As the novel coronavirus disease 2019(COVID-19)and its variants continue to rage into the second year of a global pandemic,many success stories of applying Chinese herbal medicine(CHM)to treat COVID-19 patients continue to emerge from China and other part of the ***,from a systems medicine perspective,the authors analyze those experiences and categorize them into four major treatment principles:(1)focusing on eliminating toxins in the early stage of the disease,(2)tonifying deficiency of the body throughout the entire disease course,(3)treating the affected lung and intestine simultaneously based on visceral interactions,(4)cooling blood and removing blood stasis at the later stage,as well as interpret the rationale of these *** is helpful not only in reducing the complexity of promoting the CHM applications to enhance anti-COVID-19 efficacy,but also in ramping out the process of integrating traditional Chinese medicine with modern medical practices.

In the mechanisms of acupuncture analgesia, an important aspect often overlooked is the antiinflammatory effect of acupuncture, which is mainly attained through the self-limiting inflammatory response-inflammatory reflex and its regulation of hypothalamic-pituitary-adrenal(HPA) axis and autonomic nerve system(the vagus nerve and the sympathetic postganglionic fiber). The significance of antiinflammatory effect of acupuncture in analgesia is manifested twofold: The first, inflammatory nociceptive pain is a major type of chronic pain. The second, neurogenic inflammation is the mechanism by which certain acupoints or acu-reflex points(ARPs) are formed at the body surface and have short-cut connections to the pathological focus. Selecting ARPs and applying appropriate sensory stimulation or local microtrauma(affected by different needling means or stimulation parameters), combined with lowfrequency electrical stimulation or the vagus stimulation in the concha region, are helpful to strengthen the anti-inflammatory and analgesic effect of acupuncture.

Background:Scalp combing,as an ancient method of health care,has been used for thousands of years in traditional Chinese *** this method is considered to be beneficial for the blood circulation of the head,the underlying mechanisms remain ***:Both human participants and mice were used in this *** participants,the scalp was stimulated by combing continuously for 5 min,and the temperature was measured using infrared thermal imaging before and after *** mice,the temperature was determined before and at 5,15,and 30 min after a 5-min scalp mechanical stimulation(SMS).Additionally,the vasculature of the mice was labeled with retro-orbital fluorescein isothiocyanate-dextran injection,and the capillaries were observed directly under a confocal *** in vivo CLARITY imaging and the spectrofluorometric detection of Evans Blue dye extravasation,the bloodebrain barrier permeability was ***:SMS increased the temperature of the left ear significantly in human(P=.0247)while can slightly increase the temperature of the right ear and the face without significant difference(P>.05).Moreover,SMS can significantly slow the decrease in the temperature of the external auditory canal at 5 min(P=.0153)and in body temperature at 15 min(P=.0295)after SMS whereas no significant difference in body temperature at 30 min(P>.05)compared with control ***,capillaries of the ear with a diameter of less than 8 mm were significantly dilated(P=.0006)following SMS and the number of dextran dots was higher at 15 min(P>.05)and 30 min(F=10.98,P=.037)after SMS intervention compared with control ***:Our study provides strong evidence to support the notion that scalp combing can improve extra-and intracranial blood circulation under healthy conditions.

目的:探讨头皮机械刺激(SMS),作用机理与头针相似,对脑低灌注大鼠脑水肿的脑保护作用。方法:首先,我们通过测量皮肤温度和脑血流量来筛选SMS的强度。接下来,我们观察了头皮机械刺激对2-VO模型大鼠脑血流量、脑水肿和相关病理学改变的脑保护作用。利用免疫组化和免疫印迹法检测神经凋亡相关蛋白(caspase-3、cleaved-caspase-3)和脑水肿相关蛋白(GFAP和AQP4)的表达差异。随后分析神经细胞凋亡与AQP4蛋白水平的相关性。结果:0.5 N为最适合本研究的刺激强度。与对照组相比,0.5 N、1 N、2 N组均可以维持头皮温度,改善脑血流量(均P<0.05)。SMS干预显著增加了2-VO大鼠脑血流量,降低脑含水量(P<0.001和P=0.0449)。与2-VO组相比,SMS组大鼠海马CA1区caspase-3阳性细胞数(P=0.0086)和cleavedcaspase-3阳性细胞数(P=0.036)显著减少,与caspase-3蛋白表达水平(P=0.019)相似。与sham组比较,2-VO组大鼠GFAP和AQP4表达增高(P=0.0369和P=0.0142),SMS组大鼠GFAP和AQP4表达明显降低(P=0.0484和P=0.0229)。AQP4与caspase-3、cleavedcaspase-3的表达呈正相关(R2=0.6071/R2=0.8500)。结论:本研究揭示了SMS改善脑低灌注的最佳强度及其可能的作用机制。SMS可能是一种简便、有前景的预防缺血性卒中和认知障碍的方法。

Wnt/β-catenin signaling is critical for various cellular processes in multiple cell types,including osteoblast(OB)differentiation and *** how Wnt/β-catenin signaling is regulated in OBs remain ***6AP2,an accessory subunit of V-ATPase,plays important roles in multiple cell types/organs and multiple signaling ***,little is known whether and how ATP6AP2 in OBs regulates Wnt/β-catenin signaling and bone *** we provide evidence for ATP6AP2 in the OB-lineage cells to promote OB-mediated bone formation and bone homeostasis selectively in the trabecular bone *** knocking out(CKO)ATP6AP2 in the OB-lineage cells(Atp6ap2^(Ocn-Cre))reduced trabecular,but not cortical,bone formation and bone *** and cellular biochemical studies revealed that LRP6 and N-cadherin were reduced in ATP6AP2-KO BMSCs and OBs,but not *** in vitro and in vivo studies revealed impairedβ-catenin signaling in ATP6AP2-KO BMSCs and OBs,but not osteocytes,under both basal and Wnt stimulated conditions,although LRP5 was decreased in ATP6AP2-KO osteocytes,but not *** cell biological studies uncovered that osteoblastic ATP6AP2 is not required for Wnt3a suppression ofβ-catenin phosphorylation,but necessary for LRP6/β-catenin and N-cadherin/β-catenin protein complex distribution at the cell membrane,thus preventing their *** of activeβ-catenin diminished the OB differentiation deficit in ATP6AP2-KO *** together,these results support the view for ATP6AP2 as a critical regulator of both LRP6 and N-cadherin protein trafficking and stability,and thus regulatingβ-catenin levels,demonstrating an un-recognized function of osteoblastic ATP6AP2 in promoting Wnt/LRP6/β-catenin signaling and trabecular bone formation.
