Ubiquitin-specific protease 21 promotes tumorigenicity and stemness of colorectal cancer by deubiquitinating and stabilizing ZEB1

BACKGROUND Colorectal cancer(CRC)is one very usual tumor together with higher death rate.Ubiquitin-specific protease 21(USP21)has been confirmed to take part into the regulation of CRC progression through serving as a facilitator.Interestingly,the promotive function of USP21 has also discovered in the progression of CRC.ZEB1 has illustrated to be modulated by USP7,USP22 and USP51 in cancers.However,the regulatory functions of USP21 on ZEB1 in CRC progression need more invest-igations.AIM To investigate the relationship between USP21 and ZEB1 in CRC progression.METHODS The mRNA and protein expressions were assessed through RT-qPCR,western blot and IHC assay.The interaction between USP21 and ZEB1 was evaluated through Co-IP and GST pull down assays.The cell proliferation was detected through colony formation assay.The cell migration and invasion abilities were determined through Transwell assay.The stemness was tested through sphere formation assay.The tumor growth was evaluated through in vivo mice assay.RESULTS In this work,USP21 and ZEB1 exhibited higher expression in CRC,and resulted into poor prognosis.Moreover,the interaction between USP21 and ZEB1 was further investigated.It was demonstrated that USP21 contributed to the stability of ZEB1 through modulating ubiquitination level.In addition,USP21 streng-thened cell proliferation,migration and stemness through regulating ZEB1.At last,through in vivo assays,it was illustrated that USP21/ZEB1 axis aggravated tumor growth.CONCLUSION For the first time,these above findings manifested that USP21 promoted tumorigenicity and stemness of CRC by deubiquitinating and stabilizing ZEB1.This discovery suggested that USP21/ZEB1 axis may provide novel sights for the treatment of CRC.

Structural engineering of BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures towards ultrastable and tunable photoluminescence

Construction of lead halide perovskite nanocrystals(LHP NCs)heterostructures is essential to obtain highly stable photoluminescence and expand their applications.Herein,a novel self-assembly strategy combining with a solvent-free thermal-assisted synthesis and a water-triggered reaction is developed to subsequently grow BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures at low nucleation temperature with high crystallinity.The as-obtained ternary BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(X=Cl,Br,I)heterostructures exhibit remarkably enhanced panchromatic emission and ultrastable luminescence ascribing to the low-defect growth based on lattice matching.Stable white light-emitting diodes(WLEDs)have been constructed with a high correlated color temperature(CCT)of 7225 K and luminous efficiency of 74.4 lm·W-1.Ln^(3+)-doped BaWO_(4)/CsPbX_(3)/CsPb_(2)X_(5)(Ln^(3+)=Eu^(3+),Tb^(3+),Dy^(3+),Sm^(3+),Yb^(3+)/Er^(3+))nanocomposites are further designed with excitation-dependent photoluminescence and thermochromic properties,making them excellent candidates for high-level anti-counterfeiting and encryption.This work offers a green and universal approach in assembling CsPbX_(3)(X=Cl,Br,I)on lattice-matched tungstate with adjustable panchromatic emission for versatile optical applications.

EGCG and ECG induce apoptosis and decrease autophagy via the AMPK/mTOR and PI3K/AKT/mTOR pathway in human melanoma cells

Catechins have been proven to exert antitumor effects in different kinds of cancers.However,the underlying mechanisms have not been completely clarified yet.This study aimed to assess the effects and mechanisms of(-)-epigallocatechin-3-gallate(EGCG)and(-)-epicatechin-3-gallate(ECG)on human melanoma skin A375 cells.Results showed that EGCG and ECG inhibited the proliferation of A375 cells and ECG showed better inhibitory effect.Flow cytometry analysis had shown that EGCG and ECG induced apoptosis and led to cell cycle arrest.EGCG and ECG decreased Bcl-2 expression and upregulated Caspase-3 protein level,indicating the development of apoptosis.Furthermore,EGCG and ECG could decreased mitochondrial membrane potential of A375 cells.In addition,the expression of Beclin-1,LC3 and Sirt3 were downregulated at protein levels,which known to be associated with autophagy.After autophagy was increased by rapamycin,the apoptotic trend was not change,indicating that apoptosis and autophagy are independent.Mechanistically,EGCG and ECG treatments decreased phosphorylated-AMPK(p-AMPK)and increased the ratios of P-PI3K,p-AKT and p-mTOR in melanoma cells.Conclusively,EGCG and ECG induced apoptosis via mitochondrial signaling pathway,downregulated autophagy through modulating the AMPK/mTOR and PI3K/AKT/mTOR signaling pathway.It indicated that EGCG and ECG may be utilized in human melanoma treatment.

