Limited proteomic response in the marine snail Melarhaphe neritoides after long-term emersion

Rocky intertidal organisms are commonly exposed to environmental gradients, promoting adapta-tions to these conditions. Emersion time varies along the intertidal range and in the supralittoralzone is frequently larger than a single tidal cycle, even lasting for weeks. The planktonic-dispersinggastropod Melarhaphe neritoides is a common species of the high shore, adapted to reduce waterloss in order to survive during long-term emersion. In this study, we investigated the molecular re-sponse, at the proteome level, of M. neritoides collected in high-shore tide pools to a series ofemersion periods, from 8 to 24 days, in laboratory conditions. We compared this response to indi-viduals maintained submerged during this period, because this was their original habitat. We alsoincluded a reversion treatment in the study, in which emersed individuals were returned to the sub-merged conditions. Although we detected an increase in overall protein concentration with longeremersion periods, contrary to general expectation, the two dimensional electrophoresis （2DE）-based proteomic analysis did not show significant differences between the treatments at the levelof individual protein spots, even after an emersion period of 24 days. Our results suggest that themetabolism remains unaltered independent of the treatment carried out or the changes are verysubtle and therefore difficult to detect with our experimental design. We conclude that M. neri-toides could be equally adapted to emersion and submersion without drastic physiologicalchanges.

AIM: To investigate the role of genes and kinetics of specific transcription factors in liver regeneration, and to analyze the gene expression and the activity of some molecules crucially involved in hepatic regeneration. METHODS: USING gel-shift assay and RT-PCR, transcription factors, such as NF-κb, STAT-3, SMAD3 and AP-1, and gene expression of inducible nitric oxide synthase (iNOS), hepatocyte growth factor (HGF) and c-met were analyzed in an animal model of chemically induced hepatectomy. RESULTS: Gene expression of HGF and its receptor c-met peaked at 3 h and 24 h after acute CCl4 intoxi- cation. iNOS expression was only observed from 6 to 48 h. Transcriptional factor NF-κb had an early activation at 30 min after acute liver damage. STAT-3 peaked 3 h post- intoxication, while AP-1 displayed a peak of activation at 48 h. SMAD3 showed a high activity at all analyzed times. CONCLUSION: TNF-α and IL-6 play a central role in hepatic regeneration. These two molecules are responsible for triggering the cascade of events and switch-on of genes involved in cell proliferation, such as growth factors, kinases and cyclins which are direct participants of cell proliferation.

The importance of MYC function in cancer was discovered in the late 1970s when the sequence of the avian retrovirus that causes myelocytic leukemia was *** then,over 40 years of unceasing research have highlighted the significance of this protein in malignant transformation,especially in hematologic ***,some of the earliest connections among the higher expression of proto-oncogenes(such as MYC),genetic rearrangements and their relation to cancer development were made in burkitt lymphoma,chronic myeloid leukemia and mouse *** myeloma(MM),in particular,is a plasma cell malignancy strictly associated with MYC deregulation,suggesting that therapeutic strategies against it would be beneficial in treating this ***,targeting MYC was-and,somehow,still is-challenging due to its unique properties:lack of defined three-dimensional structure,nuclear localization and absence of a targetable enzymatic *** these difficulties,however,many studies have shown the potential therapeutic impact of direct or indirect MYC *** molecules have been tested,in fact,in the context of *** this review,we summarize the current status of the different compounds,including the results of their clinical testing,and propose to continue with the efforts to identify,repurpose,redesign or improve drug candidates to combine them with standard of care therapies to overcome resistance and enable better management of myeloma treatment.

Effects of organic chromium supplementation to finishing lambs diet on growth performance, carcass characteristics and meat quality

The objective of this study was to evaluate supplemental organic chromium（Cr） to finishing lambs on growth performance, carcass characteristics, and meat quality. Eighteen Suffolk lambs（age（4.5±0.2） mon;（25.8±3.6） kg body weight（bW）） were randomly assigned to three levels of supplemental organic Cr（0.0, 0.2 and 0.4 mg kg^–1 dry matter（DM）） in a complete random design. Growth performance was evaluated for 70 d, and then lambs were slaughtered to study carcass characteristics and chemical composition of meat. Orthogonal contrasts were performed（contrast one-average level 0.2 ppm Cr vs. average level 0.4 ppm Cr; contrast two-level 0 vs. average levels（0.2＋0.4） ppm Cr）. Orthogonal polynomials were used to estimate the linear and quadratic effects of Cr concentrations. Growth and carcass performance were not affected by supplemental organic Cr. Muscle conformation and leg perimeter linearly increased（P〈0.05） as organic Cr level increased in the diet. Kidney fat decreased linearly（P〈0.05） as supplemental Cr increased. In Longissimus dorsi（LD）, the ash content decreased linearly, and shear force（kg cm^–2） increased（P〈0.05） as organic Cr level increased in the diet. It is concluded that organic Cr did not affect growth performance, but it improved positively the muscle conformation, reduced kidney fat, whereas in LD there was an increment in shear force in finishing carcass lambs.

