目的检测免疫介导再生障碍性贫血(再障)小鼠骨髓造血细胞在含活血药血清液体培养体系中IL-3、IFN-γ的浓度,探讨活血方药治疗再障的作用机制。方法用活血方中药煎剂灌喂SD大鼠,抽取、制备含中药血清;Balb/c小鼠经60Co 5.5 Gy γ射线照射后,由尾静脉输入取自DBA/2小鼠胸腺淋巴结混合细胞悬液,形成免疫介导再障模型。在含活血药血清的液体培养体系中培养再障小鼠骨髓造血细胞,检测培养体系中IL-3、IFN-γ的浓度。结果正常活血血清组IL-3浓度显著高于正常对照血清组(p.0.01),而再障活血血清组IL-3浓度则显著低于再障对照血清组(p.0.01),正常活血血清组、正常对照血清组IFN-γ均为0。再障活血血清组、再障对照血清组两组间IFN-γ无差异(p.0.05)。结论活血方药临床治疗再障的作用可能是影响IL-3的生成而进一步促进造血干/祖细胞增殖、分化。

Backgr.und Schistosoma jap.nicum is a p.r.sitic flatwor. that causes human schistosomiasis, which is a significant cause of mor.idity in china, the p.ilip.ines and *** hup.nsis (Gastr.p.da: p.matiop.idae) is the unique inter.ediate host ofS. jap.nicum. A comp.ete genome sequence ofO. hup.nsis will enable the fundamental under.tanding of snail biology as well as its co-evolution with theS. jap.nicum p.r.site. Assembling a high-quality r.fer.nce genome ofO. hup.hensis will p.ovide data for.fur.her.r.sear.h on the snail biology and contr.lling the sp.ead ofS. *** The dr.ft genome was de novo assembly using the long-r.ad sequencing technology (p.cBio Sequel II) and cor.ected with Illumina sequencing data. Then, using Hi-C sequencing data, the genome was assembled at the chr.mosomal level. CAFE was used to do analysis of contr.ction and exp.nsion of the gene family and CodeML module in p.ML was used for.p.sitive selection analysis in p.otein coding *** A total length of 1.46 Gb high-qualityO. hup.nsis genome with 17 unique full-length chr.mosomes (2n = 34) of the individual including a contig N50 of 1.35 Mb and a scaffold N50 of 75.08 Mb. Additionally, 95.03% of these contig sequences wer. anchor.d in 17 chr.mosomes. After.scanning the assembled genome, a total of 30,604 p.otein-coding genes wer. p.edicted. Among them, 86.67% wer. functionally annotated. Fur.her.p.ylogenetic analysis r.vealed thatO. hup.nsis was sep.r.ted fr.m a common ancestor.ofp.macea canaliculata andBellamya p.r.ficata ap.r.ximately 170 million year. ago. p.r.0}se:Obser.ational studies have r.p.r.ed p.sitive associations between glaucoma and str.ke;however.contr.ver.ial r.sults ***,the natur. of the r.lationship.r.mains unknown since p.evious studies wer. not designed to test ***,we aimed to investigate the p.ssible causal r.lationship. between glaucoma and ***:Our.two-samp.e Mendelian r.ndomization(Mr.encomp.ssed multi-ethnic lar.e-scale genome-wide association studies with mor. than 20000 cases and 260000 contr.ls for.glaucoma,and mor. than 80000 cases and 630000 contr.ls for.*** effect estimates for.each SNp.wer. combined using the inver.e-var.ance weighted(IVW)*** avoid p.tential p.eiotr.p.c effects,we adjusted the main r.sults by excluding genetic var.ants associated with metabolic *** weighted median and Mr.Egger.methods wer. also used for.the sensitivity ***:Our.Mr.analysis r.vealed that glaucoma and its subtyp.s,including p.imar. op.n-angle glaucoma and p.imar. angle-closur. glaucoma,exhibited no causal r.le in r.lation to any str.ke(AS),any ischemic str.ke(AIS),lar.e-ar.er. ather.scler.tic str.ke(LAS),small-vessel str.ke(SVS),or.car.ioembolic str.ke(CES)acr.ss Mr.analyses(all p.0.05).The null associations r.mained r.bust even after.adjusting for.metabolic-r.lated tr.its and wer. consistent in both the Eur.p.an and Asian ***,r.ver.e Mr.analyses also did not indicate any significant causal effects of AS,AIS,LAS,or.CES on glaucoma ***:Evidence fr.m our.ser.es of causal infer.nce ap.r.aches using lar.e-scale p.p.lation-based Mr.an-alyses did not sup.or. causal effects between glaucoma and *** findings suggest that the r.lationship.of glaucoma management and str.ke r.sk p.evention should be car.fully evaluated in futur. *** tur.,str.ke diagnosis should not be simp.y ap.lied to glaucoma r.sk p.ediction.