Spray pyrolysis-derived W-doped MoSe2/rGO paper-like microspheres:optimization of microstructure and mesostructure for enhanced lithium storage

Two-dimensional MoSe2is a promising candidate for lithium-ion battery anodes.However,its conductivity and lithium storage volumetric effect still need to be optimized.In this work,W-doped MoSe2/rGO paper-like microspheres are successfully prepared through ultrasonic spray pyrolysis,achieving optimization at both the microstructure and mesostructure to enhance the lithium storage performance of the material.Firstly,by utilizing the similar two-dimensional structure between MoSe2and rGO,self-assembly is achieved through spray pyrolysis,resulting in a well-defined van der Waals heterostructure at the interface on the microscale,enhancing the electron and ion transfer capability of the composite.Secondly,the mesoscale paper-like microsphere morphology provides additional volume expansion buffering space.Moreover,W-doping not only increases the interlayer spacing of MoSe2（0.73 nm）,thereby reducing the diffusion resistance of Li+,but also allow for the modulation of the energy band structure of the material.Density functional theory（DFT） calculations confirm that W-doped MoSe2/rGO exhibits the narrowest bandgap（0.892 eV）.Therefore,the composite demonstrates excellent lithium storage performance,maintaining a specific capacity of 732.9 mAh·g-1after 300 cycles at a current density of 1 A·g-1.

Five Rutaceae family ethanol extracts alleviate H2O2 and LPS-induced inflammation via NF-κB and JAK-STAT3 pathway in HaCaT cells

This study was designed to investigate the effects of five Rutaceae family ethanol extracts(FRFEE):Citrus medica linn(CML),Citrus aurantium L.Cv.Daidai(CAD),Citrus medica linn.***.sarcodactylis(Noot.)Swingle(CMS),Citrus sinensis L.Osbeck(CSO)and Zanthoxylum bungeanum Maxim(ZBM)on retarding the progression of H_(2)O_(2) and LPS-induced HaCaT cells.Cell inflammatory injury model was established by H_(2)O_(2) and LPS.The alleviative effects of FRFEE were evaluated by detecting the activity of superoxide dismutase(SOD),glutathione(GSH)and the generation of reactive oxygen species(ROS).The inflammatory signaling pathways of NF-κB and JAK-STAT3 were detected by Western blotting,the mRNA expression levels of inflammatory factors and skin barrier factors were detected by RT-PCR.50%ethanol extracts of five medicinal and food homologous herbs of Rutaceae family showed different levels of anti-oxidant and anti-inflammatory activities.The FRFEE effectively improved SOD and GSH content and decreased ROS levels.Meanwhile,FRFEE strongly suppressed two inflammatory signaling pathways NF-κB and JAK-STAT3.The RT-PCR examination of inflammatory factors and skin barrier factor revealed significant anti-inflammatory effects of FRFEE.It was worth noting that among the five extracts,Zanthoxylum bungeanum Maxim extract had the best anti-inflammatory and anti-oxidation effects.In addition,it could strongly inhibit the expression of psoriasis factor CCL20.In summary,these results suggested that Zanthoxylum bungeanum Maxim extract could be used as an anti-psoriatic agent in the treatment of psoriasis among FRFEE.

A new interacting capillary bundle model on the multiphase flow in micropores of tight rocks

Surfactants are widely used in the fracturing fluid to enhance the imbibition and thus the oil recovery rate. However, current numerical models cannot capture the physics behind capillary imbibition during the wettability alteration by surfactants. Although the interacting capillary bundle(ICB) model shows potential in characterizing imbibition rates in different pores during wettability alteration, the existing ICB models neglect the influence of wettability and viscosity ratio on the imbibition behavior, making it difficult to accurately describe the oil-water imbibition behavior within the porous media. In this work,a new ICB mathematical model is established by introducing pressure balance without assuming the position of the leading front to comprehensively describe the imbibition behavior in a porous medium under different conditions, including gas-liquid spontaneous imbibition and oil-water imbibition.When the pore size distribution of a tight rock is known, this new model can predict the changes of water saturation during the displacement process in the tight rock, and also determine the imbibition rate in pores of different sizes. The water saturation profiles obtained from the new model are validated against the waterflooding simulation results from the CMG, while the imbibition rates calculated by the model are validated against the experimental observations of gas-liquid spontaneous imbibition. The good match above indicates the newly proposed model can show the water saturation profile at a macroscopic scale while capture the underlying physics of the multiphase flow in a porous medium at a microscopic scale. Simulation results obtained from this model indicate that both wettability and viscosity ratio can affect the sequence of fluid imbibition into pores of different sizes during the multiphase flow, where less-viscous wetting fluid is preferentially imbibed into larger pores while more-viscous wetting fluid tends to be imbibed into smaller pores. Inonotus obliquus polysaccharides prevent UVB-induced skin photodamage by modulating Sirt3-activated Foxo3a/PINK1-Parkin pathway: an in vitro and in vivo study

Inonotus obliquus， an edible and medicinal fungus， has proven to be beneficial for relieving body aging. Previous research has shown that Inonotus obliquus polysaccharide (IOP) can effectively reduce oxidative stress and alleviate photoaging in UVB-treated HaCaT cells. However， the underlying mechanism is still unclear. This study aimed to investigate the mechanism through which IOP mitigates skin photodamage by modulating mitochondrial autophagy through both in vivo and in vitro experiments. The results indicated that IOP alleviated the thickening of the stratum corneum and inflammatory infiltration caused by UVB radiation. Moreover， IOP effectively inhibited the mRNA expression of MMP1 and MMP3 to prevent collagen breakdown and decreased the expression of aging-related proteins (P53 and P21) and inflammatory factors (TNF-α and IFN-γ) in UVB-induced mice. Both in vitro and in vivo studies demonstrated that IOP increased Sirt3 expression， activated the Foxo3a/PINK1-Parkin pathway and enhanced mitochondrial autophagy to reduce photodamage. Additionally， IOP could regulate the gut microbes in photodamaged mice and increase the relative abundance of Clostridium＿XlVa， Desulfovibrio， and Barnesiella， which are important for protecting the intestinal barrier and alleviating intestinal dysfunction. These results provide a theoretical foundation for the use of IOP as a natural component to reduce skin photodamage.

