Additive neurorestorative effects of exercise and docosahexaenoic acid intake in a mouse model of Parkinson’s disease

There is a need to develop interventions to slow or reverse the degeneration of dopamine neurons in Parkinson’s disease after *** that preclinical and clinical studies suggest benefits of dietary n-3 polyunsaturated fatty acids,such as docosahexaenoic acid,and exercise in Parkinson’s disease,we investigated whether both could synergistically interact to induce recovery of the dopaminergic ***,mice received a unilateral stereotactic injection of 6-hydroxydopamine into the striatum to establish an animal model of nigrostriatal *** weeks after lesion,animals were fed a docosahexaenoic acid-enriched or a control diet for the next 8 *** this period,the animals had access to a running wheel,which they could use or *** acid treatment,voluntary exercise,or the combination of both had no effect on(i)distance traveled in the open field test,(ii)the percentage of contraversive rotations in the apomorphine-induction test or(iii)the number of tyrosine-hydroxylase-positive cells in the substantia nigra pars ***,the docosahexaenoic acid diet increased the number of tyrosine-hydroxylase-positive terminals and induced a rise in dopamine concentrations in the lesioned *** to docosahexaenoic acid treatment or exercise alone,the combination of docosahexaenoic acid and exercise(i)improved forelimb balance in the stepping test,(ii)decreased the striatal dOPAC/dopamine ratio and(iii)led to increased dopamine transporter levels in the lesioned *** present results suggest that the combination of exercise and docosahexaenoic acid may act synergistically in the striatum of mice with a unilateral lesion of the dopaminergic system and provide support for clinical trials combining nutrition and physical exercise in the treatment of Parkinson’s disease.

Polyethylene glycol fusion repair of severed sciatic nerves accelerates recovery of nociceptive sensory perceptions in male and female rats of different strains

Successful polyethylene glycol fusion(PEG-fusion)of severed axons following peripheral nerve injuries for PEG-fused axons has been reported to:(1)rapidly restore electrophysiological continuity;(2)prevent distal Wallerian degeneration and maintain their myelin sheaths;(3)promote primarily motor,voluntary behavioral recoveries as assessed by the Sciatic Functional Index;and,(4)rapidly produce correct and incorrect connections in many possible combinations that produce rapid and extensive recovery of functional peripheral nervous system/central nervous system connections and reflex(e.g.,toe twitch)or voluntary *** preceding companion paper describes sensory terminal field reo rganization following PEG-fusion repair of sciatic nerve transections or ablations;howeve r,sensory behavioral recovery has not been explicitly explored following PEG-fusion *** the current study,we confirmed the success of PEG-fusion surgeries according to criteria(1-3)above and more extensively investigated whether PEG-fusion enhanced mechanical nociceptive recovery following sciatic transection in male and female outbred Sprague-dawley and inbred Lewis *** nociceptive responses were assessed by measuring withdrawal thresholds using von Frey filaments on the dorsal and midplantar regions of the *** von Frey filament tests were a more reliable method than plantar von Frey filament tests to assess mechanical nociceptive sensitivity following sciatic nerve *** withdrawal thresholds of the sciatic-mediated lateral dorsal region differed significantly across strain but not *** thresholds did not change significantly from baseline in chronic Unoperated and Sham-operated *** sciatic transection,all rats exhibited severe hyposensitivity to stimuli at the lateral dorsal region of the hindpaw ipsilateral to the ***,PEG-fused rats exhibited significantly earlier return to baseline withdrawal thresholds than Negative

