Molecular mechanisms of apoptosis induced by Scorpio water extract in human hepatoma HepG2 cells

AIM: To clarify the mechanism underlying the anti-mutagenic and anti-cancer activities of Scorpio water extract (SWE). METHODS: Human hepatoma HepG2 cells were incubated with various concentrations of SWE. After 24-h incubation, cytotoxicity and apoptosis evaluations were determined by MTT and DNA fragmentation assay, respectively. After treatment with SWE, mitochondrial membrane potential (MMP) was determined by measuring the retention of the dye 3,3'-dihexyloxacarbocyanine (DiOC6(3)) and the protein expression including cytochrome C and poly-(ADPribose) polymerase (PARP) were measured by Western blotting. Caspase-3 and -9 enzyme activities were measured using specific fluorescence dyes such as Ac-DEVD-AFC and Ac-LEHD-AFC. RESULTS: We found that treatment with SWE induced apoptosis as confirmed by discontinuous DNA fragmentation in cultured human hepatoma HepG2 cells. Our investigation also showed that SWE-induced apoptosis of HepG2 cells were associated with intracellular events including disruption of MMP, increased translocation of cytochrome C from mitochondria to cytosol, activation of caspase-3, and PARP. Pre-treatment of N-acetyl-Asp-Glu-Val-Asp-CHO (Ac-DEVD-CHO), a caspase-3 specific inhibitor, or cyclosporin A (CsA), an inhibitor of MMP disruption, completely abolished SWE-induced DNA fragmentation. CONCLUSION: These results suggest that SWE possibly causes mitochondrial damage, leading to cytochrome C release into cytosol and activation of caspases resulting in PARP cleavage and execution of apoptotic cell death in HepG2 cells. These results further suggest that Scorpio may be a valuable agent of therapeutic intervention of human hepatomas.

Molecular dynamics （MD） computer simulation techniques, as a powerful tool commonly utilized by the liquid crystal display （LCD） community, usually are employed for computing the equilibrium and transport properties of a classical many body system, since they are very similar to real experiments in many respects. In this paper we pre- sent molecular dynamics computer simulation results taken for a mixture of the two different kinds of nematic liq- uid crystals （LCs）. We calculated rotational viscosity from Brownian behavior with friction of the mean director of the mixture comprising pentylcyanobiphenol （5CB） and decylcyanobiphenol （10CB） by using molecular dynamics computer simulation, where intermolecular potential parameter is Generalized AMBER force field （GAFF）. Our computed results show a good agreement with the experimental results.

Label-free Detection for a DNA Methylation Assay Using Raman Spectroscopy

Background： DNA methylation has been suggested as a biomarker for early cancer detection and treatment. Varieties of technologies for detecting DNA methylation have been developed, but they are not sufficiently sensitive for use in diagnostic devices. The aim of this study was to determine the suitability of Raman spectroscopy for label-free detection of methylated DNA. Methods： The methylated promoter regions of cancer-related genes cadherin 1 （CDH1） and retinoic acid receptor beta （RARB） served as target DNA sequences. Based on bisulfite conversion, oligonucleotides ofmethylated or nonmethylated probes and targets were synthesized for the DNA methylation assay. Principal component analysis with linear discriminant analysis （PCA-DA） was used to discriminate the hybridization between probes and targets （methylated probe and methylated target or nonmethylated probe and nonmethylated target） of CDH! and RARB from nonhybridization between the probe and targets （methylated probe and nonmethylated target or nonmethylated probe and methylated target）. Results： This study revealed that the CDH1 and RARB oligo sets and their hybridization data could be classified using PCA-DA. The classification results for CDH1 methylated probe ＋ CDH1 methylated target versus CDH! methylated probe ＋ CDHI unmethylated target showed sensitivity, specificity, and error rates of 92%, 100%, and 8%, respectively. The classification results for the RARB methylated probe ＋ RARB methylated target versus RARB methylated probe ＋ RARB unmethylated target showed sensitivity, specificity, and error rates of 92%, 93%, and 11%, respectively. Conclusions： Label-free detection ofDNA methylation could be achieved using Raman spectroscopy with discriminant analysis.

Food waste is recognized as a valuable source for potential agricultural applications to supply organic matter and nutrients to arable ***,the information on the combined application of food waste and the plant growth-promoting bacterial strain,Chlorella,related to plant metabolic features and sodium chloride content in arable soil is *** present study was conducted to investigate the exogenous application of food waste along with Chlorella,which improved the physio-morphological features of red *** results revealed that this combination enhanced the organic matter in the soil,ultimately improving the fertility rate of the soil,and the physio-morphological features,such as chlorophyll a content(24.5±0.7),root(7.8±0.7)cm and shoot length(12.1±0.7)cm,fresh weight(2.1±0.05)g,dry weight(0.19±0.05)g,mineral contents,and hormonal concentration(ABA by up to 2 folds).The combined treatment also minimized free radicals via the activation of the intrinsic antioxidant series cascade and electrolyte *** findings showed that adding Chlorella and food wastes improved growth characteristics and can be used as a green bio-fertilizer for sustainable agriculture.

