An efficient and universal protoplast-based transient gene expression system for genome editing in Brassica crops

Protoplast-based transient gene expression system has been widely used in plant genome editing because of its simple operation and less *** order to establish a universal protoplast-based transient transfection system for verifying activities of genome editing vectors containing targets in Brassica,we systematically optimized factors affecting protoplast isolation and transient gene *** established an efficient protoplast-based transient gene expression system(PTGE)in Chinese cabbage,achieving high protoplast yield of 4.9×10^(5)·g^(-1)FW,viability over 95%,and transfection efficiency of 76%.We showed for the first time that pretreatment of protoplasts with a hypotonic MMG could significantly enhance the transfection ***,protoplasts incubated at 37℃ for 6 min improved the transfection efficiency to 86%.We also demonstrated that PTGE worked well(more than 50%transfection efficiency)in multiple Brassica species including cabbage,Pak Choi,Chinese kale,and ***,PTGE was used for validating the activities of CRISPR/Cas9 vectors containing targets in Chinese cabbage,cabbage,and pak choi,demonstrating the broad applicability of the established PTGE for genome editing in Brassica crops.

Amino-functionalized UiO-66-doped mixed matrix membranes with high permeation performance and fouling resistance

For the reduction of bovine serum proteins from wastewater,a novel mixed matrix membrane was prepared by functionalizing the substrate material polyaryletherketone(PAEK),followed by carboxyl groups(C-SPAEKS),and then adding amino-functionalized UiO-66-NH_(2)(Am-UiO-66-NH_(2)).Aminofunctionalization of UiO-66 was accomplished by melamine,followed by an amidation reaction to immobilize Am-UiO-66-NH_(2),which was immobilized on the surface of the membrane as well as in the pore channels,which enhanced the hydrophilicity of the membrane surface while increasing the negative potential of the membrane *** nanoparticle-loaded ultrafiltration membrane has good permeation performance,with a pure water flux of up to 482.3 L·m^(-2)·h^(-1) for C-SPAEKS/AmUiO-66-NH_(2) and a retention rate of up to 98.7%for bovine serum albumin(BSA)-contaminated ***,after several hydrophilic modifications,the flux recovery of BSA contaminants by this series of membranes increased from 56.2%to 80.55%of pure *** results of ultra-filtration flux time tests performed at room temperature showed that the series of ultrafiltration membranes remained relatively stable over a test time of 300 ***,the newly developed mixed matrix membrane showed potential for high efficiency and stability in wastewater treatment containing bovine serum proteins.

Early screening of diabetes retinopathy(DR)plays an important role in preventing irreversible *** research has failed to fully explore effective DR lesion information in fundus ***,traditional attention schemes have not considered the impact of lesion type differences on grading,resulting in unreasonable extraction of important lesion ***,this paper proposes a DR diagnosis scheme that integrates a multi-level patch attention generator(MPAG)and a lesion localization module(LLM).Firstly,MPAGis used to predict patches of different sizes and generate a weighted attention map based on the prediction score and the types of lesions contained in the patches,fully considering the impact of lesion type differences on grading,solving the problem that the attention maps of lesions cannot be further refined and then adapted to the final DR diagnosis ***,the LLM generates a global attention map based on ***,the weighted attention map and global attention map are weighted with the fundus map to fully explore effective DR lesion information and increase the attention of the classification network to lesion *** paper demonstrates the effectiveness of the proposed method through extensive experiments on the public DDR dataset,obtaining an accuracy of 0.8064.

Achieving high fouling resistance and permeability using membrane separation technology in water treatment processes remains a *** this work,a novel mixed-matrix membrane(MMM)(poly(arylene ether ketone)[PAEK]-containing carboxyl groups[PAEK-COOH]/UiO-66-NH_(2)@graphene oxide[GO])with superb fouling resistance and high permeability was prepared by the nonsolvent-induced phase separation method,by in-situ growth of UiO-66-NH_(2) on the GO layer,and by preparing hydrophilic *** the basis of the structure and performance analysis of the MMM,the maximum water flux reached 591.25 L·m^(-2)·h^(-1) for PAEK-COOH/UiO-66-NH_(2)@GO,whereas the retention rate for bovine serum albumin increased from 85.40%to 94.87%.As the loading gradually increased,the hydrophilicity of the MMMs increased,significantly enhancing their fouling *** strongest anti-fouling ability observed was 94.74%,which was 2.02 times greater than that of the pure *** the same time,the MMMs contained internal amide and hydrogen bonds during the preparation process,forming a cross-linked structure,which further enhanced the mechanical strength and chemical *** summary,the MMMs with high retention rate,strong permeability,and anti-fouling ability were successfully prepared.

