Development of a Method to Produce Chromosome Lacking Lines (CLLs) in <i>Nicotiana tabacum</i>L. “Red Russian”

Monosomic lines of Nicotiana tabacum are helpful to confirm the location of genes on specific chromosomes. In the cross N. nudicaulis and N. tabacum, hybrid seedlings express lethal symptoms, which are controlled by the S subgenome of N. tabacum. To identify the responsible chromosome, we needed to produce chromosome lacking lines (CLLs) of N. tabacum L. “Red Russian” and use them to cross with N. nudicaulis. From a cross of (N. tabacum × N. tomentosiformis) × N. tabacum, 380 BC1 individuals were obtained. Using a Haplo-Q line (a monosomic line lacking the single linkage group 11) and N. tabacum, we found that qPCR is a simple and reliable screening method for CLLs of N. tabacum. The marker PT30342 is located on linkage group 11, and the -Ct value (Ct Actin - Ct PT30342) was 2.0 for a disomic line and was 1.097 for a Haplo-Q line. By the use of flow cytometry, qPCR and chromosome counting together as a screening method, we identified 6 CLLs lacking 2 to 6 chromosomes. Compared with conventional methods, our method is a rapid technique for making and screening CLLs ofthe S or S/T subgenome of N. tabacum. Further, these CLLs will be useful to identify the location of two or more factors on chromosomes controlling a variety of genetic problems affecting breeding. Here, we only made CLLs of the S or S/T subgenome of N. tabacum. We will use the method established in this study to produce CLLs of the T subgenome of N. tabacum, and gather a full set of CLLs of N. tabacum. qPCR could also be applied to the identification of chromosome aberrations in other plants.

Safety assessment of dextrin hydrogel adhesion barrier (AdSpray®) for elective laparoscopic cholecystectomy

Objective:Adhesion barriers have not traditionally been used during laparoscopic cholecystectomy(lapchole),and so there are no data confirming the safety of the dextrin hydrogel adhesion barrier(AdSpray®)for the use during *** aim of this prospective study is to investigate the safety of AdSpray®for elective ***:Between July 2019 and March 2021,we conducted a prospective study at Saiseikai Kumamoto Hospital to assess the safety of applying AdSpray®around the hepatoduodenal ligament,liver surface,and port area after extraction of the gallbladder during elective *** assessed patient morbidity,mortality,and laboratory data on postoperative day 1 and at the outpatient follow-up ***:A total of 100 patients(43 men,57 women)received AdSpray®application during elective lapchole at our *** mean patient age was 56.5±14.8 y,and the mean body mass index was 25.1±4.8 kg/*** gallbladder drainage was performed in 3(3.0%)patients as conservative treatment for acute *** mean operative time was 91.7±35.1 min,and the median blood loss was 5 mL(range,5e120 mL).Intraoperative gallbladder perforation was observed in 4.0%of patients(n=4).There was no intraoperative bile duct ***(1.0%)patient had a postoperative subhepatic fluid collection without bacterial *** mean white blood cell count and C-reactive protein level was significantly elevated on postoperative day 1 but returned to preoperative levels by the time of the follow-up *** patients were successfully discharged after surgery,and the median postoperative hospital stay was 2 d(range,2e9 d).Conclusion:Applying AdSpray®during elective lap-chole is safe,with an acceptable rate of postoperative complications.

Objective: To assess the efficacy of interferon beta-1b (IFNβ-1b) in Japane se patients with relapsing-remitting multiple sclerosis (RRMS). Background: The effects of IFNβin RRMS have been assessed in study populations comprised predo minantly of white patients. MS in Japanese patients is different from that in wh ite patients in that there are two different presentations classic MS (C-MS) an d optic-spinal MS (OS-MS)and chronic progressive forms are infrequent. Methods : A total of 205 Japanese patients with RRMS were randomized to receive 50 μg o r 250 μg (1.6 or 8.0 MIU) IFNβ-1b administered SC every other day for up to 2 years. The primary endpoint was annual relapse rate. Secondary endpoints includ ed further relapse-related and MRI outcome measures, as well as changes in Expa nded Disability Status Scale and Neurologic Rating *** was assessed i n 188 patients, and safety was assessed in 192 patients. Supplemental ad hoc sub group analyses were also performed for patients with OS-MS and those with C-MS .Results: Annual relapse rates were 0.763 in the 250 μg group and 1.069 in the 50 μg group, a relative reduction of 28.6%(p = 0.047). Results for all seconda ry endpoints favored 250μg IFNβ-1b. Subgroup analyses suggested that the magn itude and direction of treatment effect in patients with OS-MS and C-MS was si milar, albeit not significant due to small sample size. Conclusions: Interferon beta-1b (IFNβ-1b) 250 μg significantly reduced relapse rates and change in M RI lesion area in Japanese patients with relapsing-remitting multiple sclerosis ,and seemed to be comparably effective in optic-spinal multiple sclerosis (MS) and classic MS. The response to treatment with IFNβ1b in Japanese patients with MS suggests that a common pathogenesis and underlying genetic characteristics a re shared with white patients.
