leptin Modulates the Differentiation of Keratinocytes via Autophagy in Psoriasis Patients with Metabolic Syndrome

objective: Psoriasis is associated with a high prevalence of metabolic syndrome (MS), and patients with concomitant psoriasis and MS are more severely affected and less responsive to treatment. However, the molecular mechanisms behind these effects are unknown. Recent studies have shown that leptin may serve as a molecular link between psoriasis and MS, suggesting that high leptin concentrations may ex.cerbate psoriasis. However, the molecular mechanism of this effect is still unclear. We aimed to investigate the effect of leptin on autophagy in patients with psoriasis. Methods: From January 2021 to June 2022 in PlA General Hospital, we enrolled 51 patients with psoriasis, including 21 patients with MS and 30 without MS, and 30 healthy controls who had undergone nevus surgery. We measured the epidermal leptin, P62, and lC3B concentrations of patients by immunohistochemistry, and measured the serum leptin concentration by enzyme-linked immunosorbent assay. We then performed correlation analyses to compare these proteins' concentrations between patients with concomitant psoriasis and MS, patients with psoriasis alone, and healthy control groups. Additionally, we performed western blotting after in vitro culture of HaCaT cells with different concentrations of leptin and measured the ex.ression levels of the autophagy markers Beclin1, lC3B, and P62; the differentiation markers K10, K16, and K17; and PI3K/AKT/mToR signaling pathway-related proteins of HaCat cells. Nex., we transfected ATG5 into HaCaT cells to revert autophagy and used the specific PI3K inhibitor lY294002 to block PI3K/AKT/mToR signaling. The ex.ression levels of K10, K16, and K17 of HaCat cells were again measured. one-way analysis of variance was used for the comparison of means of multiple samples, and lSD-t post hoc test was used for comparison between the 2 groups. Virtually coupled train set control subject to space-time separation: A distributed economic MPC approach with emergency braking configuration

The emerging virtual coupling technology aims to operate multiple train units in a Virtually Coupled Train Set (VCTS) at a minimal but safe distance. To guarantee collision avoidance, the safety distance should be calculated using the state-of-the-art space-time separation principle that separates the Emergency Braking (EB) trajectories of two successive units during the whole EB process. In this case, the minimal safety distance is usually numerically calculated without an analytic formulation. Thus, the constrained VCTS control problem is hard to address with space-time separation, which is still a gap in the ex.sting literature. To solve this problem, we propose a Distributed Economic Model Predictive Control (DEMPC) approach with computation efficiency and theoretical guarantee. Specifically, to alleviate the computation burden, we transform implicit safety constraints into ex.licitly linear ones, such that the optimal control problem in DEMPC is a quadratic programming problem that can be solved efficiently. For theoretical analysis, sufficient conditions are derived to guarantee the recursive feasibility and stability of DEMPC, employing compatibility constraints, tube techniques and terminal ingredient tuning. Moreover, we ex.end our approach with globally optimal and distributed online EB configuration methods to shorten the minimal distance among VCTS. Finally, ex.erimental results demonstrate the performance and advantages of the proposed approaches. © 2024 Beijing Jiaotong University

The generation of ultrashort high-power light sources in the mid-infrared(mid-IR)to terahertz(THz)range is of interest for applications in a number of fields,from fundamental research to biology and *** conventional laser technology,photon deceleration in plasma wakes provides an alternative approach to the generation of ultrashort mid-IR or THz ***,we present a photon deceleration scheme for the efficient generation of ultrashort mid-IR or THz pulses by using an intense driver laser pulse with a relatively short wavelength and a signal laser pulse with a relatively long *** signal pulse trails the driver pulse with an appropriate time delay such that it sits at the front of the second wake bubble that is driven by the driver *** to its relatively long wavelength,the signal pulse will be subjected to a large gradient of the refractive index.in the plasma wake ***,the photon deceleration in the plasma wake becomes faster and more efficient for signal pulses with longer *** greatly enhances the capacity and efficiency of photon deceleration in the generation of ultrashort high-power light sources in the long-wavelength IR and THz spectral ranges.

