Objective: A variety of ion channels have been implicated in breast cancer proliferation and *** K+(Kv) channels not only cause repolarization in excitable cells,but are also involved in multiple cellular functions in non-excitable *** this study we investigated the role of Kv channels in migration of BT474 breast cancer ***: Transwell technique was used to separate migratory cells from non-migratory ones and these two groups of cells were subject to electrophysiological examinations and microfluorimetric measurements for cytosolic *** migration was examined in the absence or presence of Kv channel ***: When compared with non-migratory cells,migratory cells had much higher Kv current densities,but rather unexpectedly,more depolarized membrane potential and reduced *** transcriptasepolymerase chain reaction(RT-PCR) analysis revealed the presence of Kv1.1,Kv1.3,Kv1.5,Kv2.1,Kv3.3,Kv3.4 and Kv4.3 *** migration was markedly inhibited by tetraethylammonium(TEA),a delayed rectifier Kv channel blocker,but not by 4-aminopyridine,an A-type Kv channel ***: Taken together,our results show that increased Kv channel expression played a role in BT474 cell migration,and Kv channels could be considered as biomarkers or potential therapeutic targets for breast cancer *** mechanism(s) by which Kv channels enhanced migration appeared unrelated to membrane hyperpolarization and Cainflux.

目的:研究PTEN抑癌基因在乳腺癌组织中的表达,探讨其与乳腺癌病人的外周血、骨髓及前哨淋巴结微小转移灶之间的关系。方法:选择53例乳腺癌病人的组织标本,用免疫组织化学方法检测原发肿瘤PTEN蛋白的表达;用定量RT鄄PCR法测定原发肿瘤PTENmRNA的表达。以免疫细胞化学法检测外周血和骨髓中的微小转移灶;HE染色和免疫组织化学法检测前哨淋巴结中的微小转移灶。结果:外周血、骨髓及前哨淋巴结中微小转移灶的检出率分别是24.5%,56.6%,26.4%和41.5%。乳腺癌组织中PTEN蛋白表达呈丢失者占35.8%,后者与外周血和骨髓微小转移灶间无显著关系,而与前哨淋巴结中的微小转移密切相关(P≤0.001)。PTENmRNA的表达与外周血、骨髓及前哨淋巴结中的微小转移灶之间均无显著相关性。结论:乳腺癌组织中PTEN蛋白表达的丢失与前哨淋巴结中的微小转移有密切关系,可作为预测其早期转移的重要指标。

Resolution of hemolysis from pump thrombus during left ventricular assist device exchange

A 50-year-old male who underwent a Heart Mate Ⅱ left ventricular assist device placement for ischemic cardiomyopathy presented with discolored urine and hemolysis 3 mo after the operation. His hemolysis was thought to be due to thrombosis within the pump. Imaging studies were not able to visualize a left ventricular thrombus. Medical management with anticoagulation failed and he underwent surgery for a pump exchange. Intraoperatively, a firm thrombus was found within the pump of the Heart Mate Ⅱ, and the color of the urine changed dramatically from cola-colored to yellow which enabled us to confirm the diagnosis.

Theoretical study of defect impact on two-dimensional MoS_2

Our theoretical findings demonstrate for the first time a possibility of band-gap engineering of monolayer MoS2 crystals by oxygen and the presence of vacancies. Oxygen atoms are revealed to substitute sulfur ones, forming stable MoS2-xOx ternary compounds, or adsorb on top of the sulfur atoms. The substituting oxygen provides a decrease of the band gap from 1.86 to 1.64 eV and transforms the material from a direct-gap to an indirect-gap semiconductor. The surface adsorbed oxygen atoms decrease the band gap up to 0.98 eV depending on their location tending to the metallic character of the electron energy bands at a high concentration of the adsorbed atoms. Oxygen plasma processing is proposed as an effective technology for such band-gap modifications.

Practice of breast self-examination among high risk Chinese women in Hong Kong

Background The incidence of breast cancer in Hong Kong is increasing and breast serf-examination (BSE)as a screening tool is becoming mere popular,especially among first-dsgree relatives (FDR) of breast cancer victims. BSE may be mere applicable to Chinese women as their breast size is smaller. This study explores the BSE practice in this group of *** A cross sectional study was conducted by sending questionnaires to 330 FDR of breast cancer patients treated in the Department of Surgeory,Univeisity of Hong Kong. BSE behavior was studied with respect to its frequency,awareness,completeness and confidence of *** 110 subjects returned the questionnaires with a response rate of 33%.The mean age of the respondent was 37 years. Only 57 women (52%)were practicers. The practicers have a stronger BSE awareness (p＜0.01) and a lower mean score on thought barriers (P=0.002) than the non-practicers.40% of the practicers gain their BSE knowledge through clinicians and their mean period of practica was 3.2 yeara. 68% of the practicers performed BSE *** overall confidence rate was 35%, but the rate was 43% among those who performled complete *** factors possibly related to the completeness and rate of confidence of BSF examination were *** only factor that significantly deternined completenass was the time spent for each examination (P=0.002).The complete practicers required a longer time than the incomplete practicers(6.60 and 2.96 min,respectively).Women with a stronger BSE intention (P=0.001)and a lower mean score on thought barrers(P=0.001)performed the examination *** Slightly over half of the FDR practice *** majority perform a complete BSE but they are not confident in finding abnormalities. Women spending more time on BSE are associated with a higher rate of completeness. However,only those with a stronger DSE: intention and lesser thought barriers are more confident in their practice.
