石斛属DendrobiumSw.中药生物碱类成分结构新颖,药理作用丰富多样,特别是在抗肿瘤方面的表现尤为突出,多种石斛属中药生物碱已被证实对肺癌、胃癌、肝癌、乳腺癌、结肠癌、宫颈癌等有显著的抗肿瘤活性,显示出新药开发潜力。但目前对其抗肿瘤药理作用及机制的系统分析仍然不足,使得在理解和应用石斛属生物碱的过程中存在一定的盲点和局限性。梳理并深入探讨石斛属中药生物碱成分对不同癌症的药理作用,对其常规的抗肿瘤作用机制如诱导肿瘤细胞凋亡、增强机体免疫功能、抑制肿瘤细胞的迁移、细胞周期调控进行解析,并揭示其可能的作用靶点,为其在抗肿瘤研究领域的深入探索和开发利用提供有力的参考和依据。

目的评价湖北金粟兰中倍半萜二聚体化合物(+)-chlorahupetenes B[(+)-CHB]对脂多糖(LPS)诱导的小鼠巨噬细胞(RAW264.7)炎症反应的影响及作用机制。方法RAW264.7细胞分为对照组(给予等体积DMSO)、模型组(给予等体积DMSO)、地塞米松磷酸钠注射液(Dex,阳性对照,1μmol·L^(-1))组和(+)-CHB低、中、高浓度(5、10、20μmol·L^(-1))组。各组分别加入相应药物孵育细胞1 h,除对照组外,其余组加入LPS(1μg·mL^(-1))诱导24 h造成炎症应答模型。细胞增殖检测法用于评估细胞活力;Griess反应检测一氧化氮(NO)浓度;酶联免疫分析检测炎症细胞因子肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β、IL-6的生成;实时荧光定量PCR(qRT-PCR)检测IL-1β、NOD样受体热蛋白结构域相关蛋白3(NLRP3)、环氧合酶-2(COX-2)、诱导型一氧化氮合酶(iNOS)、IL-6和TNF-αmRNA表达;Western blotting检测Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核因子κB(NF-κB)p65、p-NF-κBp65、NLRP3、嘌呤能受体(P2X7)蛋白表达水平。结果与模型组比较,(+)-CHB减少了梭形细胞数量,使大部分细胞恢复正常形态;显著抑制NO、TNF-α、IL-6、IL-1β生成(P<0.01),显著降低COX-2、iNOS、IL-6、TNF-α、NLRP3、IL-1βmRNA水平(P<0.05、0.01);显著降低TLR4、MyD88、NF-κB p65、p-NF-κB p65、NLRP3、P2X7蛋白表达水平(P<0.05、0.01)。结论(+)-CHB通过抑制TLR4/MyD88/NF-κB信号通路和P2X7/NLRP3/IL-1β炎症小体轴激活缓解LPS诱导的巨噬细胞炎症反应。

目的回顾性分析南京鼓楼医院使用沙库巴曲缬沙坦患者住院期间用药情况,为临床合理用药提供参考。方法系统收集该院2019年7月—2021年9月使用沙库巴曲缬沙坦住院患者相关数据,对其用药合理性进行评价。结果共收集2682例次患者资料,有868例次(32.36%)患者发生918次不合理用药现象。具体不合理用药情况为:超适应证用药182次,用法用量不合理389次,药物转换不合理251次,特殊人群用药不合理96次。结论该院使用沙库巴曲缬沙坦存在不合理用药情况,临床药师应在一定程度上参与患者的用药管理,加强用药监护,提高用药合理性。