Human bocavirus in children hospitalized for acute respiratory tract infection in Rome

background The role of human bocavirus (HboV) as a respiratory pathogen has not been fulfilled *** aimed to describe clinical and serological characteristics of children with HboV hospitalized for acute respiratory tract infection and to evaluate whether differences occur between HboV alone and in *** We retrospectively reviewed data from 60 children (median age of 6.2 months,range 0.6-70.9) hospitalized for acute respiratory symptoms,with HboV detected from a respiratory sample,using a reverse transcriptase-PCR for 14 respiratory viruses (including respiratory syncytial virus (RSV),influenza virus A and b,human coronavirus OC43,229E,NL-63 and HUK1,adenovirus,rhinovirus,parainfluenza virus 1-3,and human metapneumovirus).Results HboV was detected alone in 29 (48.3%) patients,while in co-infection with other viruses in 31 patients (51.7%),with a peak between December and *** the 60 patients,34 were bronchiolitis,19 wheezing,3 pneumonia,2 upper respiratory tract infection,and 2 whooping *** children (11.6%) required admission to the paediatric intensive care unit (PICU) for respiratory *** differences was observed in age,family history for atopy and/or asthma,clinical presentations,chest X-ray,or laboratory findings in children with HboV alone *** viral *** was the most frequently co-detected virus (61.3%).When compared with HboV detection alone,the co-detection of RSV and HboV was associated with male sex (P=0.013),younger age (P=0.01),and lower blood neutrophil count (P=0.032).Conclusions HboV can be detected alone and in co-infection respiratory samples of children with an acute respiratory tract infection.A cause-effect relationship between HboV and respiratory infection is not clear,so further studies are needed to clarify this point.

Effects of basic traditional Chinese diet on body mass index,lean body mass,and eating and hunger behaviours in overweight or obese individuals

ObJECTIVE:To compare the effects of a basic traditional Chinese diet with a Western standard diet on body mass index(bMI),lean body mass(LbD),and eating and hunger ***:A sample of 284 patients were randomized into 2 groups:group A(n = 142) followed a basic traditional Chinese diet(bCTD) and group b(n =142) followed a Western standard diet(WSD).both diets were set at approximately 1200 *** patients enrolled were compared before treatment and 6 weeks after treatment,and then follow-ups were made at 1 year and 5 ***:In the bCTD group,bMI decreased by0.46 kg/m2 and LbM by 0.25 kg,versus respectively0.28 kg/m2 and 0.41 kg in the WSD *** of eating self-assessment,hunger measurement and psychophysical indices of health were also in favour of the ***:Outcomes show that the bCTD has a better effect on bMI and LbM,as well as over the medium/long term,and provides stronger psychophysical support to obese patients.

Effect of Lactobacillus rhamnosus HN001 and bifidobacterium longum bb536 on the healthy gut microbiota composition at phyla and species level: A preliminary study

AIMTo evaluate the ability of Lactobacillus rhamnosus HN001 and bifidobacterium longum bb536 to colonize the intestinal environment of healthy subjects and modify the gut microbiota *** healthy Italian volunteers, eight males and twelve females, participated in the study. Ten subjects took a sachet containing 4 × 109 colony-forming units (CFU) of bifidobacterium longum bb536 and 109 CFU of Lactobacillus rhamnosus HN001, 30 min before breakfast (pre-prandial administration), while ten subjects took a sachet of probiotic product 30 min after breakfast (post-prandial administration). The ability of Lactobacillus rhamnosus HN001 and bifidobacterium longum bb536 to colonize human gut microbiota was assessed by means of quantitative real-time PCR, while changes in gut microbiota composition were detected by using Ion Torrent Personal Genome *** after 1-mo of probiotic administration, b. longum bb536 and L. rhamnosus HN001 load was increased in the majority of subjects in both pre-prandial and post-prandial groups. This increase was found also 1 mo after the end of probiotic oral intake in both groups, if compared to samples collected before probiotic consumption. At phyla level a significant decrease in Firmicutes abundance was detected immediately after 1-mo of b. longum bb536 and L. rhamnosus HN001 oral intake. This reduction persisted up to 1 mo after the end of probiotic oral intake together with a significant decrease of Proteobacteria abundance if compared to samples collected before probiotic administration. Whereas, at species level, a higher abundance of blautia producta, blautia wexlerae and Haemophilus ducrey was observed, together with a reduction of Holdemania filiformis, Escherichia vulneris, Gemmiger formicilis and Streptococcus sinensis abundance.
