimetic environments regarding the topographies found in vivo. This review will highlight the use of natural surface topographies as a template to generate substrates for in-vitro cell culture. This review starts with an analysis of the main cell functions that can be regulated by the substrate’s surface topography through cell-substrate interactions. Then, we will discuss research works wherein substrates for cell biology decorated with natural surface topographies were used and investigated regarding their influence on cellular performance. At the end of this review, we will highlight the advantages and challenges of the use of natural surface topographies as a template for the generation of advanced substrates for cell culture.

Dexamethasone intravitreal implant(Ozurdex) in diabetic macular edema: real-world data versus clinical trials outcomes

AIm: To investigate the safety and efficacy of intravitreal dexamethasone implants(Ozurdex?/DEX) in patients with diabetic macular edema(DmE) either na?ve or nonna?ve to anti-VEGF therapies who switched to DEX implant independent of response to anti-vascular endothelial growth factors(anti-VEGFs).mETHODS: This was an audit retrospective review of medical records of patients with DmE who switched to the DEX intravitreal implant. Patients were divided into 2 groups: patients na?ve to antiangiogenic therapy and patients who were previously treated with anti-VEGFs. Data regarding demographics, changes in mean best-corrected visual acuity(BCVA), central macular thickness(CmT), and intraocular pressure(IOP) was collected over 6 mo. The demographic data mean changes in BCVA, CmT, and IOP were compared. Six-month follow-up data of 47 patients(57 eyes), who either switched to DEX implant irrespective of response to previous treatments or were treatment na?ve before receiving DEX implant, was ***: Improvement in mean BCVA was observed from 1-4 mo after injection with a decreased effect at month 6 as expected, with better outcomes in na?ve compared to non-na?ve patients. A statistically relevant decrease in mean CmT was observed during the follow-up period. An increase in mean IOP was observed in the first 2 mo after DEX therapy. The mean number of injections of the overall population during the 6 mo was 1.3. A subgroup analysis showed no relevant difference between phakic versus pseudophakic patients relative to measured outcomes. There was no cataract progression during the follow-up period and no adverse events ***: This real-life setting study shows that intravitreal DEX implant is effective and safe. The timings of greater therapeutic impact are concordant with previous studies and suggest that earlier treatment with corticosteroids may have an additional benefit in na?ve patients.

magnesium-based materials show great potential for producing biodegradable stents,but their high corrosion rates are a *** study investigates whether ultrasound melt treatment can change the corrosion response of an extruded AZ91D-1.0%Ca(wt.%)in Earle's Balanced Salt Solution by tailoring the intermetallics'morphology in the as-extruded *** results showed that the wires from ultrasound-treated ingots corroded faster than non-treated ones in immersion for up to 6 *** trend shifted for longer periods,and ultrasound-treated material showed lower corrosion rates and uniform corrosion,while the non-treated material displayed localized corrosion *** testing of the wires demonstrated that immersion in EBSS lowered the tensile strength and elongation at fracture due to material degradation,regardless of the processing ***,this decline was sharper in the non-treated *** findings suggest that ultrasound melt processing can be a promising method for improving the corrosion resistance of magnesium-based materials,paving the way for their use in manufacturing biodegradable stents.

The tissue engineering triad comprises the combination of cells,scaffolds and biological ***,we prepared cell-and drug-loaded hydrogels using in situ silk fibroin(SF)hydrogels induced by dimyristoyl glycerophosphoglycerol(DmPG).DmPG is reported to induce rapid hydrogel formation by SF,facilitating cell encapsulation in the hydrogel matrix while maintaining high cell viability and proliferative *** addition,DmPG can be used for liposome formulations in entrapping drug ***(Dex)was loaded into the DmPG-induced SF hydrogels together with human osteoblast-like SaOS-2 cells,then the osteogenic differentiation of the entrapped cells was evaluated in vitro and compared to cells cultured under standard *** production by cells cultured in DmPG/Dex-SF hydrogels with Dex-depleted osteogenic medium was equivalent to that of cells cultured in conventional osteogenic medium containing *** extended-release of the entrapped Dex by the hydrogels was able to provide a sufficient drug amount for osteogenic *** controlled release of Dex was also advantageous for cell viability even though its dose in the hydrogels was far higher than that in osteogenic *** results confirmed the possibility of using DmPG-induced SF hydrogels to enable dual cell and drug encapsulation to fulfil the practical applications of tissue-engineered constructs.

Helium ion scanning microscopy(HIm)is a novel high-resolution scanning microscopy technique that uses helium ions instead of electrons to form images of the highest quality and resolution,providing a sub-nanometer resolution sputter uncoated biological ***,we took advantage of HIm to explore the cell surface of Tritrichomonas foetus,a protist parasite of cattle that provokes hard infection and abortion in *** describe thin protrusions,like nanotubes described in other cells,with different sizes(27 nm to 81 nm in thickness)and various lengths(from 73 nm to 2μm),as well bulbous structures either budding from the cell surface or present in the extremities of some *** flagella also presented these thin protrusions and different protein decoration,similar to those previously described using freeze-fracture *** between two cells were also seen,and their role in infection is *** cell surface was also examined and showed several pits indicative of endocytic activity and other types of arrays of *** observations were confirmed using Scanning Electron microscopy(SEm),negative staining,and conventional thin sectioning for observation by transmission electron *** findings provide new and relevant information that may contribute to a better understanding of protozoan biology and its interaction with mammalian cells.

Comparative stability analyses of traditional and selective room-and-pillar mining techniques for sub-horizontal tungsten veins

The stability and productivity concerning a modification on the traditional room and pillar for a new selective technique at the Portuguese Panasqueira mine have been *** traditional room-and-pillar stoping uses 5.0-m wide rooms with 3.0 m×3.0 m pillars,while the selective room-and-pillar mining technique consists in stoping with rooms of 4.0 m wide and pillars of 4 m×4 m with a subsequent selective cutting of the quartz veins at the mid pillar of approximately 0.5 m high,to obtain a pillar section with an area of 3.0 m×3.0 *** stability and productivity analyses indicate that the selective technique obtains smaller average pillar safety factor,more rock mass displacement,more extraction and selectivity ratios,and ore grade improvement,compared with the traditional *** results show that the selective technique is also more *** proposed selective room-and-pillar mining technique is applicable to any sub-horizontal narrow quartz veins with wolfram,gold,*** as the famous La Rinconada gold mine in the Peruvian Andes.
