The ePSPS Pro106Ser substitution solely accounts for glyphosate resistance in a goosegrass(eleusine indica) population from Tennessee, United States

Previous studies have documented the occurrence of glyphosate-resistant （GR） goosegrass （eleusine indica （L.） Gaertn.） and, in at least some cases, resistance is due to an altered target site. Research was performed to determine if an altered target site was responsible for GR in a Tennessee, United States goosegrass population （TennGR）. DNA sequencing revealed a mutation in TennGR plants conferring the Prol06Ser 5-enolpyruvylshikimate-3-phosphate synthase （ePSPS） substitution previously identified in other GR populations. F2 populations were derived from TennGR plants crossed with plants from a glyphosate-susceptible population （TennGS） and analyzed for their response to glyphosate and genotyped at the ePSPS locus. Plants from the F2 populations segregated 1：2：1 sensitive：intermediate：resistant in response to a selec- tive dose of glyphosate, and these responses co-segregated with the ePSPS genotypes （PP106, PS106, and SS106）. To separately investigate the effect of the Prol06Ser substitution on GR, glyphosate dose-response curves and 50% effective dose （eDso） values were compared among the three genotypes and the two parental populations. The SS106 genotype was 3.4-fold resistant relative to the PP106 genotype, identical to the resistance level obtained when comparing the resistant and susceptible parental populations. We conclude that the mutation conferring a Prol06Ser ePSPS mutation is solely responsible for GR in the TennGR goosegrass population.

Frontiers of 3D Printing/Additive Manufacturing: from Human Organs to Aircraft Fabrication

It has been more than three decades since stereolithography began to emerge in various forms of additive manufacturing and 3D printing. Today these technologies are proliferating worldwide in various forms of advanced manufacturing. The largest segment of the 3D printing market today involves various polymer component fabrications, particularly complex structures not attainable by other manufacturing *** printer head systems have also been adapted to selectively print various speciated human cells and special molecules in attempts to construct human organs, beginning with skin and various tissue patches. These efforts are discussed along with metal and alloy fabrication of a variety of implant and bone replacement components by creating powder layers, which are selectively melted into complex forms（such as foams and other open-cellular structures） using laser and electron beams directed by CAD software. efforts to create a ＂living implant＂ by bone ingrowth and eventual vascularization within these implants will be discussed briefly. Novel printer heads for direct metal droplet deposition as in other 3D printing systems are briefly described since these concepts will allow for the eventual fabrication of very large and complex products, including automotive and aerospace structures and components.

Strategies for creating living, additively manufactured, open-cellular metal and alloy implants by promoting osseointegration,osteoinduction and vascularization: An overview

Additive manufacturing of porous, open-cellular metal or alloy implants, fabricated by laser or electron beam melting of a powder bed, is briefly reviewed in relation to optimizing biomechanical compatibility by assuring elastic(Young's) modulus matching of proximate bone, along with corresponding pore sizes assuring osseointegration and vasculature development and migration. In addition, associated, requisite compressive and fatigue strengths for such implants are described. Strategies for optimizing osteoblast(bone cell) development and osteoinduction as well as vascularization of tissue in 3 D scaffolds and tissue engineering constructs for bone repair are reviewed in relation to the biology of osteogenesis and neovascularization in bone, and the role of associated growth factors, bone morphogenic proteins, signaling molecules and the like. Prospects for infusing hydrogel/collagen matrices containing these cellular and protein components or surgically extracted intramedullary(bone marrow) concentrate/aspirate containing these biological and cell components into porous implants are discussed, as strategies for creating living implants, which over the long term would act as metal or alloy scaffolds.

Vascularization in interconnected 3D printed Ti-6Al-4V foams with hydrogel matrix for biomedical bone replacement implants

Vascularization on newly implanted metal orthopedic implants has remained a challenge in the field of tissue engineering. To address this challenge, in this research,an interconnected foam structure of Ti-6 Al-4 V was microfabricated by electron beam melting(eBM) technique. The foam in question has a density of 1.77 g cm-3 with 60% porosity and a tensile strength of 18 GPa. An extracellular matrix based hydrogel was added as an aqueous matrix to the *** mimetic stress has been closely related to many wound healing biomedical applications as it increases survival and proliferation molecular signals. To that end, increased expression of hypoxia-inducible factor-1α(Hif-1α) and vascular endothelial growth factor(VeGF) in the aqueous hydrogel matrix was achieved by the addition of a hypoxia mimetic deferoxamine mesylate(DFM). In this study, the formation of an endothelial network was achieved in a hydrogel matrix in the presence of the before mentioned 3 D printed metal foam. Cellular viability, fluorescent microscopy and scanning electron microscopy imaging analysis demonstrate that pre-osteoblasts undergo proliferation and also attach efficiently to the foam when exposed to DFM. Human umbilical vascular endothelial cells(HUVeCs) were grown in an extracellular matrix-like 3 D hydrogel and a hypoxia-like stress was achieved. This research demonstrates that pre-osteoblast cells undergo cell differentiation and increase the production of hydroxyapatite on exposure to the hypoxia mimetic molecule. This proposed approach encompasses an ideal prototype for a completely living implanted structure for future orthopedic implants.