Molecular dielectric switches constitute a type of intelligent materials that are highly coveted for their distinctive advantages of switchable dielectric responses,lightweight,and mechanical ***(2D)hybrid perovskites have demonstrated excellent promise for assembling dielectric switches,in which the dynamic motions of organic moieties afford driving force to trigger switchable dielectric phase ***,we successfully assembled a new lead-free hybrid double perovskite,(CHA)4Cu Bi Br8(1,CHA=cyclohexylammonium),adopting a typical 2D structural motif,which shows dielectric anisotropy and bistable behaviors during the reversible phase transition near T_(c)=378 K(the Curie temperature).That is,its dielectric constants could be switched and tuned between high-dielectric and low-dielectric *** analyses reveal that the ordered-disordered transformation of the organic CHA+moiety and distortion of inorganic framework account for its phase *** result will stimulate further exploration of molecular dielectric switches in this 2D environmentally friendly family.

针对单个烯烃生产系统节能潜力有限的问题,提出基于烯烃生产子系统规模调配的方法,构建混合整数非线性规划(MINLP)优化模型。以单位产量1 t×h^(-1)为基准规模,当丙烷脱氢和乙烷裂解的规模系数分别为50.0和89.4时(即产量分别为50.0 t×h^(-1)和89.4 t×h^(-1)),系统的能量回收率达到最大,为82.6%。与两者独立子系统相比,公用工程负荷减少502.7 MW,对应的中低压蒸气消耗量减少544.9 t×h^(-1),电能减少0.5 MW,碳排放量减少0.5 t×h^(-1),节能减排的效果非常显著。结果表明,多个系统间能量交互可以有效提高能量回收率,且最优的规模系数使系统整体能量回收率达到最大。

Study on the mechanism of Euphorbia fischeriana Steud.- Jujubae Fructus in the treatment of hepatocirrhosis based on network pharmacology

The active components,targets,and pathways of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis and the mechanism of action were explored by means of network ***,the active components and related targets of Jujubae Fructus were screened by TCMSP database and standardized by Uniprot *** compounds of Euphorbia fischeriana *** obtained by searching the literature and finally screened by PubChem database,Swiss ADME,and *** targets were obtained through Genecards database,PPI network analysis was conducted on common targets of Euphorbia fischeriana Steud.-Jujubae Fructus and hepatocirrhosis by using String database,GO enrichment analysis and KEGG pathway enrichment analysis was conducted through Metascape database by using intersection targets of Euphorbia fischeriana Steud.-Jujubae Fructus and hepatocirrhosis,and the results were drawn by using Weishengxin online drawing ***,the network of drug-compound-target-pathway was constructed by the software of ***,the above results were verified by molecular docking.47 active compounds from Euphorbia fischeriana Steud.-Jujubae Fructus were screened out,which had 38 common targets,162 intersection targets,and 340 signal pathways with hepatocirrhosis,mainly involving hepatitis C,JAK-STAT signal pathway and AGE-RAGE signal ***,such as MAPK1,AKT1,TNF,JUN,IL6 and PTGS2,play important roles in the *** findings suggested that the main active ingredients of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis are quercetin,scopolamine,physcion,7-deoxyrangduin,17-Hydroxyjolkinolide A,*** docking results showed that the main active components and core targets might have a good binding *** study preliminarily explored the potential mechanism of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis and provided a theoretical basis for the clin

工业生产中的自动化智能化离不开自动目标检测,而高准确性的自动目标检测则依赖于与实际场景相适应的数据集。本文针对工业实际场景,发布了一个密集控制阀零件数据集,命名为PD4CV(Part Detection for Control Valve)2023。该数据集的图像全部来源于控制阀生产车间,图像采集完成后,首先对数据集图片进行预处理操作,接着对数据集图片中的零件目标进行标注,然后再对数据集图片进行训练集、验证集以及测试集的划分。PD4CV2023数据集共涵盖9类零件,包括510张工盘图像和15 015个零件样本,平均每张图像含有约29个零件样本。与现有的目标检测数据集相比,该数据集具有零件摆放密集、遮挡,零件尺寸差异大,部分零件外形相似,零件样本数量不均衡等特点。最后,在不同类型数据集上的预训练对比实验表明,一般场景数据集、特定工业场景数据集只适用于一般和特定任务,而代表实际控制阀生产工况下的PD4CV2023数据集,可用于控制阀零件目标检测,其具有其特殊性和不可替代性;一系列目标检测算法在该数据集上的综合对比则验证了PD4CV2023数据集在一般性目标检测、多尺度目标检测、小规模、不均衡数据下目标检测中的有效性。PD4CV2023数据集可用于面向工业的目标检测的相关研究。
