1,25-dihydroxyvitamin D_3 regulates LPS-induced cytokine production and reduces mortality in rats

AIM： To study the immunoregulatory effect of 1,25-dihydroxyvitamin-D3 Von dominant Thl response in rats. METHODS： Sixty adult Lewis rats were randomized into three groups. Rats in group 1 （n=25） were treated with 1,25-（OH）2D3 first and then challenged with LPS, rats in group 2 （n=25） were treated with vehicle first and then challenged with LPS. Ten animals in groups 1 and 2 were preserved for mortality observation. The remaining animals were injected （i.p） with endotoxin, 24 h after the last administration of 1,25-（OH）2D3 and vehicle. Rats in group 3 （n=10） were treated with 1,25-（OH）2D3 only. Serum IL-12, IFN-y, IL-2 and IL-4 levels were measured and target gene of 1,25-（OH）2D3 on Th cells was studied after 6 h. Gene abundance was verified by real-time quantitative PCR. RESULTS： No death occurred in rats pretreated with 1,25-（OH）2D3 after LPS injection. Death occurred 9 h after LPS injection in rats pretreated with the vehicle, and the number of deaths was 5 within 24 h, with a mortality rate of 50%. There was no change in the number of deaths within 96 h. Six hours after endotoxin stimulation, serum IL-12 and IFN-y levels decreased significantly in rats pretreated with 1,25-（OH）2D3 as compared with those in rats pretreated with the vehicle. The serum content of these two cytokines was very low in rats not challenged by endotoxin, and there was a significant difference as compared with the previous two groups. CONCLUSION： 1,25-（OH）2D3 attenuates injuryinduced by the lethal dose of 1PS, regulates Thl and Th2 cells at the transcription level, and dominantly responds to cytokine production in rats.

Solid lipid nanoparticles loaded with bovine serum albumin（BSA） were prepared by a double emulsion method. As the mass fraction of the model drug BSA increased from 0 to 15%, the particle size gradually increased. The physical stability of the nanoparticles was investigated by zeta potential measurement and they were shown to be quite stable. Fluorescence spectroscopy confirmed that the loaded position of BSA was on the interface between the inner aqueous phase and the solid lipid phase. Both Fourier-transform infrared spectroscopy and circular dichroism spectra indicate that BSA in the nanoparticles was not destroyed, but the secondary structure was disrupted slightly.

The microstructural evolution of 18Crl 8Mn2Mo0.77N high nitrogen austenitic stainless steel in aging treatment was investigated by optical microscopy （OM）, scanning electron microscopy （SEM）, and transmission electron microscopy （TEM）. The results show that hexagonal intergranular and cellular Cr2N with a=0.478 nm and c=0.444 nm and body-centered cubic intermetaUic X phase with a=0.892 nm precipitate gradually in the isothermal aging treatment. The matrix nitrogen depletion due to the intergranular Cr2N precipitation induces the decay of Vickers hardness, and the formation of cellular Cr2N and X phase causes the increase in the values. The impact toughness presents a monotonic decrease and SEM morphologies show the leading brittle intergranular fracture. The tensile strength and elongation deteriorate obviously except for the sample aged for 1 h in yield strength. Stress concentration occurs when the matrix dislocations pile up at the pre- cipitation and matrix interfaces, and the interracial dislocations may become precursors to the misfit dislocations, which can form small cleavage steps and accelerate the formation of cracks.

AIM:To evaluate the inhibitory effect of antisense phosphorothioate oligonucleotide (asON) complementary to the initiator of human telomerase catalytic subunit (hTERT) on the growth of hepatoma ***:The as-hTERT was synthesized by using a DNA synthesizer. HepG2.2.15 cells were treated with ashTERT at the concentration of 10μmol/L. After 72h, these cells were obtained for detecting growth inhibition,telomerase activity using the methods of MTT,TRAP-PCR-EliSA, respectively. BALB/c(nu/nu) mice were injected HepG2.2.15 cells and a human-nude mice model was obtained. There were three groups for anti-tumor activity study. Once tumors were established, these animals in the first group were administered as-hTERT and *** of tumor cells was detected by FCM. In the 2nd group, the animals were injected HepG2.2.15 cells together with as-hTERT. In the third group, the animals were given as-hTERT 24 hours postinjection of HepG2.2.15 cells. The anti-HBV effects were assayed with EliSA in vitro and in ***: Growth inhibition was observed in cells treated with as-hTERT in vitro. A significant different in the value of A570-A630 was found between cells treated with as-hTERT and control (PliSA. The peak of apoptosis in tumor cells given as-hTERT was 21.12%, but not seen in saline-treated control. A prolonged period of carcinogenesis was observed in the second and third group animals. There was inhibitory effect on the expression of HBsAg and HBeAg in vivo and in ***: As-hTERT has an anti-tumor activity, which may be useful for gene therapy of tumors.

We theoretically investigate the spin resolved Friedel oscillation （FO） and quasiparticle interference （QPI） in graphene induced by an Anderson *** the impurity becomes magnetic,the resulted FO becomes spin dependent,which gives rise to a local magnetic moment oscillation with an envelop decaying as r-2in real space in the doping ***,at half filling,the electron density and local magnetic moment will not oscillate but decay as *** spin resolved FO has both sublattice and spin ***,the local magnetic moment decay at half filling only occurs at one sublattice of graphene,which is quite like the phenomenon observed in the STM experiment (***ález-Herrero et al.,Science 352,437 （2016）).We further give an analytic formula about such spin dependent FO based on the stationary phase ***,we study the interference of quasiparticles around the magnetic impurity by calculating the spin dependent Fourier-transformed local density of states （FT-LDOS）.Our work gives a comprehensive understanding about the local magnetic moment oscillation around an Anderson impurity on graphene.

By testing the temperature of the coal and the stress of the working surface, we got the variation law of coal temperature and coal stress during the excavation. The result shows that the activities of mining affect the coal temperature, the fluctuation of coal temperature and the coal stress is synchronous. During the smooth change of crustal stress, the coal temperature basically keeps unchanged, when the dynamic phenomenon appears, the coal temperature changes, as well the coal stress. Therefore, we can use the online coal temperature monitoring system to test the coal temperature of the working surface continuously, and it can provide basic information for forecasting coal mine power disaster before it happens.