From Cape Town to Cambridge: Orthopaedic trauma in contrasting environments

AIM: To compare the trauma experience gained by a trainee at a United Kingdom major trauma centre and a secondary level hospital in South ***: A profile of inpatient trauma cases during a five-week period in Addenbrooke's Hospital, Cambridge and Somerset Hospital, Cape Town was created. This was achieved by recording various parameters for each patient admitted including age, gender, injury, mechanism of injury and postal/area code. This, together with details of the departments themselves, allows a comparison of the amount and variety of orthopaedic trauma cases experienced by an individual trainee in each setting. ReSULTS: The trauma profiles differed significantly. Patients in Cape Town were younger and more likely to be male. In the young, injury in Cape Town was more likely to occur due to assault or being struck by a vehicle, whilst patients in Cambridge were more likely to be injured whilst in a vehicle or in high energy falls. In older patients, trauma at both centres was almost exclusively due to mechanical falls. In a given age group, injuries at the two centres were similar, however the majority of patients admitted to Addenbrooke's were elderly, resulting in less variation in the overall injury ***: The trauma profile of a major trauma centre in the United Kingdom is less varied than that of a South African secondary centre, with significantly fewer cases per surgeon. This suggests a more variedtraining experience in the developing world with a greater caseload.

Foot burns and diabetes: a retrospective study

Background: Diabetes in conjunction with a foot burn can compound the challenges in wound healing;however, the impact of diabetes on outcomes of patients with foot burns has not been examined. Methods: A retrospective notes audit was conducted at the Concord Hospital Burns Unit for patients with foot burns who were admitted from 1st January 2012 to 31st December 2013. Data were collected for 15 subjects with foot burns and diabetes and 18 subjects with foot burns and no diabetes as a control group. Subjects were matched for percentage total body surface area of burns. Results: The mean inpatient and total lengths of stay for the diabetic group were 21.27 days and 64.80 days, which were significantly longer (P = 0.090 and P = 0.054) than the 9.61 days and 30.56 days in the control, based on a significance level of 0.10. The diabetic group was significantly older (P = 0.001), at 56.60 years versus 39.44 years in the control. Significantly (P = 0.033) more patients with diabetes were not working (n = 12/15 or 80.00 % versus n = 7/18 or 38.89 %) compared to the control. The diabetic group had higher rates of regrafting (n = 3/15 or 20.00 % versus n = 1/18 or 5.55 %) than the control and significantly (P = 0.013) more amputations (n = 5/15 or 33.33 % versus n = 0 or 0.00 %) compared to the control. Fewer patients with diabetes were prescribed pressure garments (n = 2/15 or 13.33 % versus n = 9/18 or 50.00 %), which was significant (P = 0.034). The increased age of patients in the diabetic group correlates with results from other studies. Healing time may be reflected by total length of stay, which was more than double for patients with diabetes, increasing demand and cost of inpatient and outpatient services. Conclusion: This study highlights the importance of recognizing the potential for poorer outcomes for patients with diabetes and indicates the need for more burn prevention education and promotion in this 'at risk' patient group.

Dynamic distribution of TTK in HeLa cells: insights from an ultrastructural study

entry into mitosis is driven by signaling cascades of mitotic kinases. Our recent studies show that TTK, a kinetochore-associated protein kinase, interacts with CeNP-e, a mitotic kinesin located to corona fiber of kinetochore. Using immunoelectron microscopy, here we show that TTK is present at the nuclear pore adjacent complex of interphase HeLa cells. Upon nuclear envelope fragmentation, TTK targets to the outermost region of the developing kinetochores of monoorient chromosome as well as to spindle poles. After stable attachment, throughout chromosome congression, TTK is a constituent of the corona fibers, extending up to 90 nm away from the kinetochore outer plate. Upon metaphase alignment, TTK departs from the kinetochore and migrates toward the centrosomes. Taken together, this evidence strongly supports a model in which TTK functions in spindle checkpoint signaling cascades at both kinetochore and centrosome.

Precise dating of abrupt shifts in the Asian Monsoon during the last deglaciation based on stalagmite data from Yamen Cave, Guizhou Province, China

Based on 33 U/Th dates and 1020 oxygen isotopic data from stalagmite Y1 from Yamen Cave, Guizhou Province, China, a record of the Asian Summer Monsoon (ASM) was established. The record covers the last deglaciation and the early Holocene (from 16.2 to 7.3 ka BP) with an average oxygen isotope resolution of 9 years. The main millennial-scale deglacial events first identified in Greenland (Greenland Interstadial events: GIS 1e through GIS 1a) and later in China are clearly present in the Y1 record. By analogy to earlier work, we refer to these as Chinese Interstadials (CIS): CIS A.1e to CIS A.1a. The onset of these events in Y1 δ18O records are nominally dated at: 14750±50, 14100±60, 13870±80, 13370±80, and 12990±80 a BP. The end of CIS A.1a or the beginning of the Younger Dryas (YD) event is nominally at 12850±50 a BP and the end of the YD dates to 11500±40 a BP. The δ18O values shift by close to 3‰ during the transition into the Bφlling-Allerφd (BA, the onset of CIS A.1e) and at the end of the YD. Comparisons of Y1 to previously published early Holocene records show no significant phase differences. Thus, the east Asia Monsoon and the Indian Monsoon do not appear to have been out of phase during this interval. The Y1 record confirms earlier work that suggested that solar insolation and North Atlantic climate both affect the Asian Monsoon.
