Effects of different vasopressors on hemodynamics in patients undergoing orthotopic liver transplantation

Background The hyperdynamic circulatory state in end-stage liver disease is similar to the hemodynamic state in endotoxic shock. Recent research indicated that proper use of norepinephrine （NE） in patients with endotoxic shock could improve the perfusion of visceral organs and raise the survival rate. In this study, dopamine （DA） or NE combined with DA was infused during the orthotopic liver transplantation （OLT） to observe and compare their effects on hemodynamics, oxygenation, and renal function during different stages of the operation.Methods Thirty American Society of Anesthesiology （ASA） Ⅲ-Ⅳ patients undergoing OLT were randomly divided into group DA and group NE with 15 patients in each group. Vasopressors were infused after induction of anesthesia. DA was infused in group DA; DA and NE in group NE. Data of hemodynamics, oxygenation and renal function were collected after induction, 1 hour in preanhepatic, anhepatic, neohepatic phase and at the end of operation.Results Heart rate （HR） and mean arterial pressure （MAP） of the two groups were stable. In anhepatic phase, central venous pressure （CVP）, mean pulmonary arterial pressure （MPAP）, pulmonary arterial wedge pressure （PAWP）, cardiac output （CO）, and cardiac index （CI） decreased, whereas systemic vascular resistance （SVR） and systemic vascular resistance index （SVRI） increased significantly （P 〈 0.05 ）. The hemodynamic variables of group NE were more stable than that of group DA. Pulmonary vascular resistance （PVR）, pulmonary vascular resistance index （PVRI）, power of hydrogen （pH）, and mixed venous oxygen saturation （ SvO2 ） had no significant changes. Oxygen delivery （ DO2 ） and oxygen consumption （ VO2 ） decreased during anhepatic phase （P〈0. 05 ）, but lactic acid （ LAC ） increased since anhepatic phase. Blood urea nitrogen （BUN） maintained relatively stable during different phases. Group NE had more urine output （F=4. 733, P=0. 039）.Conclusions During OLT, both DA and NE combined with DA can maint

Gene functional research using polyethylenimine-mediated in vivo gene transfection into mouse spermatogenic cells

Aim： To study polyethylenimine （PEI）-mediated in vivo gene transfection into testis cells and preliminary functional research of spermatogenic cell-specific gene NYD-SP12 using this method. Methods： PEI/DNA complexes were introduced into the seminiferous tubules of mouse testes using intratesticular injection. Transfection efficiency and speciality were analyzed on the third day of transfection with fluorescent microscopy and hematoxylin staining. The long-lasting expression of the GFP-NYD-SP12 fusion protein and its subcelluar localization in spermatogenic cells at different stages were analyzed with fluorescent microscopy and propidium iodide staining. Results： With the mediation of PEI, the GFP-NYD-SP12 fusion gene was efficiently transferred and expressed in the germ cells （especially in primary spermatocytes）. Transfection into Sertoli cells was not observed. The subcellular localization of the GFP-NYD-SP2 fusion protein showed dynamic shifts in spermatogenic cells at different stages during spermatogenesis. Conclusion： PEI can efficiently mediate gene transfer into spermatocytes. Thus, it might be useful for the functional research of spermatogenic-cell specific genes such as the NYD-SP12 gene. In our gtudy, the NYD-SP12 protein was visualized and was involved in the formation of acrosome during spermatogenesis. Expression of a novel dipeptidyl peptidase 8(DPP8)transcript variant,DPP8-v3,in human testis

Aim： To investigate the role of a novel dipeptidyl peptidase 8 transcript variant （DPP8-v3） gene in testis development and/or spermatogenesis. Methods： A human testis cDNA microarray was hybridized with mRNA of human adult and fetal testes. Differentially expressed clones were sequenced and characterized and their expression was analyzed by real-time reverse transcription polymerase chain reaction （RT-PCR） and Southern-blot analysis. Results： A new transcript variant of the human dipeptidyl peptidase （DPP8）, exhibiting a 5-fold higher expression level in human adult than that in fetal testes, was cloned and was named DPP8 variant 3 （DPP8-v3）. The full-length sequence of DPP8-v3 was 3,030 bp, encoding a protein of 898 amino acids. Conclusion： DPPS-v3 is a novel human DPP8 transcript variant highly expressed in the adult testis. Similar to DPPIV, DPP8-v3 may play a key role in the immunoregulation of testes and accordingly may influence spermatogenesis and male fertility. （Asian J Androl 2005 Sep; 7： 245-255）

Formation of Mycorrhiza-like Structures in Cultured Root/Callus of Cathaya argyrophylla Chun et Kuang Infected with the Ectomycorrhizal Fungus Cenococcum geophilum Fr.

