Improvements of TKC Technology Accelerate Isolation of Transgene-Free CRISPR/Cas9-Edited Rice Plants

Elimination of the CRISPR/Cas9 constructs in edited plants is a prerequisite for assessing genetic stability, conducting phenotypic characterization, and applying for commercialization of the plants. However, removal of the CRISPR/Cas9 transgenes by genetic segregation and by backcross is laborious and time consuming. We previously reported the development of the transgene killer CRISPR(TKC) technology that uses a pair of suicide genes to trigger self-elimination of the transgenes without compromising gene editing efficiency. The TKC technology enables isolation of transgene-free CRISPR-edited plants within a single generation, greatly accelerating crop improvements. Here, we presented two new TKC vectors that show great efficiency in both editing the target gene and in undergoing self-elimination of the transgenes. The new vectors replaced the CaMV35 S promoter used in our previous TKC vector with two rice promoters to drive one of the suicide genes, providing advantages over our previous TKC vector under certain conditions. The vectors reported here offered more options and flexibility to conduct gene editing experiments in rice.

Dear Editor CRISPR gene-editing technology has successfully generated targeted mutations in rice and many other plant species （Ma et al., 2015）. Assessment of heritability and phenotypic stability of CRISPR-edited plants requires the elimination of the CRISPR construct. The presence of the CRISPR construct makes it difficult to distinguish the mutations transmitted from the previous generation from newly generated mutations by the CRISPR construct at the current generation. The existence of the CRISPR construct also greatly increases the risk of off-target effects.

Given the increased circulation time after fishing,a series of changes take place in live clams,leading to a deterioration in quality even after ***,in this study,we aimed to explore the optimal mode of transportation of *** container for holding clams was reformed,and a water circulation temperature control system was *** physiological responses of clams during anhydrous and watery transportation at two temperatures(4 and 15℃)were investigated based on the aforementioned *** comparing the transportation patterns after 3 d of transport,a higher survival rate was observed at 4℃(97%)than at 15℃(63%)in the anhydrous transportation groups and a lower survival rate was observed at 4℃(93%)than at 15℃(99%)in the watery transportation *** addition,the glycogen content,condition index(CI),and adenylate energy charge(A.E.C)value were higher at4℃((40.87±0.99)mg g^(-1),13.71%±0.50%and 57.45%±1.60%)than at 15℃((30.54±0.81)mg g^(-1),9.09%±0.30%and 43.12%±1.65%)in the anhydrous transportation *** the watery transportation groups,a lower glycogen content,CI,and *** were observed at 4℃((33.78±0.84)mg g^(-1),9.78%±0.50%and 64.65%±1.25%)than at 15℃((41.53±0.93)mg g^(-1),12.72%±0.83%and 71.58%±1.27%).Results from this study show that anhydrous transportation(4℃)is the optimal transport condition for clams to maintain a high quality and good physiological ***,this study will be particularly useful for establishing shellfish transportation systems.

Radiative cooling materials have gained prominence as a zero-energy solution for mitigating global warming. However, a comprehensive understanding of the atomic-scale optical properties and macroscopic optical performance of radiative cooling materials remains elusive, limiting insight into the underlying physics of their optical response and cooling efficacy. La2O3 and HfO2, which represent rare earth and third/fourth subgroup inorganic oxides, respectively, show promise for radiative cooling applications. In this study, we used multiscale simulations to investigate the optical properties of La2O3 and HfO2 across a broad spectrum. First-principles calculations revealed their dielectric functions and intrinsic refractive indices, and the results indicated that the slightly smaller bandgap of La2O3 compared to HfO2 induces a higher refractive index in the solar band. Additionally, three-phonon scattering was found to provide more accurate infrared optical properties than two-phonon scattering, which enhanced the emissivity in the sky window. Monte Carlo simulations were also used to determine the macroscopic optical properties of La2O3 and HfO2 coatings. Based on the simulated results, we identified that the particle size and particle volume fraction play a dominant role in the optical properties. Our findings underscore the potential of La2O3 and HfO2 nanocomposites for environment-friendly cooling and offer a new approach for high-throughput screening of optical materials through multiscale simulations.

Previous studies indicate that individuals’default behaviors on online peer-to-peer(P2P)lending platforms greatly influence other borrowers’default ***,the mechanism of this impact is not ***,there is scarce research in regard to which factors influence the relationship between an individual’s default behavior and an observer’s default *** important questions are yet to be resolved;hence,we conducted two experiments using the scenario-based research method,focusing on Chinese online P2P lending *** results indicate that an individual’s default behavior can trigger an observer’s default intention as a result of the imperfect punitive measures as they currently exist on Chinese online P2P lending *** the observer’s moral disengagement level and pragmatic self-activation level serve as mediating *** situations where an observer knows an individual’s default behavior,the level of intimacy between the defaulter and observer positively affects the relationship between their default behavior and *** intimacy level also positively influences the relationship between the individual’s default behavior and the two mediator *** on the findings,we provide management suggestions in the context of online P2P *** study sets a foundation for future research to utilize other methods to extend the present research findings to other regions and domains.

Pepper(Capsicum spp.)is an important vegetable crop that provides a unique pungent sensation when *** construction of a pepper variome map,we examined the main groups that emerged during domestication and breeding of ***,their relationships and temporal succession,and the molecular events underlying the main *** results showed that the initial differentiation in fruit shape and pungency,increase in fruit weight,and transition from erect to pendent fruits,as well as the recent appearance of large,blocky,sweet fruits(bell peppers),were accompanied by strong selection/fixation of key alleles and introgressions in two large genomic ***,we identified Up,which encodes a BIG GRAIN protein involved in auxin transport,as a key domestication gene that controls erect vs pendent fruit *** up mutation gained increased expression especially in the fruit pedicel through a 579-bp sequence deletion in its 5′upstream region,resulting in the phenotype of pendent *** function of Up was confirmed by virus-induced gene *** together,these findings constitute a cornerstone for understanding the domestication and differentiation of a key horticultural crop.

在成熟市场经济中,作为介于政府与市场之间的第三部门,社会组织具有弥补政府缺陷和市场不足的一般功能。在我国转型时期,社会组织在市场不断发育成熟与政府职能不断转变过程中,扮演一种怎样的角色?文章通过在对我国转型期社会组织发展省域间差异进行描述性分析的基础上,运用全国省域面板数据对社会组织发展差异性的原因和影响因素进行实证研究。结果显示:在转型时期,市场化程度比较高的省份,对公共事务的治理需求也在不断增加,社会组织的发展也会比较快;在现行政府职能转变过程中,政府与社会组织在社会治理中并不是相互替代的关系,而是呈相互补充、相互促进的关系;区域经济发展水平、人口规模以及对外开放程度对社会组织发展的均具有显著的促进作用,但城市化进程的推进对社会组织发展的影响并不显著。