Zebrafish embryo toxicity of 15 chlorinated, brominated, and iodinated disinfection by-products

disinfection to protect human health occurs at drinking water and wastewater facilities through application of non-selective oxidants including chlorine. Oxidants also transform organic material and form disinfection by-products（dBPs）, many of which are halogenated and cyto-and genotoxic. Only a handful of assays have been used to compare dBP toxicity,and researchers are unsure which dBP（s） drive the increased cancer risk associated with drinking chlorinated water. The most extensive data set employs an in vitro model cell,Chinese hamster ovary cells. Traditionally, most dBP research focuses on the threat to human health, but the effects on aquatic species exposed to dBPs in wastewater effluents remain ill defined. We present the developmental toxicity for 15 dBPs and a chlorinated wastewater to a model aquatic vertebrate, zebrafish. Mono-halogenated dBPs followed the in vivo toxicity rank order： acetamides 〉 acetic acids 〉 acetonitriles ~ nitrosamines, which agrees well with previously published mammalian in vitro data. di-and tri-halogenated acetonitriles were more toxic than their mono-halogenated analogues, and bromine-and iodine-substituted dBPs tended to be more toxic than chlorinated analogues. No zebrafish development effects were observed after exposure to undiluted or non-concentrated,chlorinated wastewater. We find zebrafish development to be a viable in vivo alternative or confirmatory assay to mammalian in vitro cell assays.

Are the fish of the upper and lower Mekong interconnected?

The Mekong supports one of the richest inland fisheries in the world, with many of the fish migrating long distance to spawn. Little is known about the fisheries and migration strategies of the Upper Mekong whilst it is supposed that many fish species move between the Lower and Upper Mekong. Most likely, natural fish migration in the river has been altered by dam construction across the mainstream of the Upper Mekong. In this paper, the interconnectivity of fish species between different sections of the Mekong and negative impacts of dams on migratory fish are studied. Of the 162 fish species in the Upper Mekong and the 869 species in the Lower Mekong, 61 species are common. Results show that there is no significant difference at order level between the UM and LM. Similarity coefficients are used to evaluate interconnectivity at species, genus and family levels among four different sections of the Upper Mekong with each other and with the Lower Mekong as a whole. The highest similarity is found between the middle and lower reach of the Upper Mekong at species and genus levels and the middle and upper reach at family level. Of the eight cascade dams, Mengsong dam in planning is considered as the biggest threat to migratory fish from the Lower Mekong and should be particularly concerned.

Forest tree species reproduction is a key factor in maintaining the genetic diversity of future generations and the stability of forest *** ongoing ash dieback disease could affect the reproductive ecology of Fraxinus excelsior *** have a major impact on the quantity and quality of pollen and *** this study,we investigated pollen production and viability of pollen and seeds of ash trees with different health status from 2018 to *** were collected from 105 trees(pollen production),pollen from 125 trees(pollen viability),and seeds from 53 trees(seed quality)in two seed orchards and in one floodplain forest in southern *** all parameters were examined at every site every *** average pollen production per tree was estimated at 471.2±647.9 billion pollen *** addition,we found that a high number of inflorescences did not equate to high pollen production per *** production of healthy and diseased trees did not differ significantly,although only 47%of severely diseased male trees(vs.72%for healthy trees)produced *** regards to pollen viability,the TTC test showed an average viability of 73%±17%.Overall,there was a slight tendency for diseased trees to have less viable ***,a significant difference could only be calculated for trees in the floodplain *** percentage of germinable seeds in 2018 was 38%in the floodplain forest and 57%in one of the seed *** percentage of viable seeds(TTC test)ranged from 17 to 22%in the orchards in ***-viable seeds were usually heavily infested by *** general,seed quality was not significantly different between healthy and diseased *** results indicate that ash dieback affects flower formation and pollen viability but not pollen production or seed ***,the fact that hardly any flowering was observed,especially for trees that were seriously affected,suggests a negative effect of ash dieback on reproductive per

A recent research article published in Science by Ritter et *** that endosomal sorting complexes required for transport(ESCRT)proteins mediate repair of lesions in the cell membrane caused by the pore-forming toxin perforin at sites of cytotoxic T cell engagement.1 Subsequent entry of granzymes and initiation of apoptosis of cancer cells is thus limited,leading to reduced sensitivity against T cell‑mediated cytotoxicity.1 Along these lines,this could constitute a new critical resistance mechanism for T cellbased treatment approaches.
