Chondroitinase ABC combined with Schwann cell transplantation enhances restoration of neural connection and functional recovery following acute and chronic spinal cord injury

Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal *** we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord *** report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal *** Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder ***,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord *** findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.

In the intricate process of natural product biosynthesis, a metabolon can enhance metabolic flux by associating sequential enzymes. A fungal metabolon, comprising of flavin-dependent monooxygenase Spe F and P450 monooxygenase Spe G, is identified in the biosynthesis of spiro polycyclic alkaloids(+)-notoamide B and its diastereomer(+)-versicolamide B. Using notoamide E as a substance, Spe F/Spe G metabolon can control the stereoselectivity of its 2,3-epoxidation, followed by hydrogen atom ion at C-17 to generate reactive epoxide tau-m A with dienyl iminium unit. Subsequently,(+)-notoamide B and(+)-versicolamide B are produced via tandem nonenzymatic inverse-electron-demand Diels-Alder reaction and semipinacol rearrangement. This provides the first example of metabolon in the biosynthesis of spiro-prenylated indole alkaloids.

Tropical cyclone extreme rainfall(TCER)causes devastating floods and severe damage in China and it is therefore important to determine its long-term climatological distribution for both disaster prevention and operational *** on the tropical cyclone(TC)best-track dataset and TC precipitation data from 1960 to 2019,the spatiotemporal distribution of TCER affecting China is *** show that there were large regional differences in the threshold for TCER in China,decreasing from the southeastern coast to the northwest *** occurred infrequently in northern China but had a high intensity and was highly *** frequency and intensity of TCER showed slightly increasing trends over time and was most likely to occur in August(41.0%).Most of the TC precipitation processes with extreme rainfall lasted for four to six days,with TCER mainly occurring on the third to fourth *** with wide areas showed a northwestward prevailing track and a westward prevailing *** TCs are not always accompanied by extreme precipitation while some weak TCs can lead to very extreme rainfall.A total of 64.7%(35.3%)of the TCER samples occurred when the TC was centered over the land(sea).TCER≥250 mm was located within 3°of the center of the *** the center of the TC was located over the sea(land),the extreme rainfall over land was most likely to appear on its northwestern(northeastern)side with a dispersed(concentrated)*** has unique climatic characteristics relative to the TC precipitation.

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease.

Several promising plasma biomarker proteins,such as amyloid-β(Aβ),tau,neurofilament light chain,and glial fibrillary acidic protein,are widely used for the diagnosis of neurodegenerative ***,little is known about the long-term stability of these biomarker proteins in plasma samples stored at-80°*** aimed to explore how storage time would affect the diagnostic accuracy of these biomarkers using a large *** samples from 229 cognitively unimpaired individuals,encompassing healthy controls and those experiencing subjective cognitive decline,as well as 99 patients with cognitive impairment,comprising those with mild cognitive impairment and dementia,were acquired from the Sino Longitudinal Study on Cognitive Decline *** samples were stored at-80°C for up to 6 years before being used in this *** results showed that plasma levels of Aβ42,Aβ40,neurofilament light chain,and glial fibrillary acidic protein were not significantly correlated with sample storage ***,the level of total tau showed a negative correlation with sample storage ***,in individuals without cognitive impairment,plasma levels of total protein and tau phosphorylated protein threonine 181(p-tau181)also showed a negative correlation with sample storage *** was not observed in individuals with cognitive ***,we speculate that the diagnostic accuracy of plasma p-tau181 and the p-tau181 to total tau ratio may be influenced by sample storage ***,caution is advised when using these plasma biomarkers for the identification of neurodegenerative diseases,such as Alzheimer's ***,in cohort studies,it is important to consider the impact of storage time on the overall results.

