BACKGROUND An isthmocele is a scar defect in the uterine wall due to a cesarean section. Its prevalence is unknown,but it has been associated with secondary infertility.Here,we present a case where the patient suffered from an isthmocele that was associated with persistent hydrometra,which developed during in vitro fertilization.CASE SUMMARY The patient underwent hysteroscopic surgery,which successfully resolved the isthmocele as well as the hydrometra. Afterward,two high-quality,euploid embryos,determined by morphological assessment and pre-implantation genetic diagnostic testing,were transferred. This resulted in uterine pregnancy,as determined byserum β-human chorionic gonadotropin levels on day 14(180 m U/ml) and ultrasound-confirmed presence of a gestational sac with a positive embryocardia at week 6. The pregnancy reached 36 wk without any complications,and the product was born in good health. We report a successful isthmocele treatment in a patient with secondary infertility,in which the isthmocele was the cause of persistent hydrometra.CONClUSION Hydrometra caused by secondary cesarean is an infertility factor,which can be corrected by hysteroscopy plus ablation of the isthmocele.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

Bioactive thermal spray coatings produced via high-velocity oxygen fuel spray(HVOF)from hydroxyapatite(HAp)and bioactive glasses(BG)have the potential to be employed on temporary implants due to the ability of both HAp and BG to dissolve and promote osseointegration,considering that both phases have different reaction and dissolution rates under in-vitro conditions.In the present work,75%wt.HAp-25%wt.S53P4 bioactive glass powders were HVOF-sprayed to obtain HAp/S53P4 BG composite coatings on a bioresorbable AZ31 alloy.The study is focused on exploring the effect of the stand-off distance and fuel/oxygen ratio variation as HVOF parameters to obtain stable structural coatings and to establish their effect on the phases and microstructure produced in those coatings.Different characterization techniques,such as scanning electron microscopy,X-ray diffraction,and Fourier transform infrared spectroscopy,were employed to characterize relevant structural and microstructural properties of the composite coatings.The results showed that thermal gradients during coating deposition must be managed to avoid delamination due to the high temperature achieved(max 550℃)and the differences in coefficients of thermal expansion.It was also found that both spraying distance and oxygen/fuel ratio allowed to keep the hydroxyapatite as the main phase in the coatings.In addition,in-vitro electrochemical studies were performed on the obtained HAp/S53P4 BG composite coatings and compared against the uncoated AZ31 alloy.The results showed a significant decrease in hydrogen evolution(at least 98%)when the bioactive coating was applied on the Mg alloy during evaluation in simulated body fluid(SBF).

Non-alcoholic fatty liver disease(NAFlD)is the leading cause of chronic liver disease in the western world(1).Currently,the global prevalence of NAFlD is estimated at 30%and is notably higher in South America and the Middle East(1).NAFlD prevalence has been rising during the last decades,and cases of decompensated NAFlD cirrhosis could increase by 168%in 2030(Figure 1A)(2).These trends are probably explained by several factors including the decrease in viral hepatitis incidence and the increasing prevalence of obesity and type 2 diabetes mellitus(T2DM)among the general population(2).

Melanin is the basis of coloration in many animals,and although it is often used in communication,thermoregulation,or camouflage,melanin has many other physiological functions.For example,in polluted habitats,melanin can have a detoxifying function.Melanic coloration would help to sequester in the skin the heavy metal contaminants from inside the body,which will be expelled to the exterior when the skin is sloughed.Moreover,animals should have evolved more melanic colorations in more polluted habitats(“industrial melanism”hypothesis).We examined whether the fossorial amphisbaenian reptile,Trogonophis wiegmanni,is able to eliminate heavy metals,derived from soil pollution by seagull depositions,through sloughing its skin.Our results suggest a covariation between levels of soil pollution by heavy metals and the concentration of heavy metals in the sloughed skins of amphisbaenians.This suggests that amphisbaenians may expel heavy metals from their bodies when they slough the skins.We also tested whether amphisbaenians inhabiting soils with higher levels of heavy metal pollution had darker(melanin-dependent)body colorations.However,contrary to predictions from the“industrial melanization”hypothesis,we found a negative relationship between soil pollution and proportions of melanic coloration.This contradictory result could,however,be explained because heavy metals have endocrine disruption effects that increase physiological stress,and higher stress levels could result in decreased melanogenesis.We suggest that although amphisbaenians might have some detoxifying mechanism linked to melanin in the skin,this process might be negatively affected by stress and result ineffective under conditions of high soil pollution.