Stark splitting of intense red emission for Er^(3+)in O_(h) symmetry sites realizing optical temperature sensing in biological applications

Optical temperature sensing based on the fluorescence intensity ratio(FIR)of red emission for lanthanide ions holds significant relevance in non-contact temperature measurement for biological application.In this study,the perovskite-structured KZnF_(3)is utilized as a host material for Er^(3+)to achieve a high-purity upconversion(UC)red emission.The observed Stark splitting of the red emission peak provides evidence of the energy level splitting of Er^(3+).Group theory is employed to decompose the spectral branching of Er^(3+)under the point group symmetry of KZnF_(3),allowing for the derivation of Stark splitting energy levels induced by the crystal field effect.The optical temperature-sensing behavior of the red UC luminescence was investigated,specifically examining the FIR of the splitting sub-peaks,which exhibited an exponential relationship with temperature.The KZnF_(3):Yb^(3+),Er^(3+)demonstrated a relative sensitivity(S_(r))of 0.00182%·K^(-1)at 298 K,highlighting its excellent response to temperature.Ex vivo bio-thermometry experiments conducted on chicken breast validated the material's ability to penetrate biological tissues and showed its significant sensitivity of the FIR to temperature.These results establish KZnF_(3):Yb^(3+),Er^(3+)as a promising material for optical thermometry in various biological applications.

BACKGROUND Radiotherapy stands as a promising therapeutic modality for colorectal cancer(CRC);yet,the formidable challenge posed by radio-resistance significantly undermines its efficacy in achieving CRC remission.AIM To elucidate the role played by microRNA-298(miR-298)in CRC radio-resistance.METHODS To establish a radio-resistant CRC cell line,HT-29 cells underwent exposure to 5 gray ionizing radiation that was followed by a 7-d recovery period.The quantification of miR-298 levels within CRC cells was conducted through quantitative RT-PCR,and protein expression determination was realized through Western blotting.Cell viability was assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and proliferation by clonogenic assay.Radio-induced apoptosis was discerned through flow cytometry analysis.RESULTS We observed a marked upregulation of miR-298 in radio-resistant CRC cells.MiR-298 emerged as a key determinant of cell survival following radiation exposure,as its overexpression led to a notable reduction in radiation-induced apoptosis.Intriguingly,miR-298 expression exhibited a strong correlation with CRC cell viability.Further investigation unveiled human dual-specificity tyrosine(Y)-regulated kinase 1A(DYRK1A)as miR-298’s direct target.CONCLUSION Taken together,our findings underline the role played by miR-298 in bolstering radio-resistance in CRC cells by means of DYRK1A downregulation,thereby positioning miR-298 as a promising candidate for mitigating radioresistance in CRC.

Background Macrophages are involved in various immune inflammatory disease conditions.This study aimed to investigate the role and mechanism of macrophages in regulating acute intestinal injury in neonatal necrotizing enterocolitis(NEC).Methods CD68,nucleotide-binding oligomerization domain,leucine-rich repeat,and pyrin domain-containing 3(NLRP3),cysteine aspartate-specific protease-1(caspase-1),and interleukin-1β(IL-1β)in paraffin sections of intestinal tissues from NEC and control patients were detected with immunohistochemistry,immunofluorescence,and western blot.Hypertonic pet milk,hypoxia and cold stimulation were used to establish a mouse(wild type and Nlrp3^(-/-))model of NEC.The mouse macrophage(RAW 264.7)and rat intestinal epithelial cell-6 lines were also cultured followed by various treatments.Macrophages,intestinal epithelial cell injuries,and IL-1β release were determined.Results Compared to the gut“healthy”patients,the intestinal lamina propria of NEC patients had high macrophage infiltration and high NLRP3,caspase-1,and IL-1β levels.Furthermore,in vivo,the survival rate of Nlrp3^(-/-)NEC mice was dramatically improved,the proportion of intestinal macrophages was reduced,and intestinal injury was decreased compared to those of wild-type NEC mice.NLRP3,caspase-1,and IL-1β derived from macrophages or supernatant from cocultures of macrophages and intestinal epithelial cells also caused intestinal epithelial cell injuries.Conclusions Macrophage activation may be essential for NEC development.NLRP3/caspase-1/IL-1β cellular signals derived from macrophages may be the underlying mechanism of NEC development,and all these may be therapeutic targets for developing treatments for NEC.
