3,4-Epoxybisabola-7（14）,10-dien-2-one, a natural sesquiterpoind, was first synthesized from （-）-carvone through 7 steps, in an overall yield of 35.9%. An efficient synthetic route to muurolane type sesquiterpenes starting from （R）-carvone, employing allylic diazene rearrangement and the ring closing methesis （RCM） reaction as key steps, is described. The first asymmetric total synthesis of （-）-10α- hydroxy-4-muurolen-3-one 11 and （-）-10β-hydroxy-4-muurolen-3-one C was accomplished. Through the total synthesis, the absolute configurations of the natural products A, B and C were established.

Based on a large number of researches and engineering practices both domestic and overseas, it is shown that the building parameters to be determined during scheme phase can exert a great effect on the building energy consumption. In this paper, through a combination of the popular design method of building parameterization at present and the design goat of energy saving during the scheme phase, the author carries out researches on the design methods and toot development which are applicable to parameterization of building energy saving in this stage. In connection with the characteristics of both modeling process of parameterization and energy saving design, and by means of steady calculation as wetl as simulation, this paper establishes an simplified model to calculate the overall energy consumption of air-conditioning, heating, lighting and equipments, and ultimately gives suggestions on design of scheme for energy saving by optimization with the genetic algorithm （GA）. On the basis of the model, a software platform is developed by computer language QTand openGL interface and is oriented to the design users and sets up the MMI （human-computer interaction） software interface for parameterization of building energy saving, which achieves automatic modeling of parameterization and promotes research on oractical design cases.

Objective 5-azacytidine could induce the differentiation of stem cells into cardiomyocytes （CMs）. The aim of this study was to screen the optimal condition for 5-azacytidine inducing differentiation of human mesenchumal stem cells （hMSCs） into CMs, and the effect of 5-azacytidine on adherence, cell vigor and chromosome karyotype of hMSCs. Methods hMSCs were isolated from human bone marrow and cultured in vitro. The phenotypes ofhMSCs were identified by flow cytometric analyses. MTT test was used to investigate the effect of different concentrations of 5-azacytidine on proliferation ofhMSCs. Four weeks after 5-azacytidine induction, semi-quantitative RT-PCR, transmission electron microscopy （TEM）, single-cell action potentials, detection of cardio-enzyme AST and LDH, cell adherence, cell viability and chromosome karyotype test were performed. Results The typical morphological features of hMSCs were fibroblast-like in shape, hMSCs expressed CD44 and CD105,and did not express CD34, CD45 and CD31. The optimal concentration of 5-azacytidine was 10μ mol/L. The shape of hMSCs treated with 5-Azacytidine changed from fusiform to polygon or astrocyte gradually, and passaged cells were evenly arranged as polarity structure. Indueed-hMSCs connected with neighbouring cells, fbrming myotube-like structures 4 weeks later. It was confirmed that induced hMSCs shaped myotubule-like structure and had some of micro-histologic structures of CMs by TEM. RT-PCR showed that induced hMSCs expressed cardiac specific product BNNP and early cardio-myogenesis specific transcription factor NKX2.5mRNA. Besides, induced-MSCs led to the weak action potential and secreted cardio-enzyme AST and LDH. There was no significant difference in cell adherence and viability before and after induction. Both hMSCs and induced-hNSCs kept stable normal diploid nucleus.