An in vitro system was used for ectomycorrhizal synthesis of Cenococcum geophilum Fr. with Cathaya argyrophylla Chun et Kuang, an endangered species. Calli initiated from stem segments and adventitious roots differentiated from young seedlings were removed and cocultured with Cenococcum geophllum on a modified Murashlge-Skoog medium. Fungal hyphae were visible within intercellular spaces of the callus 4 weeks after inoculation, but definite and well-developed Hartig net structures did not form in the calu 8 weeks after inoculation. The typical ectomycorrhizal structures （i.e. hyphal mantle and Intracortical Hartig net） were observed in root segments 8 weeks after inoculation. This is the first report of aseptic ectomycorrhlzal-like formation/infection between root organ/callus of Cathaya argyrophylla and the ectomycorrhizal fungus Cenococcum geophflum. This culture system is useful for further investigation of mycorrhizal synthesis in Cathaya trees.

Alcohols＇ Classification by Infrared Spectra Segment Based on Support Vector Machines

This paper studies various classifiers to identify primary, secondary or tertiary alcohols by using segmental spectra and their combinations to support vector machines （SVMs）. The results showed that the O-H in-plane bending absorption contributed most to identification their substitute. This conclusion disagrees with related known research results.

POLY(THIOPHENE-3-ACETIC ACID)-PALLADIUM NANOPARTICLE COMPOSITE MODIFIED ELECTRODES FOR SUPERSENSITIVE DETERminATION OF HYDRAZINE

A promising electrochemical sensor was fabricated by electrodeposition of Pd nanoparticles （PdNPs） on poly（thiophene-3-acetic acid）（PTAA）-modified glassy carbon electrode （GCE）, forming a PdNPs/PTAA composites-modified GCE （PdNPs/PTAA/GCE）. Scanning electron microscope （SEM） and electrochemical techniques were used for the characterization of these composites. It was found that the PdNPs/PTAA layer was very uniform. ]Electrochemical experiments showed that this proposed PdNPs/PTAA composites-modified electrode exhibited excellent electrocatalytic activity towards the oxidation of hydrazine. Under the optimum conditions, the proposed sensor can be applied to the quantification of hydrazine with a wide linear range from 8.0 × 10-9 mol/L to 1.0 × 10-5 mol/L with a low detection limit of 2.67 × 10-9 mol/L. The experiment results also showed that the sensor exhibited good reproducibility and long-term stability, as well as high selectivity with no interference from other potential competing species.

The vitamin C（VC） in crops was successfully determined using ascorbate oxidase（AO） electrochemical biosensor based on the biocompatible poly（3,4-ethylenedioxythiophene）（PEDOT） matrices,which was easily prepared by one-step electrodeposition technique in ionic liquid *** fabricated biosensor displayed excellent bioelectrocatalytic performance to the oxidation of VC,wide linear range,low detection limit,fast response time,good operational and storage stability,the good results of the determination of VC in vegetable crops indicated that the fabricated biosensor will be a good candidate for the physiological and biochemical studies of crops in near future.

Surface iodine and pyrenyl-graphdiyne co-modified Bi catalysts for highly efficient CO_(2) electroreduction in acidic electrolyte

CO_(2) electroreduction to formic acid/formate would contribute to alleviating the energy and climate *** work reports a Bi-based catalyst derived from the in-situ electroreduction of Bi_(2)O_(2)CO_(3) modified with iodine and pyrenyl-graphdiyne(PGDY)on the surface for efficient electroreduction of CO_(2) in acidic electrolyte,with a high partial current density of 98.71 mA·cm^(-2) and high Faradaic efficiency(FE)>90%over the potential range from^(-1).2 to-1.5 V *** hydrogen electrode(RHE),as well as the long-term operational stability over 240 h without degradation in H-type *** results and density function theory calculations show that the synergistic effect of surface iodine and PGDY is responsible for this active and extremely stable process of CO_(2) electroreduction via lowering the energy barriers for formation of*OCHO intermediate,suppressing the competitive HER by enhancing the concentration of both K+and CO_(2) at reaction interface,as well as preventing the dissolution and re-deposition of active Bi atoms on surface during catalytic *** work provides new insight into designing highly active and stable electrocatalysts for CO_(2) reduction.

Renewable biosynthesis of isoprene from wastewater through a synthetic biology approach:the role of individual organic compounds

Electroanalytical method for determination of shikonin based on the enhancement effect of cyclodextrin functionalized carbon nanotubes

A simple and sensitive electroanalytical method for determination of shikonin, a widely used antitumoral agent, using β-cyclodextrin-functionalized multiwalled carbon nanotubes composite modified glassy carbon electrodes （MWCNTs/β-CD/GCE） was presented. CDs are water-soluble and environmentally friendly and can improve the dispersibility of MWCNTs/βCD functional materials, which was confirmed by SEM. The electrochemical behaviors of shikonin on different electrodes were investigated by cyclic voltammetry （CV） and differential pulse voltammograms （DPVs）. The results demonstrated that the redox peak currents of shikonin obtained at MWCNTs/fl-CD/GCE were much higher than those at the βCD/GCE and MWCNTs/GCE, which can be attributed to the combination of the excellent electrocatalytic properties of MWCNTs and the molecular recognition ability of βCD. At MWCNTs/ βCD/GCE, the response current exhibits a linear range from 5.0 nmol/L to 10.0 μmol/L with a detection limit of 1.0nmol/L （S/N=3）. As a practical application, the proposed method was applied to quantitatively determine shikoninin urine samples with satisfying results.