Primary adenocarcinoma of the appendix is a rare malignancy that constitutes 〈 0.5% of all gastrointestinal neoplasms. Moreover, primary signet ring cell carc noma of the appendix is an exceedingly rare entity. We have encountered 15 cases of primary appendiceal cancer among 3389 patients who underwent appen- dectomy over the past 18 years. In the present report, we describe a rare case of primary signet ring cell carcinoma of the appendix with ovarian metastases and unresectable peritoneal dissemination occurring in a 67-year-old female patient. She underwent ap- pendectomy and bilateral salpingo-oophorectomy with a laparoscopy procedure. She then received palliative systemic chemotherapy with 12 cycles of oxaliplatin, 5-flurorouracil, and leucovorin （FOIFOX-4）. The patient currently is well without progression of disease 12 mo after beginning chemotherapy.

背景:由于关节软骨再生修复能力有限,自体软骨细胞移植技术为关节软骨损伤的修复提供了较好的修复材料。目的:多中心评估自体软骨细胞移植的短期疗效与安全性。设计、时间及地点:自身前后对照观察,于2001-03/2006-04在韩国80所附属医院完成。对象:软骨缺损患者261例,男169例,女92例,年龄15～70岁,平均36.47岁。软骨缺损面积2.0～20.0cm2。其中股骨髁损伤215例,滑车30例,髌骨25例,胫骨2例。方法:取关节非负重区软骨组织200～300mg,体外培养扩增后将软骨细胞植入患者关节缺损区。于移植后6个月对所有患者进行随访,膝关节功能评估标准采用美国膝关节协会评分系统[Knee society score-A(KSS-A)和Knee society score-B(KSS-B)]。主要观察指标:自体软骨细胞膝关节缺损区移植后KSS-A和KSS-B评分。结果:获得随访患者261例。患者移植前KSS-A评分为63.55±18.47,移植后6个月为88.74±11.47,差异有显著性意义(P<0.05)移植前KSS-B评分为59.56±24.92,移植后6个月为85.13±14.67,差异有显著性意义(P<0.05)。患者移植前后膝关节疼痛、活动度、前后稳定性、屈曲挛缩、伸直迟滞、行走能力和上下楼能力均明显改善(P<0.05)。综合评价移植有效率为97.3%,优良率为89.3%。结论:自体软骨细胞移植可有效的治疗关节软骨缺损,促进膝关节功能的恢复。

The use of natural killer(NK)cells is a promising and safe immunotherapeutic approach in the field of cancer ***,combination treatments are required to enhance the effector functions and therapeutic efficacy of NK *** this study,we investigated the potential of daratumumab(Dara),bortezomib,and dexamethasone(Dvd)to augment the antitumor effects of NK cells in a multiple myeloma(MM)xenograft mouse *** cells were expanded and activated using the K562-OX40 ligand and membrane-bound IL-18 and IL-21 in the presence of IL-2 and IL-15 from peripheral blood mononuclear cells from MM patients.A human MM xenograft model was established using human RPMI8226-RFP-FLuc cells in NOD/SCID IL-2Rγnull(NSG)***-bearing mice were divided into six treatment groups:no treatment,expanded NK cells(eNKs),Dara,Dara+eNKs,Dvd,and Dvd+*** treatment strongly enhanced the cytotoxicity of eNKs by upregulating expression of NK cell activation ligands,downregulating expression of NK cell inhibitory ligands,and promoting antibody-dependent cellular *** combination of eNKs with Dvd significantly prolonged mouse survival and reduced the tumor burden and serum M-protein ***,Dvd pretreatment significantly increased eNK persistence and homing to MM *** findings suggest that Dvd treatment potentiates the antimyeloma effects of NK cells expanded and activated ex vivo by modulating immune responses in MM-bearing mice.

Transmission and Suppression of Avian Influenza Virus

The growth of AIV is possible with supply of minerals from aeolian desert dust, as cofactors of enzymes, and thus the blockage of the aeolian desert dusts to the poultry farmhouse is important to suppress the enzymatic activity of AIV. South Georgia of Subantarctic Islands may be the source of Continental AIV, so that Continental AIV can become endemic within the local bird population when surrounding environments are close to Subantarctica of South Georgia. There are food webs in sequence of aquatic virus, bacteria, phytoplankton, zooplankton, krill, fish and squid while penguins feed on the fish or squid infected by mutant virus to generate LPAI after an incubation period. When migratory birds move to the Continents, HPAI can spread in the Continents due to low UV-B radiation, enough feed of rice, wheat, corn, wetland, and mineral-enriched desert dust. The clean room is equipped with air washers to remove aeolian dusts and migratory birds feces from working personnel and equipments, heaters to keep the pasteurisation of ultra-high temperature at 135°C for 1 - 2 seconds to inactivate AIV, humidifier to keep wet state above 65% relative humidity for weak virus activity, and the UV-C lamps (254 nm) to finally inactivate AIV. Since AIV doesn’t like the high salinity and high relative humidity, seawater is blanketed as sprayer to maintain high salinity (>20 ppt) and high relative humidity (>65%) from the top to the bottom of the poultry farmhouses for suppression of AIV transmission from infected aeolian dust coming from outside.
