Expression of auxin synthesis gene tms1 under control of tuber-specific promoter enhances potato tuberization in vitro

Phytohormones, auxins in particular, play an important role in plant development and productivity. Earlier data showed positive impact of exogenous auxin on potato (Solanum tuberosum L.) tuberization. The aim of this study was to generate potato plants with increased auxin level predominantly in tubers. To this end, a pBinB33-tms1 vector was constructed harboring the Agrobacterium auxin biosynthesis gene tms1 fused to tuber-specific promoter of the class i patatin gene (B33-promoter) of potato. Among numerous independently generated B33:tms1 lines, those without visible differences from control were selected for detailed studies. in the majority of transgenic lines, tms1 gene transcription was detected, mostly in tubers rather than in shoots. indoleacetic acid (iAA) content in tubers and the auxin tuber-to-shoot ratio were increased in tms1-expressing transformants. The organ-specific increase in auxin synthesis in B33:tms1-transformants accelerated and intensified the process of tuber formation, reduced the dose of carbohydrate supply required for in vitro tuberization, and decreased the photoperiodic dependence of tuber initiation. Overall, a positive correlation was observed between tms1 expression, iAA content in tubers, and stimulation of tuber formation. The revealed properties of B33:tms1 transformants imply an important role for auxin in potato tuberization and offer prospects to magnify potato productivity by a moderate organ-specific enhancement of auxin content.

New approach for particle size and shape analysis of iron-based oxygen carriers at multiple oxidation states

One of the crucial issues in the chemical looping technology lies in its bed material:the oxygen *** size analysis of an oxygen carrier is important since in a fluidized bed the material can only work well within a specific size *** the favorable size ranges for oxygen carrier materials have already been reported,none of the published studies has analyzed the particle size and shape of oxygen carriers in ***,the effect of oxygen carriers'oxidation degree on such properties has not been considered *** study aimed to report the particle size and shape analysis of five iron-based oxygen carriers,one natural ore,one synthetic material,and three residue products,at different oxidation degrees using dynamic image analysis(DiA).The oxygen carriers were prepared at different mass conversion degrees in a fluidized bed batch *** size distribution,sphericity,and aspect ratio of the oxygen carrier particles were examined experimentally using a Camsizer *** results show that the DiA method was successfully able to analyze the particle size and shape of our oxygen carriers with satisfying accuracy for *** oxidation state of the investigated materials seems to only affect the particle size and shape of oxygen carriers to a minor ***,exposures to redox cycles in a fluidized bed reactor may alter the particle size and shape of most oxygen carriers.

Natural variation of hormone levels in Arabidopsis roots and correlations with complex root architecture

Studies on natural variation are an important tool to unravel the genetic basis of quantitative traits in plants. Despite the significant roles of phytohormones in plant development, including root architecture, hardly any studies have been done to investigate natural variation in endogenous hormone levels in plants. Therefore, in the present study a range of hormones were quantified in root extracts of thirteen Arabidopsis thaliana accessions using a ultra performance liquid chromatography triple quadrupole mass spectrometer. Root system architecture of the set of accessions was quantified, using a new parameter （mature root unit） for complex root systems, and correlated with the phytohormone data. Significant variations in phytohormone levels among the accessions were detected, but were remarkably small, namely less than three-fold difference between extremes. For cytokinins, relatively larger variations were found for ribosides and glucosides, as compared to the free bases. For root phenotyping, length-related traits--lateral root length and total root length--showed larger variations than lateral root number-related ones. For root architecture, antagonistic interactions between hormones, for example, indole-3-acetic acid to trans-zeatin were detected in correlation analysis. These findings provide conclusive evidence for the presence of natural variation in phytohormone levels in Arabidopsis roots, suggesting that quantitative genetic analyses are feasible.

