Development and mapping of SSR markers linked to resistance-gene homologue clusters in common bean

Common bean is an important but often a disease-susceptible legume crop of temperate,subtropical and tropical regions worldwide. The crop is affected by bacterial, fungal and viral pathogens. The strategy of resistance-gene homologue(RGH) cloning has proven to be an efficient tool for identifying markers and R(resistance) genes associated with resistances to diseases. Microsatellite or SSR markers can be identified by physical association with RGH clones on large-insert DNA clones such as bacterial artificial chromosomes(BACs). Our objectives in this work were to identify RGH-SSR in a BAC library from the Andean genotype G19833 and to test and map any polymorphic markers to identify associations with known positions of disease resistance genes. We developed a set of specific probes designed for clades of common bean RGH genes and then identified positive BAC clones and developed microsatellites from BACs having SSR loci in their end sequences. A total of 629 new RGH-SSRs were identified and named BMr(bean microsatellite RGH-associated markers). A subset of these markers was screened for detecting polymorphism in the genetic mapping population DOR364 × G19833. A genetic map was constructed with a total of 264 markers,among which were 80 RGH loci anchored to single-copy RFLP and SSR markers. Clusters of RGH-SSRs were observed on most of the linkage groups of common bean and in positions associated with R-genes and QTL. The use of these new markers to select for disease resistance is discussed.

Using presence signs to discriminate between similar species

The Lusitanian and the Mediterranean pine voles(Microtus lusitanicus Gerbe,1879 and Microtus duodecimcostatus de Selys-Longchamps,1839)are fossorial sister species and have an allopatric pattern of distribution in Portugal,which includes a potential sympatry area in the centre of the *** present study aimed to determine the validity of using presence signs in the field for discrimination of the two species in an area of sympatry(Northern Alentejo)and the characteristics that achieve the best classification accuracy.A total of 175 trapping plots were sampled across the study *** to the set up of traps,ten presence signs were randomly selected for measure-ments of four variables:proportion of soil mounds,mean diameter of mounds,proportion of burrow openings and mean diameter of burrow *** the basis of a classification tree analysis,results showed that presence signs can be used to discriminate plots inhabited by one or the other species in the studied sympatry *** character-istic that most accurately enables species identification is the proportion of burrow openings:for every ten pres-ence signs found in a plot,if more than eight have an opening,then it is inhabited by ***(*** burrow openings with few or no mounds present);if eight or fewer have an opening,*** is present(*** mounds with few or no burrow openings).

Dear Editor,Monkeypox is a zoonotic disease caused by the monkeypox virus(MPXV),a double-stranded DNA virus in the genus Orthopoxvirus,family *** is known to be transmitted between animals such as rats and squirrels,but animal-to-human and human-to-human transmission has been known in Africa since *** is linked to direct contact with body fluids,skin lesions,and patient items as well as respiratory droplets in case of prolonged face-to-face contact[1].Until recently,only a few cases were associated with travel to endemic countries or contact with infected animals imported from endemic countries.

Control of APN/CD13 and NEP/CD10 on sperm motility

Aminopeptidase N （APN/CD13） and neutral endopeptidase （NEP/CD10） are enzymes present in human sperm cells and involved in regulation of sperm motility of noncapacitated spermatozoa. We investigated the involvement of APN/CD 13 and NEP/CD 10 in motility and in kinematic parameters of human capacitated spermatozoa. Sperm cells isolated by a discontinuous Percoll gradient （40%-80%） followed up by swim-up techniques were incubated with the APN/CD 13 -specific inhibitor, leuhistin （100 μmol L^-1）, and the NEP/CD 10-specific inhibitor, thiorphan （1 μmol L^-1）. The complete inhibition of both APN/CD 13 and NEP/CD 10 improved sperm motility. Spermatozoa incubated with the APN/CD13-specific inhibitor lenhistin showed asymmetrical trajectories, whereas sperm trajectories were more regular after treatment with the NEP/CD 10-specific inhibitor thiorphan. In conclusion, APN/CD 13 and NEP/CD 10 modulate the motility of capacitated spermatozoa, although each of the enzymes seems to participate in the control of different aspects of sperm motility. Therefore, both inhibitors may be useful for sperm activation at different functional stages of spermatozoa.

Alluvial sedimentation and tectonostratigraphic evolution in a narrow extensional zigzag basin margin (northern Teruel Basin, Spain)

The northern part of the eastern margin of the extensional Neogene Teruel Basin(central-eastern Spain)consists of a non-linear,zigzag fault zone made of alternating ca.2 km long,NNW-SSE trending segments and shorter NNESSW *** outcrop conditions made possible a comprehensive integrated stratigraphic and structural study,especially focused on coarse clastic sediments deposited along the basin ***-exposed stratal relationships with boundary faults,allowed the analysis of tectonic influence on *** deformation includes growth faulting,rollover anticlines,and monoclines and associated onlap stratal terminations,angular unconformities,and other complex growth strata *** of them is the onlap-over-rollover bed arrangement described here for the first time,which reveals the competition between tectonic subsidence and sedimentary ***,the structural inheritance(dense Mesozoic fracture grid)and the dominant,nearly‘multidirectional’(σ1 vertical,σ2≈σ3),Pliocene extensional regime withσ3 close to E-W,are considered to have controlled the margin structure and ***-stratigraphic evolution includes:(i)reactivation of inherited NNW-SSE faults and development of W-SW-directed small alluvial fans(SAF)while NNE-SSW segments acted as gentle relay ramp zones;(ii)progressive activation of NNE-SSW faults and development of NW-directed very small alluvial fans(VSAF);during stages i and ii sediments were trapped close to the margin,avoiding widespread progradation;(iii)linking of NNW-SSE and NNE-SSW structural segments,overall basin sinking and widespread alluvial progradation;(iv)fault activity attenuation and alluvial *** particular structure and kinematic evolution of this margin controlled alluvial system *** of alluvial fans,directly set up at the border faults,was conditioned by the narrowness of the margin,small catchment areas,and proximity between faults,which prevented the development of l

Vitellogenin content in fat body and ovary homogenates of workers and queens of Melipona quadrifasciata anthidioides during vitellogenesis

Vitellogenin （Vg） is an egg yolk protein that is produced primarily in the fat body of most female insects. In the advanced social structure of eusocial honeybees, the presence of the queen inhibits egg maturation in the workers＇ ovaries. However in the stingless bee Melipona quadrifasciata, the workers always develop ovaries and lay a certain amount of eggs while provisioning the brood cells with larval food during what is known as the worker nurse phase. The present work is a comparative study of the presence of Vg in homogenates of the fat bodies and ovaries of the nurse workers, and the virgin and physogastric queens of M. quadrifasciata. The presence of Vg was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis and irnmunoblotting using Apis mellifera anti-egg antibody. Vg was not detected in the fat bodies or ovaries of the workers, but it was found in the ovaries of virgin and physogastric queens and in the fat body ofphysogastric queens. The results are discussed, taking into account the reproductive state of the individuals and the other possible roles of Vg, such as a storage protein for metoabolism of other organs.
