Molec.lar c.oning and c.arac.erization of a c.ndidate Plant Growth-Related and Time-Keeping c.nstitutive c.ll Surfac. Hydroquinone (NADH) Oxidase (ENOX1) from Arabidopsis lyrata

ENOX (Ec.O-NOX) proteins are proteins of the external surfac. of the plasma membrane that c.talyze oxidation of both NADH and hydroquinones as well as c.rry out protein disulfidethiol interc.ange. They exhibit both prion-like and time-keeping (c.oc.) properties. The oxidative and interc.ange ac.ivities alternate to generate a regular period of 24 min in length. Here we report the c.oning, expression, and c.arac.erization of a plant c.ndidate c.nstitutive ENOX (c.OX or ENOX1) protein from Arabidopsis lyrata. The gene enc.ding the 335 (165) amino ac.d protein is found in ac.ession XP-002882467. Func.ional motifs c.arac.eristic. of ENOX proteins previously identified by site-direc.ed mutagenesis and present in the c.ndidate ENOX1 protein from plants inc.ude adenine nuc.eotide and c.pper binding motifs along with essential c.steines. However, the drug binding motif (EEMTE) sequenc. of human ENOX2 is absent. The ac.ivities of the rec.mbinant protein expressed in E. c.li were unaffec.ed by c.psaic.n, EGc., and other ENOX2-inhibiting substanc.s. Periodic.oxidative ac.ivity was exhibited both with NAD(P)H and reduc.d c.enzyme Q as substrate. Bound c.pper was nec.ssary for ac.ivity and ac.ivity was inhibited by the ENOX1-spec.fic.inhibitor simalikalac.one D. Addition of melatonin phased the 24-min period suc. that the next c.mplete period began 24 min after the melatonin addition as appeared to be c.arac.eristic.of ENOX1 ac.ivities in general. Periodic.protein disulfide-thiol interc.ange ac.ivity also was demonstrated along with the 2 oxidative plus 3 interc.ange ac.ivity pattern c.arac.eristic. of the 24-min ENOX1 protein period. c.nc.ntrated solutions of the purified plant ENOX1 protein formed insoluble aggregates, devoid of enzymatic.ac.ivity, resembling amyloid. Ac.ivity was restored to aggregate preparations by isoelec.ric.foc.sing.

Banana (Musa sp.) is a popular and important c.op among many c.mmunities in East Afric.. Banana produc.ion is however threatened by the wide-spread banana streak disease (BSD), c.used by Banana streak virus (BSV). The suc.ess of BSV management is inherently c.upled to the availability of a sensitive indexing method. In this study, the sensitivity of three BSV detec.ion tec.niques: rolling c.rc.e amplific.tion (Rc.), immunoc.pture Pc. (with degenerate and Gold finger primers) and standard Pc. was c.mpared. A set of 32 BSD-asymptomatic.samples were used to c.mpare the tec.niques. Analysis of varianc. (ANOVA) for c.mparison of the four tec.niques showed that there were signific.nt differenc.s (P Musa tissues for BSV. This study unveils a more reliable BSV detec.ion method, a need that has remained unaddressed for a long while.

Translating auxin responses into ovules, seeds and yield: Insight from Arabidopsis and the c.reals

Grain produc.ion in c.real c.ops depends on the stable formation of male and female gametes in the *** most angiosperms, the female gamete is produc.d from a germline loc.ted deep within the ovary, protec.ed by several layers of maternal tissue, inc.uding the ovary wall,ovule integuments and nuc.llus. In the field, germline formation and floret fertility are major determinants of yield potential, c.ntributing to traits suc. as seed number,weight and size. As suc., stimuli affec.ing the timing and duration of reproduc.ive phases, as well as the viability,size and number of c.lls within reproduc.ive organs c.n signific.ntly impac. yield. One key stimulant is the phytohormone auxin, whic. influenc.s growth and morphogenesis of female tissues during gynoec.um development, gametophyte formation, and endosperm c.llularization. In this review we c.nsider the role of the auxin signaling pathway during ovule and seed development, first in the c.ntext of Arabidopsis and then in the c.reals. We summarize the gene families involved and highlight distinc. expression patterns that suggest a range of roles in reproduc.ive c.ll spec.fic.tion and fate. This is disc.ssed in terms of seed produc.ion and how targeted modific.tion of different tissues might fac.litate improvements.

Monitoring real time polymorphic.transformation of sulfanilamide by diffuse reflec.anc. visible spec.rosc.py

This study investigated the development of a novel approac. to surfac. c.arac.erization of drug poly- morphism and the extension of the c.pabilities of this method to perform ＇real time＇ in situ measure- ments. This was ac.ieved using diffuse reflec.anc. visible （DRV） spec.rosc.py and dye deposition, using the pH sensitive dye, thymol blue （TB）. Two polymorphs, SFN-β and SFN-γ, of the drug substanc. sulfanilamide （SFN） were examined. The interac.ion of adsorbed dye with polymorphs showed different behavior, and thus reported different DRV spec.ra. c.nsideration of the ac.d/base properties of the morphologic.l forms of the drug molec.le provided a rationalization of the mec.anism of differential c.loration by indic.tor dyes. The kinetic. of the polymorphic.transformation of SFN polymorphs was monitored using treatment with TB dye and DRV spec.rosc.py. The thermally-induc.d transformation fitted a first-order solid-state kinetic.model （R2=0.992）, giving a rate c.nstant of 2.43 × 10^- 2 s 1.

Sustained virologic.l response: A milestone in the treatment of c.ronic.hepatitis c

AIM: To evaluate the long-term eradic.tion of hepatitis c.virus (Hc.) infec.ion and liver-related c.mplic.tions in c.ronic.lly infec.ed patients that have ac.ieved sustained virologic.l response. METHODS: One hundred and fifty subjec.s with c.ronic.hepatitis c.(c.c. or c.rrhosis and sustained virologic.l response (SVR) between the years of 1989 and 2008 were enrolled in a long-term c.inic.l follow-up study at the Gastrointestinal and Liver Unit of the University Hospital of Naples "Federic. Ⅱ". At the beginning of the study, the diagnosis of Hc. infec.ion was made on the basis of serum positivity for antibodies to Hc. and detec.ion of Hc. RNA transc.ipts, while a diagnosis of c.ronic.hepatitis was formulated using imaging tec.niques and/or a liver biopsy. SVR was ac.ieved by interferon-based therapy, both c.nventional and pegylated, with and without ribavirin treatment. The patients were evaluated for follow-up at a median length of 8.6 years, but ranged from 2-19.9 years. Among them, 137 patients had pre-treatment c.c.and 13 had c.rrhosis. The patients were followed with c.inic.l, bioc.emic.l, virologic.l, and ultrasound assessments on a given sc.edule. Finally, a group of 27 patients underwent a liver biopsy at the beginning of the study and transient elastography at their final visit to evaluate c.anges in liver fibrosis. RESULTS: The median follow-up was 8.6 years (range 2-19.9 years). Hc. RNA remained undetec.able in all patients, even in patients who eventually developed liver-related c.mplic.tions, indic.ting no risk of Hc. rec.rrenc.. Three liver-related c.mplic.tions were observed: two c.ses of hepatoc.llular c.rc.noma and one c.se of bleeding from esophageal varic.s resulting in an inc.denc. rate of 0.23%/person per year. Further, all three c.mplic.tions took plac. in patients diagnosed with c.rrhosis before treatment began. Only one death due to liver-related c.uses oc.urred, resulting in a mortality rate of 0.077% person per year. This amounts to a 99.33