Microarray analysis of signalling interactions between inflammation and angiogenesis in subchondral bone in temporomandibular joint osteoarthritis

Inflammation and angiogenesis, the major pathological changes of osteoarthritis (oA), are closely associated with joint pain; however, pertinent signalling interactions within subchondral bone of osteoarthritic joints and potential contribution to the peripheral.origin of oA pain remain to be elucidated. Herein we developed a unilateral anterior crossbite mouse model with osteoarthritic changes in the temporomandibular joint. Microarray-based transcriptome analysis, besides quantitative real-time polymerase chain reaction, was performed to identify differentially ex.ressed genes (DEGs). overall, 182 DEGs (fold change ≥ 2, P ontrol and unilateral anterior crossbite groups: 168 were upregulated and 14 were downregulated. on subjecting significant DEGs to enrichment analyses, inflammation and angiogenesis were identified as the most affected. Inflammation-related DEGs were mainly enriched in T cell activation and differentiation and in the mammalian target of rapamycin/nuclear factor-κB/tumour necrosis factor signalling. Furthermore, angiogenesis-related DEGs were mainly enriched in the Gene ontology terms angiogenesis regulation and vasculature development and in the KEGG pathways of phosphoinositide 3-kinase-protein kinase B/vascular endothelial growth factor/hypox.a-inducible factor 1 signalling. Protein– protein interaction analysis revealed a close interaction between inflammation-and angiogenesis-related DEGs, suggesting that phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit delta (Pi3kcd), cathelicidin antimicrobial peptide (Camp), C-x.C motif chemokine receptor 4 (Cx.r4), and MYB proto-oncogene transcription factor (Myb) play a central role in their interaction. Background: Hypox.a is a significant risk factor of hypertension. However, no studies have used transcutaneous tissue partial pressure of ox.gen (TcPo2) and partial pressure of carbon diox.de (TcPCo2) monitors to measure the respective partial pressures in healthy individuals. ox.gen saturation (Spo2) is often used for traditional monitoring of vital signs. This study investigated the changes in TcPo2 and Spo2 values during rapid changes in altitude. The trial was registered at *** (registration no. NCT06076057). Methods: Healthy adult volunteers were instructed to sit vertically in a hypobaric ox.gen chamber, which ascended from 0 m to 2500 m at a uniform speed within 10 min. The Danish Radiometer TCM4 was used to measure TcPo2 and TcPCo2 with the ventral side of the upper arm as the measurement site. The Shenzhen Kerokan P0D-1 W pulse ox.meter was used to measure heart rate and Spo2, with values recorded once every 500 m. Results: Al.ogether, 49 healthy volunteers were recruited between March 2023 and August 2023. With increasing altitude, TcPo2 and Spo2 decreased significantly (P om 0 m, TcPo2 began to change statistically at 500 m (P o2 began to change statistically at 1000 m (P ltitude, the difference in TcPo2 was greater than the difference in Spo2. Reconstruction of bone defects or fractures caused by ageing, trauma and tumour resection is still a great challenge in clinical treatment. Although autologous bone graft is considered as gold standard, the source of natural bone is limited. In recent years, regenerative therapy based on bioactive materials has been proposed for bone reconstruction. Specially, numerous studies have indicated that bioactive ceramics including silicate and phosphate bioceramics ex.ibit ex.ellent osteoinductivity and osteoconductivity, further promote bone regeneration. In addition, magnesium (Mg) element, as an indispensable mineral element, plays a vital role in promoting bone mineralisation and formation. In this review, different types of Mg-containing bioceramics including Mg-containing calcium phosphate-based bioceramics (such as Mg-hydrox.apatite, Mg-biphasic calcium phosphate), Mg-containing calcium silicate-based bioceramics (such as Mg2Sio4, Ca2MgSi2o7and Mg-doped bioglass), Mg-based biocements, Mg-containing metal/polymer-bioceramic composites were systematacially summarised. Additionally, the fabrication technologies and their materiobiological effects were deeply discussed, Clinical applications and perspectives of magnesium-containing bioceramics for bone repair are highlighted. overall, Mg-containing bioceramics are regarded as regenerative therapy with their optimised performance.