Solid solution samples of the three-component system(1−x)Pb(Ti_(0.5)Zr_(0.5))O_(3)−xCdNb_(2)O_(6)with x=0.0125-0.0500,Δx=0.0125 were obtained by solid phase synthesis followed by sintering using conventional ceramic *** crystal structure,microstructure,electrophysical,and thermophysical properties of these ceramics have been *** is shown that all studied solid solutions can be divided into two groups(with x=0.0125 and with x>0.0125),characterized by different characteristics of the change in properties with variations in external *** is probably due to the transition from a perovskite-type structure with a tetragonal(T)unit cell to inhomogeneous solid solutions consisting of a series of T-phases with similar cell parameters.A conclusion is made about the expediency of using the data obtained in the development of similar materials for devices based on them.

Effects of Agrobacterial<i>rol</i>-Genes on the Thermodynamic and Structural Features of Starches Extracted from Potato Microtubers

Wild-type potato (Solanum tuberosum L.) plants and their transformants harboring agrobacterial rolB or rolC genes under control of the patatin class i promoter were cultured in vitro. These plants were used as a source of single-node stem cuttings. The structure of native starch in tubers formed on cuttings was determined using methods of X-ray scattering and differential scanning microcalorimetry (DSC). it was found that in starch from tubers of rolB plants the melting temperature of crystalline lamella was lower and their thickness was less than that in wild-type potato. in tubers of rolC plants starch differed from starch in wild-type plants by a higher melting temperature, reduced melting enthalpy, and a greater thickness of crystalline lamellae. The melting of starch from tubers of rolC plants proceeded as the melting of two independent crystalline structures with melting temperatures of 338.0°K and 342.8°K. Overall data show that starches of different structure can be obtained by using transgenic approach.

Nucleation of methane hydrate and ice in emulsions of water in crude oils and decane under non-isothermal conditions

Achievable supercooling for the formation of methane hydrate from water emulsions was studied in seven different crude oils and in decane. The experiments were performed under constant rate cooling from + 20 to-15 °C and a pressure of methane of 12 MPa. it was demonstrated that the shapes and positions of the resulting survival curves depend on the density, viscosity and dispersive power of oil samples used in the experiments, as well as on the degree of oil oxidation. in addition, results of the experiments on ice freezing under the same emulsions are presented. The results obtained in the work allowed us to discuss the possibility and features of primary and secondary nucleation of the hydrate and ice in the systems under consideration.

Tourmalinization at the Darasun goldfield,Eastern Transbaikalia:Compositional,fluid inclusion and isotopic constraints

Zoned tourmaline （schorl-dravite） in the matrix of hydrothermal explosive breccia and ore veins in gold deposits, Chita region, Eastern Transbaikalia, Russia, are associated with Na- and K-rich porphyry-type subvolcanic intrusives. 61So values of tourmaline from three gold deposits （Darasun, Talatui, Teremkinskoye） are ＋8.3%, ＋7.6%, and --6.0% and calculated 6180 values of fluids respon- sible for the tourmalinization are ＋7.3%, ＋7.7%, and ＋4.2%, respectively. These data imply an igneous fluid source, except at the Teremkin deposit where mixing with meteoric water is indicated. Wide ranges of Fe3＋/Fetot and the presence of vacancies characterize the Darasun deposit tourmaline indicating wide ranges off（O2） and pH of mineralizing fluids. initial stage tourmalines from the gold deposits of the Darasun ore district are dravite or high mg schorl. Second stage tourmaline is characterized by oscillatory zoning but with Fe generally increasing towards crystal rims indicating decreasing temperature. Third stage tourmaline formed unzoned crystals with XMg （mole fraction of Mg） close to that of the first stage tourmaline, due to a close association with pyrite and arsenopyrite. From Fe^3＋/Fetot values, chemical composition and crystallization temperatures, logf（O2） of mineralizing fluids ranged from ca. -25 to 20, much higher than for the gold-bearing beresite-listvenite association, indicating that tourmalinization was not related to gold mineralization.