Background The Ibizan Hound is a canine breed native to the Mediterranean region,where leishmaniasis is an endemic zoonosis.Several studies indicate a low prevalence of this disease in Ibizan Hound dogs,whereas other canine breeds present a high prevalence.However,the underlying molecular mechanisms still remain unknown.The aim of this work is to analyse the relationship between serum levels of cytokines and the genomic profiles in two canine breeds,Ibizan Hound(resistant canine breed model)and Boxer(susceptible canine breed model).Methods In this study,we analyse the haplotypes of genes encoding cytokines related to immune response of leishmania infantum infection in twenty-four Boxers and twenty-eight Ibizan Hounds apparently healthy using CanineHD DNA Analysis BeadChip including 165,480 mapped positions.The haplo.glm extension of haplo.score was used to perform a General linear Model(GlM)regression to estimate the magnitude of individual haplotype effects within each cytokine.Results Mean levels of interferon gamma(IFN-γ),interleukin 2(Il-2)and Il-18 in Boxer dogs were 0.19±0.05 ng/ml,46.70±4.54 ng/ml,and 36.37±30.59 pg/ml,whereas Ibizan Hound dogs present 0.49±0.05 ng/ml,64.55±4.54 ng/ml,and 492.10±31.18 pg/ml,respectively.The GlM regression shows fifteen haplotypes with statistically significant effect on the cytokine serum levels(Plevant are Il6-CGAAG and IFNG-GCA haplotypes,which increase and decrease the Il-2,Il-8 and IFN-γserum levels,respectively.Conclusions Haplotypes in the IFNG and Il6 genes have been correlated to serum levels of IFN-γ,Il-2 and Il-18,and a moderate effect has been found on Il8 haplotype correlated to Il-8 and Il-18 serum levels.The results indicate that the resistance to l.infantum infection could be a consequence of certain haplotypes with a high frequency in the Ibizan Hound dog breed,while susceptibility to the disease would be related to other specific haplotypes,with high frequency in Boxer.Future studies are needed to elucidate whether these differences and h

From aerial parts of Austroeupatorium inulifolium was isolated the ent-nor-furano triol labdane austroeupatol 1.The compound 1 was treated with IBX showing an unexpected selectivity at the potentially oxidizable sites of the sub-strate yielding the 2-oxoaustroeupatol(2)and 2,19-dioxoaustroeupatol(3).The treatment of 2 with sodium periodate yields a heterocyclic derivative(ε-caprolactone derivate 4)formed by oxidative cleavage and unexpected intramo-lecular attack of the hydroxymethylene(C-19)oxygen to the ketonic carbon(C-2).A plausible mechanistic pathway for the obtention of compound 4 is proposed.

Flowers are composed of organs whose identity is defined by the combinatorial activity of transcription factors(TFs).The interactions between MADS-box TFs and protein complex formation have been schematized in the floral quartet model of flower development.The gynoecium is the flower’s female reproductive part,crucial for fruit and seed production and,hence,for reproductive success.After the establishment of carpel identity,many tissues arise to form a mature gynoecium.TFs have been described as regulators of gynoecium development,and some interactions and complexes have been identified.However,broad knowledge about the interactions among these TFs and their participation during development remains scarce.In this study,we used a systems biology approach to understand the formation of a complex reproductive unit—as the gynoecium—by mapping binary interactions between well-characterized TFs.We analyzed almost 4500 combinations and detected more than 250 protein-protein interactions(PPIs),resulting in a process-specific interaction map.Topological analyses suggest hidden functions and novel roles for many TFs.In addition,we observed a close relationship between TFs involved in auxin and cytokinin-signaling pathways and other TFs.Furthermore,we analyzed the network by combining PPI data,expression,and genetic data,which helped us to dissect it into several dynamic spatio-temporal subnetworks related to gynoecium development processes.Finally,we generated an extended PPI network that predicts new players in gynoecium development.Taken together,all these results serve as a valuable resource for the plant community.

lipid peroxidation and iron accumulation are closely associated with neurodegenerative diseases,such as Alzheimer’s,Parkinson’s,and Huntington’s diseases,or neurodegeneration with brain iron accumulation disorders.Mitochondrial dysfunction,lipofuscin accumulation,autophagy disruption,and ferroptosis have been implicated as the critical pathomechanisms of lipid peroxidation and iron accumulation in these disorders.Currently,the connection between lipid peroxidation and iron accumulation and the initial cause or consequence in neurodegeneration processes is unclear.In this review,we have compiled the known mechanisms by which lipid peroxidation triggers iron accumulation and lipofuscin formation,and the effect of iron overload on lipid peroxidation and cellular function.The vicious cycle established between both pathological alterations may lead to the development of neurodegeneration.Therefore,the investigation of these mechanisms is essential for exploring therapeutic strategies to restrict neurodegeneration.In addition,we discuss the interplay between lipid peroxidation and iron accumulation in neurodegeneration,particularly in PlA2G6-associated neurodegeneration,a rare neurodegenerative disease with autosomal recessive inheritance,which belongs to the group of neurodegeneration with brain iron accumulation disorders.