Autumn Tropical Cyclones over the Western North Pacific during 1949–2016:A Statistical Study

We used tropical cyclone(TC)best track data for 1949–2016,provided by the Shanghai Typhoon Institute,China Meteorological Administration(CMA-STI),and a TC size dataset(1980–2016)derived from geostationary satellite infrared images to analyze the statistical characteristics of autumn TCs over the western North Pacific(WNP).We investigated TC genesis frequency,location,track density,intensity,outer size,and landfalling features,as well as their temporal and spatial evolution *** average,the number of autumn TCs accounted for 42.1%of the annual total,slightly less than that of summer TCs(42.7%).However,TCs classified as strong typhoons or super typhoons were more frequent in autumn than in *** most years of the 68-yr study period,there was an inverse relationship between the number of autumn TCs and that of summer *** genesis of autumn TCs was concentrated at three centers over the WNP:the first is located near(14°N,115°E)over the northeastern South China Sea and the other two are located in the vast oceanic area east of the Philippines around(14°N,135°E)and(14°N,145°E),*** terms of intensity,the eight strongest TCs during the study period all occurred in *** is revealed that autumn TCs were featured with strong typhoons and super typhoons,with the latter accounting for28.1%of the total number of autumn ***,the average 34-knot radius(R34)of autumn TCs increased with TC *** 1949 to 2016,164 autumn TCs made landfall in China,with an average annual number *** TCs were most likely to make landfall in Guangdong Province,followed by Hainan Province and Taiwan Island.

OBJECTIVE:To investigate the effect of parthenolide(PTL),a sesquiterpene lactone medicinal compound,on the sensitivity of the gastric cancer cell line SGC7901 and the DPP-and ADR-resistant sublines SGC7901/DDP and SGC7901/ADR to cisplatin[diamminedichloroplatinum(Ⅱ),DDP]and adriamycin(ADR)in ***:SGC7901,SGC7901/DDP,and SGC7901/ADR were treated with various concentrations of PTL alone or in combination with DDP or *** effects on cell proliferation,apoptosis,and expression/activity of several proliferation/apoptosis-related proteins[B-cell lymphoma-2(Bcl-2),cyclin D1,nuclear factor-kappa B(NF-κB),and Caspase-8]and drug transporters(P-glycoprotein and multidrug resistance protein-1)were measured using flow cytometry,Western blotting,and in vitro activity ***:Treatment of SGC7901 cells with PTL inhibited cell growth,increased apoptosis,and sensitized the cells to ***,PTL treatment resulted in downregulation of NF-κB activity and Bcl-2 expression,and upregulation of Caspase-8 ***,PTL co-treatment of SGC7901/DDP and SGC7901/ADR overcame their resistance to DDP and ADR,respectively,with concomitant inhibition of NF-κB,Bcl-2,Cyclin D1,P-glycoprotein,and multidrug resistance protein-1 expression and/or ***:PTL treatment decreases drug resistance in SGC7901,SGC7901/DDP,and SGC7901/ADR cells,as reflected by induction of apoptosis,inhibition of proliferation,downregulation of pro-survival and drug resistance pathways,and upregulation of pro-apoptotic *** results suggest that co-treatment with PTL may thus complement existing therapies for gastric cancer.

Pulp health and calcific healing of a complicated crown–root fracture with additional root fracture in a maxillary incisor: A case report

BACKGROUND Complicated crown–root fracture (CRF) involves severe injury to the crown, root,and pulp, and may be accompanied by multiple root fractures. The loss of a toothhas lifelong consequences for children and teenagers, but the maintenance of pulphealth and the calcific healing of multiple root fractures are rarely reported in *** SUMMARY This case reports healing of a permanent tooth with complicated crown–root andadditional root fractures, in which pulp health was maintained. A 10-year-old girlfell and fractured the root of her maxillary left central incisor at the cervical *** the coronal fragment was repositioned, the tooth was splinted until thetooth was no longer mobile, 2 years later. Eight years after treatment, the toothhas remained asymptomatic with vital pulp and localized gingival ***-beam computed tomography revealed not only calcified healing of the CRFbut also spontaneous healing in an additional undiagnosed root fracture. Thefracture line on the enamel could not be healed by hard tissue and formed agroove in the cervical crown. It was speculated that the groove was related to thelocalized gingival *** This case provides a clinical perspective of the treatment of a tooth with acomplicated CRF and an additional root fracture.
