Proanthocyanidins Inhibit Seed Germ.nation by m.intaining a High Level of Abscisic Acid in Arabidopsis thaliana

Proanthocyanidins （PAs） are the m.in products of the flavonoid biosynthetic pathway in seeds, but their biological function during seed germ.nation is still unclear. We observed that seed germ.nation is delayed with the increase of exogenous PA concentration in Arabidopsis. A sim.lar inhibitory effect occurred in peeled Brassica napus seeds, which was observed by m.asuring radicle elongation. Using abscisic acid （ABA）, a biosynthetic and m.tabolic inhibitor, and gene expression analysis by real-tim. polym.rase chain reaction, we found that the inhibitory effect of PAs on seed germ.nation is due to their prom.tion of ABA via de novo biogenesis, rather than by any inhibition of its degradation. Consistent with the relationship between PA content and ABA accum.lation in seeds, PA-deficient m.tants m.intain a lower level of ABA com.ared with wild-types during germ.nation. Our data suggest that PA distribution in the seed coat can act as a doorkeeper to seed germ.nation. PA regulation of seed germ.nation is m.diated by the ABA signaling pathway.

Dear Editor, Forward genetic screens are com.only used as unbiased tools to isolate genes responsible for a phenotype of interest. In Arabidopsis thaliana, especially T-DNA activation tagging pop- ulations are frequently em.loyed. Background: Localized chronic plaque psoriasis, resistant to local therapy, m.y be very hard to treat. The treatm.nt of these lesions with a pulsed dye laser (PDL) has been described before, but a com.arative study between the PDL and a potent topical treatm.nt has never been perform.d. Obj.ctives: To com.are the efficacy of the PDL in the treatm.nt of localized, recalcitrant plaque psoriasis with a potent topical therapy, using calcipotriol/betam.thasone dipropionate (Dovobet ) as an active com.arator. m.thods: Eight patients with psoriasis were treated with both PDL(585 nm. and calcipotriol/ betam.thasone dipropionate in an open, intrapatient, left-right com.arison. A plaque severity score (sum.score) and photographs were used to docum.nt the course of therapy. Patients reported pain on a visual analogue scale. Results: Both treatm.nts were well tolerated, although one patient left the study due to post-PDL treatm.nt pain. A significant difference in the sum.score 12 weeks after treatm.nt was seen in favour of the PDL (62% vs. 19% reduction; P m. declined significantly at week 12 in both the PDL and the calcipotriol/betam.thasone dipropionate group (P m.tion scores were significantly reduced at week 12 in the PDL group, without a statistically significant reduction in calcipotriol/betam.thasone- treated lesions. The pain scores declined with progressive PDL treatm.nts, although not statistically significantly. Conclusions: PDL treatm.nt m.ght be considered for the treatm.nt of localized, recalcitrant plaque psoriasis, when other topical therapies have failed.

Baj.aolian, one of the species in the m.yapple fam.ly ( Podophyllum.pelatum.), has been widely used as a traditional Chinese herbal m.dicine for the rem.dies of snake bites, general weakness, poisons, condylom. accum.nata, lym.hadenopathy, and certain tum.rs in China. In Western m.dicine, Podophyllum.was first used m.dically as a laxative in the early 19th century. Since 1940, the resin of podophyllum.has also been used topically for various skin lesions, such as warts and condylom.. Hum.n poisonings have been reported.An anim.l m.del was established to investigate the neurotoxic effects of Baj.aolian. Podophyllotoxin, the m.j.r active ingredient in Podophyllum. was inj.cted (ip) to young adult m.le rats at doses of 0, 5, 10, or 15 m.-kg-1 b.w.. The anim.ls were sacrificed 72 h after inj.ction.Neuronal changes were readily observable in anim.ls treated with 10 or 15 m.-kg-1 of the toxin. Edem.tous changes of the anterior horn m.toneurons were observed in the spinal cord. No neuronal necrosis was found. The type of neuronal swelling is believed to be only a transient change and would probably subside with tim. if no further assaults occur. m.re serious and perhaps longer term.of changes were found in the dorsal ganglion neurons and the nerve fibers (axons) in the central and peripheral nervous system. Severe depletion of the Nissl substance (RNA/polyribosom.s) was observed in the dorsal root ganglion neurons. Alterations in these sensory neurons would give rise to and correlate with the sensory disturbances experienced by the patients. Bodian staining also revealed a dose-related increase in the coarseness (thickness) of the nerve fibers (axons) in the cerebellum. cerebral cortex, brainstem. and spinal cord.This is the first scientific study showing the neurotoxicity of Baj.aolian, a com.only used Chinese herbal m.dicine.
