A backcrossed population （BC1） derived from a cross between C100 and Dazao was obtained. The quantitative trait loci （QTLs） of the economically important traits for whole cocoon weight, cocoon shell weight, ratio of cocoon shell and weight of pupae, etc., were analyzed for the first time using the multiple interval mapping software WinQTLCart2.0. In total 40 QTLs were detected and contributed to 21 groups based on the constructed linkage map. According to the mapping results, 2, 2, 3, and 2 major QTLs explained over 20% of total phenotypic variations, whereas four QTLs, namely qCW-19, qSW-2, qCSR-4, and qPW-23, explained more than 30% of total phenotypic variations for whole cocoon weight, cocoon shell weight, ratio of cocoon shell and weight of pupae, respectively. Correlated traits QTLs often share the same location. Furthermore, most of the detected QTLs were closed to one-side marker. By using the very closed markers, positive QTLs can be aggregated, which can form a basis for molecular marker-assisted selection and breeding.

The Rana temporaria chensinensis is an amphibian distributing mainly over the north of China. Its skin gland contains a variety of peptides, including antibacterial peptides. One of them, component 1, has been purified from the dried skin of *** chensinesis using a five step procedure. Purified component 1 has a molecular weight of 5 700 and its amino acid composition shows that it is devoid of disulfide bridges but rich in histine and glycine residues. Moreover, it has been found that it has a broad spectrum of antibacterial activity for Pseuctomonas aeruginosa, Escherichia coli, Bacillus sublilis, S. faccalis and Proteus vulgaris .

利用家蚕品系C100和大造的回交一代BC1群体,用WinQTLCart2.0软件对家蚕全茧量、茧层量、茧层率和蛹体重等4个重要经济性状在分子连锁图的基础上进行了多重区间作图分析。共检测到40个QTL,分布于21个连锁群,其中全茧量、茧层量、茧层率和蛹体重的QTL中,贡献率超过20%的主效QTL数分别为2、2、3、2个,贡献率超过30%的主效QTL有4个,分别是控制全茧量的qCW-19、茧层量的qSW-2、茧层率的qCSR-4和蛹体重的qPW-23。相关显著的经济性状的QTL具有集中分布的特点,并且检测到的QTL均与其单侧标记紧密连锁,利用紧密连锁的分子标记将不同的增效QTL进行聚合,为优质高产家蚕新品种的选育打下基础。

MicroRNAs(miRNAs)are involved in lymphoma progression by regulating the tumor *** miR130b is overexpressed in diffuse large B-cell lymphoma(DLBCL),inducing Th17 cell *** further illustrate its biological significance and therapeutic rationale,miR130b was detected by quantitative real-time PCR in the serum samples of 532 newly diagnosed DLBCL *** mechanism of miR130b on lymphoma progression and the tumor microenvironment was investigated both in vitro and in *** targeting miR130b was also evaluated,including OX40 agonistic antibody and lipid nanoparticles(LNPs)-miR130b *** results showed that serum miR130b significantly correlated with tumor miR130b and serum interleukin-17,indicating lymphoma relapse and inferior survival of DLBCL ***130b overexpression altered tumor microenvironment signaling pathways and increased Th17 cell *** mechanism of action,miR130b downregulated tumor OX40L expression by directly targeting IFNAR1/p-STAT1 axis,recruiting Th17 cells via OX40/OX40L interaction,thereby promoting immunosuppressive function of Th17 *** co-culture systems of B-lymphoma cells with immune cells,miR130b inhibited lymphoma cell autophagy,which could be counteracted by OX40 agonistic antibody and LNPs-miR130b *** murine xenograft model established with subcutaneous injection of A20 cells,both OX40 agonistic antibody and LNPs-miR130b antagomir remarkably inhibited Th17 cells and retarded miR130b-overexpressing tumor *** conclusion,as an oncogenic biomarker of DLBCL,miR130b was related to lymphoma progression through modulating OX40/OX40L-mediated lymphoma cell interaction with Th17 cells,attributing to B-cell lymphoma sensitivity towards OX40 agonistic *** miR130b using LNPs-miR130b antagomir could also be a potential immunotherapeutic strategy in treating OX40-altered lymphoid malignancies.

国际蚕分子育种计划(International Silkworm Project)是Rhode Island大学的***在1991年提出来的,也叫基因标记育种计划[1].该计划主要包括两步工作:第一步是制作蚕的分子基因图, 第二步是数量性状定位分析(简称QTL分析,也叫数量性状定位作图:QTL Mapping).最后利用分子标记直接在DNA水平进行重要经济性状(数量性状)的选择、固定,即实施'分子育种',用很短时间育成兼具高抗、强健、丝多、质优、易繁等特性的新蚕品种,本文对此进行简要介绍.

