BACKGROUND Chronic renal failure(CRF) patients are predisposed to arrhythmias, while the detailed mechanisms are unclear. We hypothesized the chronic inflammatory state of CRF patients may lead to cardiac sympathetic remodeling, increasing the incidence of ventricular arrhythmia(VA) and sudden cardiac death. And explored the role of atorvastatin and etanercept in this *** A total of 48 rats were randomly divided into sham operation group(Sham group), CRF group, CRF + atorvastatin group(CRF + statin group), and CRF + etanercept group(CRF + rhTNFR-Fcgroup). Sympathetic nerve remodeling was assessed by immunofluorescence of growth-associated protein 43(GAP-43) and tyrosine hydroxylase positive area fraction. Electrophysiological testing was performed to assess the incidence of VA by assessing the ventricular effective refractory period and ventricular fibrillation threshold. The levels of tumor necrosis factor-alpha(TNF-α) and interleukin-1beta were determined by Western blotting and enzyme-linked immunosorbent *** Echocardiogram showed that compared with the Sham group, left ventricular end-systolic diameter and ventricular weight/body weight ratio were significantly higher in the CRF group. Hematoxylin-eosin and Masson staining indicated that myocardial fibers were broken, disordered, and fibrotic in the CRF group. Western blotting, enzyme-linked immunosorbent assay,immunofluorescence and electrophysiological examination suggested that compared with the Sham group, GAP-43 and TNF-α proteins were significantly upregulated, GAP-43 and tyrosine hydroxylase positive nerve fiber area was increased, and ventricular fibrillation threshold was significantly decreased in the CRF group. Using the plate/shell elements in commercial software,accurate analysis of interlaminar initial damage in typical composite structures is still a challenging *** propose an accurate and efficient model for analysis of interlaminar initial damage,the following work is carried out:(A)A higher-order theory is firstly proposed by introducing the local Legendre polynomials,and then a novel shell element containing initial damage prediction is developed,which can directly predict transverse shear stresses without any postprocessing *** variables at each node are independent of number of layers,so the proposed model is more efficient than the 3D-FEM.(B)Compression experiment is carried out to verify the capability of the proposed *** results obtained from the proposed model are in good agreement with experimental data.(C)Several examples have been analyzed to further assess the capability of the proposed model by comparing to the 3D-FEM ***,accuracy and efficiency have been evaluated in different damage criterion by comparing with the selected *** numerical results show that the proposed model can well predict the initial interlaminar damage as well as other ***,the model is implemented with UEL subroutine,so that the present approach can be readily utilized to analyze the initial damage in typical composite structures.

Autonomous navigation is a fundamental problem in *** methods generally build point cloud map or dense feature map in perceptual space;due to lack of cognition and memory formation mechanism,traditional methods exist poor robustness and low cognitive *** a new navigation technology that draws inspiration from mammal’s navigation,bionic navigation method can map perceptual information into cognitive space,and have strong autonomy and environment *** improve the robot’s autonomous navigation ability,this paper proposes a cognitive map-based hierarchical navigation ***,the mammals’navigation-related grid cells and head direction cells are modeled to provide the robots with location *** then a global path planning strategy based on cognitive map is proposed,which can anticipate one preferred global path to the target with high efficiency and short ***,a hierarchical motion controlling method is proposed,with which the target navigation can be divided into several sub-target navigation,and the mobile robot can reach to these sub-targets with high confidence ***,some experiments are implemented,the results show that the proposed path planning method can avoid passing through obstacles and obtain one preferred global path to the target with high efficiency,and the time cost does not increase extremely with the increase of experience nodes *** motion controlling results show that the mobile robot can arrive at the target successfully only depending on its self-motion information,which is an effective attempt and reflects strong bionic properties.

Given a k×n integer primitive matrix A(i.e.,a matrix can be extended to an n×n unimodular matrix over the integers)with the maximal absolute value of entries‖A‖bounded by an integer λ from above,the authors study the probability that the m×n matrix extended from A by appending other m-k row vectors of dimension n with entries chosen randomly and independently from the uniform distribution over{0,1,…,λ-1}is still *** authors present a complete and rigorous proof of a lower bound on the probability,which is at least a constant for fixed m in the range[k+1,n-4].As an application,the authors prove that there exists a fast Las Vegas algorithm that completes a k×n primitive matrix A to an n×n unimodular matrix within expected O(n^(ω)log‖A‖)bit operations,where O is big-O but without log factors,ω is the exponent on the arithmetic operations of matrix multiplication.

Regulatory Mechanisms for Transcriptional Bursting Revealed by an Event-Based Model

Gene transcription often occurs in discrete bursts, and it can be difficult to deduce the underlying regulatory mechanisms for transcriptional bursting with limited experimental data. Here, we categorize numerous states of single eukaryotic genes and identify 6 essential transcriptional events, each comprising a series of state transitions;transcriptional bursting is characterized as a sequence of 4 events, capable of being organized in various configurations, in addition to the beginning and ending events. By associating transcriptional kinetics with mean durations and recurrence probabilities of the events, we unravel how transcriptional bursting is modulated by various regulators including transcription factors. Through analytical derivation and numerical simulation, this study reveals key state transitions contributing to transcriptional sensitivity and specificity, typical characteristics of burst profiles, global constraints on intrinsic transcriptional noise, major regulatory modes in individual genes and across the genome, and requirements for fast gene induction upon stimulation. It is illustrated how biochemical reactions on different time scales are modulated to separately shape the durations and ordering of the events. Our results suggest that transcriptional patterns are essentially controlled by a shared set of transcriptional events occurring under specific promoter architectures and regulatory modes, the number of which is actually limited.
