SARS-CoV-2 productively infects primary human immune system cells in vitro and in CoviD-19 patients

The severe acute respiratory syndrome coronavirus 2(SARS-CovV-2)infection is associated with a hyperinflammatory state and lymphocytopenia,a hallmark that appears as both signature and prognosis of disease severity *** cytokine storm and a sustained inflammatory state are commonly associated with immune cell depletion,it is still unclear whether direct SARS-Cov-2 infection of immune cells could also play a role in this scenario by harboring viral *** found that monocytes,as well as both B and T lymphocytes,were susceptible to SARS-Cov-2 infection in vitro,accumulating double-stranded RNA consistent with viral RNA replication and ultimately leading to expressive T cell *** addition,flow cytometry and immunofluorescence analysis revealed that SARS-Cov-2 was frequently detected in monocytes and B lymphocytes from coronavirus disease 2019(CoVID-19)*** rates of SARS-Cov-2-infected monocytes in peripheral blood mononuclear cells from CoVID-19 patients increased over time from symptom onset,with SARS-CoV-2-positive monocytes,B cells,and CD4+T lymphocytes also detected in postmortem lung *** results indicated that SARS-CoV-2 infection of blood-circulating leukocytes in covID-19 patients might have important implications for disease pathogenesis and progression,immune dysfunction,and virus spread within the host.

Investigation of Particle Breakdown in the Production of Composite Magnesium Chloride and Zeolite Based Thermochemical Energy Storage Materials

Production of human polyclonal antibodies by transgenic animals

Polyclonal antibodies collected from the blood of animals and humans experimentally immunised or spontaneously immunised respectively can be injected into patients to protect them against pathogens, toxins, tumours etc. This approach is severely limited by the availability of human polyclonal antibodies of interest. Moreover, polyclonal antibodies from animals are recognised as antigens by patients and are thus rapidly rejected and inactivated. To circumvent this problem, animals (essentially rabbits, chicken, pigs and cows) are being genetically engineered. Their immunoglobulin genes are being inactivated and the corresponding human immunoglobulin genes are being transferred to them. These animals will be immunized and it is expected that large amounts of pure human polyclonal antibodies will be extracted from their blood to be administered to patients. The possible acceptability problem of this approach is under a case study of the European Union Pegasus project.

Genome-wide association study(GWAS)was performed for 16 agronomic traits including nitrogen use efficiency(NUE)and yield-related components using a panel of 190 mainly japonica rice varieties and a set of 38390 single nucleotide polymorphism(SNP)*** panel was evaluated under rainfed upland conditions in Madagascar in two consecutive cropping seasons with two contrasted nitrogen input *** another set of five grain traits,we identified previously known genes(GW5,GS3,Awn1 and Glabrous1),thus validating the pertinence and accuracy of our datasets for GWAS.A total of 369 significant associations were detected between SNPs and agronomic traits,gathered into 46 distinct haplotype groups and 28 isolated *** association signals were identified for the complex quantitative trait NUE,however,larger number of quantitative trait loci(QTLs)were detected for its component traits,with 10 and 2 association signals for nitrogen utilization efficiency and nitrogen uptake efficiency,*** detected association signals co-localized with genes involved in nitrogen transport or nitrogen remobilization within 100 *** present study thus confirmed the potential of GWAS to identify candidate genes and new loci associated with agronomic ***,because of the quantitative and complex nature of NUE-related traits,GWAS might have not captured a large number of QTLs with limited effects.

本文对用SPS法通过熔融LiAlSiO4制备a-Si3N4和用简单的溶胶凝胶法制备LiYO2进行了研究。尽管具有高强度、低膨胀系数、耐高温冲击和耐化学腐蚀等优良的热力学性能,但是由于Si-N之间具有强的共价键从而导致低的扩散系数,致密化氮化硅的制备依然是一个不小的挑战。近来人们通过液相烧结增加物质的传输性制备了接近于理论密度的氮化硅陶瓷。然而,高成本的添加剂和高的烧结温度限制了氮化硅基陶瓷的应用。因此,高效低成本地制备烧结助剂和最终产物的后续研究成为了我们的主要课题。本论文中,我们用溶胶凝胶法制备了钇氧化锂(LiYO2),用固相法制备了硅酸锂铝(LiAlSiO4),这两种材料是中温陶瓷烧结重要的添加剂材料。事实上LiYO2是一种具有碳吸附能力和防暴晒涂层应用的环境友好型材料。它是用锂含量高的前躯体制备的,相比于固相合成反应,这种前躯体可以在更加低的温度下燃烧溶胶凝胶法制备的无定形粉末从而得到高纯的LiYO2粉末。950℃保温6小时,Li(CH3COO)·2H2O和Y(NO3)3·6H2O的摩尔比为2以及1000℃保温6小时,Li(CH3COO)·2H2O和Y(NO3)3·6H2O的摩尔比为3时均可以得到溶胶凝胶制备法的最优化条件。LiYO2的反应和制备机理已经被人们所研究。人们发现燃烧干凝胶制备LiYO2的这个放热合成反应只是简单的Li20和Y203之间的反应。而后,用碳酸锂、氧化铝和氧化硅以1：1.38：1.63的摩尔比制备的前躯体在1100℃下煅烧2小时后,将其用氧化铝球在高压下进行行星球磨16h,制备了纳米晶LiAlSiO4。最终得到的粒径平均尺寸为400nm,粉末粒子以合适的分布表现出不同的形状。此外,最高至1500℃中温SPS烧结制备密实化Si3N4/LiAlSiO4研究中,当温度高于1350℃时表现出了良好的性能,得到了高于理论值95%的体积密度,并且只发生了少量的α/β-Si3N4相变。结果也表明,对于大多数样品,线收缩随着温度和添加剂含量的增加而增大,液相再热处理过程中并不是主相而且含量不足抑制了α/β-Si3N4的相变,从而产生不完全致密。因此,由于LiAlSiO4的介电特性在1300℃以上产生放电等离子体,致密化过程也大多在这个条件下完成。加入20%和30%添加剂的样品在退火时间长达30分钟是有大约0.2%的气孔残存在致密材料中。LiAlSiO4质量分数为30%的LiAlSiO4/Si3N4样品经过SPS快速烧结制备(1450℃,保温15min)几乎完全致密(相对密度为98.5%),其维式硬度为13.34±0.50 GPa,断裂韧性为6.34MPam1/2。

Modeling the antigen recognition by B-cell and T-cell receptors through thermostatted kinetic theory methods

The activation and the resulting response of the immune system to antigens comprise different complex processes and cells. This paper aims at modeling the processes of recognition and learning of the immune system by means of the thermostatted kinetic theory methods. Specifically, the thermostatted kinetic framework is firstly generalized for taking into account that in some processes of proliferation of the cells, the rate is also function of the degree of information exchanged amongst cells. In particular, within the new framework, a mathematical model is proposed for miming the recognition process of the immune system through the definition of interactions between the cytoto^ic and humoral components of the adaptive immune system via T- and B-cells. The model validation is obtained by performing a sensitivity analysis on the parameters which depicts the main emerging phenomena and the different phases of the recognition and learning of the immune system.