The Omicron variant of the severe acute respiratory syndrome coronavirus 2(SARS‑CoV‑2)infected a substantial proportion of Chinese population,and understanding the factors underlying the severity of the disease and fatality is valuable for future prevention and clinical *** recruited 64 patients with invasive ventilation for COVID-19 and performed metatranscriptomic sequencing to profile host transcriptomic profiles,plus viral,bacterial,and fungal content,as well as virulence factors and examined their relationships to 28-day mortality were *** addition,the bronchoalveolar lavage fluid(BALF)samples from invasive ventilated hospital/community-acquired pneumonia patients(HAP/CAP)sampled in 2019 were included for *** analysis revealed that all Omicron strains belong to BA.5 and BF.7 sub-lineages,with no difference in 28-day mortality between *** to HAP/CAP cohort,invasive ventilated COVID-19 patients have distinct host transcriptomic and microbial signatures in the lower respiratory tract;and in the COVID-19 non-survivors,we found significantly lower gene expressions in pathways related viral processes and positive regulation of protein localization to plasma membrane,higher abundance of opportunistic pathogens including bacterial Alloprevotella,Caulobacter,Escherichia-Shigella,Ralstonia and fungal Aspergillus sydowii and Penicillium *** analysis further revealed significant associations between host immune responses and microbial compositions,besides synergy within viral,bacterial,and fungal *** study presents the relationships of lower respiratory tract microbiome and transcriptome in invasive ventilated COVID-19 patients,providing the basis for future clinical treatment and reduction of fatality.

家蚕茧质性状的QTL定位研究

采用QTLMapper2.0QTL作图软件,对F2群体的家蚕全茧量、茧层量、茧层率和蛹体重等性状进行了QTL定位分析,分别检测出7个、6个、2个、8个有显著效应分量的QTLs,分布于7个、5个、2个、7个不同的连锁群。控制全茧量、茧层量的QTLs一般存在复杂的上位性效应。对全茧量性状,有3对QTLs存在显著的加加上位性效应,其中1对还存在加显、显显互作;共有3个QTLs存在显著的显性效应,1个存在显著的加性效应。对茧层量QTLs,发现1 对QTLs存在极显著的各项遗传效应,包括上位性效应;1对QTLs被检测到显著的显显互作,1个QTL具有显著的显性效应,并与另一个QTL存在显著的加加互作。茧层率、蛹体重主要受加性或显性的QTLs作用,没有发现茧层率QTLs的上位性效应,蛹体重的有效QTL大都呈现显著的负向显性效应,只有一对QTLs存在显著的加加上位性效应。第2、3、4、11、13、24、34、37、40连锁群是两个或多个性状QTLs分布的共同连锁群。全茧量和茧层量存在共同的QTL 或染色体区域,育种上可通过适当选配,利用基因的互作效应,同步改良这两个性状。

采用混合线性模型,对家蚕全茧量、茧层量、茧层率和蛹体重等性状的性别效应进行理论估算:全茧量、茧层量、茧层率和蛹体重等4性状的性别随机效应的方差的概率和性别随机效应的预测值概率都达到极显著水平,证明全茧量、茧层量、茧层率和蛹体重等4性状的性别效应极显著,这完全符合实际情况。全茧量、茧层量、茧层率和蛹体重等4性状的性别效应预测值雌(雄)分别为0.248g(-0.247g)、2.423cg(-2.394)cg、-1.976%(1.992%)和0.224g(-0.223g)。性别效应调整后各性状均呈单峰正态分布,符合QTL分析对数量性状连续正态分布的要求。

Widespread soil acidification due to atmospheric acid deposition and agricultural fertilization may greatly accelerate soil carbonate dissolution and ***,to date,few studies have addressed these ***,we use meta-analysis and nationwide-survey datasets to investigate changes in soil inorganic carbon(SIC) stocks in *** observe an overall decrease in SIC stocks in topsoil(0-30 cm)(11.33 g C m-2yr-1) from the 1980 s to the 2010 *** SIC stocks have decreased by～8.99±2.24%(1.37±0.37 Pg C).The average SIC losses across China(0.046 Pg C yr-1) and in cropland(0.016 Pg C yr-1) account for～17.6%-24.0% of the terrestrial C sink and 57.1% of the soil organic carbon sink in cropland,*** deposition and climate change have profound influences on SIC *** estimate that～19.12%-19.47% of SIC stocks will be further lost by *** consumption of SIC may offset a large portion of global efforts aimed at ecosystem carbon sequestration,which emphasizes the importance of achieving a better understanding of the indirect coupling mechanisms of nitrogen and carbon cycling and of effective countermeasures to minimize SIC loss.