Background:The thermoregulatory ability of animals is strongly influenced by the temperature of their *** to cold requires a range of physiological and morphological *** this study,we tested the hypothesis that a small passerine,the Red-billed Leiothrix(Leiothrix lutea),can maintain homeothermy in cold conditions by adjusting the physiology and biochemistry of its tissue and organs and return to its former physiological and biochemical state when moved to a warm ***:Phenotypic variation in thermogenic activity of the Red-billed Leiothrixs(Leiothrix lutea)was investigated under warm(35℃),normal(25℃)or cold(15℃)ambient temperature *** consumption was measured using an open-circuit respirometry *** state-4 respiration and cytochrome-c oxidase(COX)activity in liver,kidney heart and pectoral muscle were measured with a Clark ***:Birds acclimated to an ambient temperature of 15℃ for 4 weeks significantly increased their basal metabolic rate(BMR)compared to a control group kept at 25℃.Birds acclimated to 35℃ decreased their BMR,gross energy intake(geI)and digestible energy intake(DEI).Furthermore,birds acclimated to 15℃ increased state-4 respiration in their pectoral muscles and cytochrome-c oxidase(COX)activity in their liver and pectoral muscle,compared to the 25℃ control *** acclimated to 35℃ also displayed lower state-4 respiration and COX activity in the liver,heart and pectoral muscles,compared to those kept at 25℃.There was a positive correlation between BMR and state-4 respiration,and between BMR and COX activity,in all of the above organs except the liver and ***:Our study illustrates that the morphological,physiological,and enzymatic changes are associated with temperature acclimation in the Red-billed Leiothrix,and supports the notion that the primary means by which small birds meet the energetic challenges of cold conditions is through metabolic adj

为实现植被固沙,设计了一种沙漠植树机器人,通过深度相机实现视觉跟随进而判断树坑标记点,将获取的信息反馈后通过计算得到运动路线,植树机器人抵达工作位置后通过控制电机实现挖坑装置的升降和钻头旋转,基于沙子的高流动性特征设计了树坑挖掘装置来减少进入树坑的回流沙,进而保证树坑的完整性,随后通过传送带将植被运输至树坑内,灌溉的同时带动后部的回填装置完成散沙回填,稳定植被。本设计可为新疆干旱沙漠化治理提供参考。

Bioeffects of different functionalized silica nano-particles on HaCaT cell line

The bioeffects of silica nanoparticles (SiNP), phosphorylate-terminated nanoparticles (PO4- NP) and amino-terminated nanoparticles (NH2NP) on HaCaT cell line have been studied in this paper. The effects of the three kinds of functionalized silica nanoparticles on adherence, proliferation and cycle of HaCaT cells have been investigated. And the cel- lular uptake of the three kinds of functionalized silica nanoparticles by HaCaT cells has also been exam- ined. Results indicated that the bioeffects of the three kinds of functionalized nanoparticles on HaCaT cells were concentration-dependent. And the three kinds of functionalized nanoparticles all exhibited well bio- compatibility if the concentration was below 0.2 μg/μL. While the cytotoxicities of the three kinds of function- alized nanoparticles on HaCaT cells would increase with the increasing of nanoparticles concentration, and the following order was observed: NH2NP > SiNP > PO4NP. In addition, the quantity and rapidity of cellular uptake of nanoparticles by HaCaT cells were diverse due to the different functional groups. Under the same conditions, NH2NP was most and fast internalized by HaCaT cells, followed by SiNP, and PO4NP was the least and slowest. These results provided theoretical foundation for the safe applica- tion and further modification of silica nanoparticles, which could broaden the application of silica nano- particles in biomedicine.

Spinal cord injury(SCI)involves diverse injury responses in different cell types in a temporally and spatially specific ***,using single-cell transcriptomic analyses combined with classic anatomical,behavioral,electrophysiological analyses,we report,with single-cell resolution,temporal molecular and cellular changes in crush-injured adult mouse spinal *** revealed pathological changes of 12 different major cell types,three of which infiltrated into the spinal cord at distinct times *** discovered novel microglia and astrocyte subtypes in the uninjured spinal cord,and their dynamic conversions into additional stage-specific subtypes/*** dynamic changes occur at 3-days post-injury and by day-14 the second wave of microglial activation emerged,accompanied with changes in various cell types including neurons,indicative of the second round of *** day-38,major cell types are still substantially deviated from uninjured states,demonstrating prolonged *** study provides a comprehensive mapping of cellular/molecular pathological changes along the temporal axis after SCI,which may facilitate the development of novel therapeutic strategies,including those targeting microglia.

Dear Editor,Coronavirus disease 2019(COVID-19)is a highly infectious respiratory disease that continues to pose a serious global public health *** disease shows a high infection rate,long incubation period,and rapidly emerging variants,which have led to its rapid spread worldwide(Krammer 2020).Many vaccines have been developed for the control of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),the virus responsible for COVID-19,including vaccines based on messenger RNA(mRNA)(Polack et al.2020),viral vectors(Zhu et al.2020),recombinant proteins(yang et al.2020),and inactivated SARS-CoV-2(Zhang et al.2021).
