Galvanic replacement reactions have been widely used to transform solution dispersed silver template structures into intricate nanoshell geometries. Here, we report on the use of these same reactions to form hollow substrate-supported Au-Ag nanoshells from silver templates having a heteroepitaxial relationship with the underlying single crystal substrate. The structures obtained exhibit a nanohut geometry, show highly tunable plasmonic properties and are formed as periodic arrays using a lithography-free technique. When removed from the substrate the inverted nanohuts appear as nanobowls with a notch in the rim. The study lays the groundwork for wafer-based devices utilizing nanoshells located at site-specific locations.

Expression Profiling and Structural Characterization of Micro RNAs in Adipose Tissues of Hibernating Ground Squirrels

Micro RNAs（mi RNAs） are small non-coding RNAs that are important in regulating metabolic stress. In this study, we determined the expression and structural characteristics of 20 mi RNAs in brown（BAT） and white adipose tissue（WAT） during torpor in thirteen-lined ground squirrels. Using a modified stem-loop technique, we found that during torpor, expression of six mi RNAs including let-7a, let-7b, mi R-107, mi R-150, mi R-222 and mi R-31 was significantly downregulated in WAT（P 〈 0.05）, which was 16%–54% of euthermic non-torpid control squirrels,whereas expression of three mi RNAs including mi R-143, mi R-200 a and mi R-519 d was found to be upregulated by 1.32–2.34-fold. Similarly, expression of more mi RNAs was downregulated in BAT during torpor. We detected reduced expression of 6 mi RNAs including mi R-103 a, mi R-107, mi R-125 b, mi R-21, mi R-221 and mi R-31（48%–70% of control）, while only expression of mi R-138 was significantly upregulated（2.91 ± 0.8-fold of the control, P 〈 0.05）. Interestingly,mi RNAs found to be downregulated in WAT during torpor were similar to those dysregulated in obese humans for increased adipogenesis, whereas mi RNAs with altered expression in BAT during torpor were linked to mitochondrial b-oxidation. mi RPath target prediction analysis showed that mi RNAs downregulated in both WAT and BAT were associated with the regulation of mitogen-activated protein kinase（MAPK） signaling, while the mi RNAs upregulated in WAT were linked to transforming growth factor b（TGFb） signaling. Compared to mouse sequences, no unique nucleotide substitutions within the stem-loop region were discovered for the associated pre-mi RNAs for the mi RNAs used in this study, suggesting no structure-influenced changes in pre-mi RNA processing efficiency in the squirrel. We report the development and experimental implementation of the automated experiment workflows for the identification of thebest predictive channel for a phenomenon of interest in spectroscopic measurements. The approach is based on the combinationof ensembled deep kernel learning for probabilistic predictions and a basic reinforcement learning policy for channel selection. Itallows the identification of which of the available observational channels, sampled sequentially, are most predictive of selectedbehaviors, and hence have the strongest correlations. We implement this approach for multimodal imaging in piezoresponse forcemicroscopy (PFM), with the behaviors of interest manifesting in piezoresponse spectroscopy. We illustrate the best predictivechannel for polarization-voltage hysteresis loop and frequency-voltage hysteresis loop areas is amplitude in the model samples. Thesame workflow and code are applicable for any multimodal imaging and local characterization methods.

Strategies are sought to reduce the impact of nonpoint sources of pollution,including fecal bacteria,and meet TMDL *** filter strips(VFSs)are one such strategy,though the data of VFS function under field conditions,particularly for fecal bacteria,are *** objective of this study was to evaluate the efficiencies in removing fecal bacteria,N,and P from VFSs installed to treat contaminated runoff from a working *** were found to be effective in removing a broad range of constituents from beef feedlot runoff pretreated by a settling *** first 30 m provided most or all of the reductions found within the 150 m VFSs studied:reductions average 85%of inflow water,85%of sediment,77%of N,and 84%of *** bacteria removal by the VFSs was about one order of magnitude:reductions at 30 m ranged from 83.5%for FC and FS to 91%for *** the site being studied,this provided an important level of protection and reduced surface-flow concentrations of fecal coliforms to below the 200 CFU/100 mL(coliform-forming units,CFU)water-quality standard for Kansas.

Although the structural and functional effects of exercise on the heart are well established,the metabolic changes that occur in the heart during and after exercise remain *** this study,we used metabolomics to assess time-dependent changes in the murine cardiac metabolome following 1 session of treadmill *** the exercise bout,we also recorded blood lactate,glucose,and ketone body levels and measured cardiac mitochondrial *** both male and female mice,moderate-and high-intensity exercise acutely increased blood lactate *** both sexes,low-and moderate-intensity exercise augmented circulating 3-hydroxybutryrate levels immediately after the exercise bout;however,only in female mice did high-intensity exercise increase 3-hydroxybutyrate levels,with significant increases occurring 1 h after the exercise *** metabolomics analyses of sedentary female and male hearts suggest considerable sex-dependent differences in basal cardiac metabolite levels,with female hearts characterized by higher levels of pantothenate,pyridoxamine,homoarginine,tryptophan,and several glycerophospholipid and sphingomyelin species and lower levels of numerous metabolites,including acetyl coenzyme A,glucuronate,gulonate,hydroxyproline,prolyl-hydroxyproline,carnosine,anserine,and carnitinylated and glycinated species,as compared with male *** after a bout of treadmill exercise,both male and female hearts had higher levels of corticosterone;however,female mice showed more extensive exercise-induced changes in the cardiac metabolome,characterized by significant,time-dependent changes in amino acids(e.g.,serine,alanine,tyrosine,tryptophan,branched-chain amino acids)and the ketone body *** from experiments using isolated cardiac mitochondria suggest that high-intensity treadmill exercise does not acutely affect respiration or mitochondrial coupling;however,female cardiac mitochondria demonstrate generally higher adenosi

BACKGROUND： Cortical motor neurons, also known as upper motor neurons, are large projection neurons whose axons convey signals to lower motor neurons to control the muscle movements. Degeneration of cortical motor neuron axons is implicated in several debilitating disorders including hereditary spastic paraplegia （HSP）. Since the discovery of the first HSP gene, SPASTthat encodes spastin, over 70 distinct genetic loci associated with HSP have been identified. How the mutations of these functionally diverse genes result in axonal degeneration and why certain axons are affected in HSP remain largely unknown. The development of induced pluripotent stem cell （iPSC） technology has provided researchers an excellent resource to generate patient-specific human neurons to model human neuropathological processes including axonal defects. METHODS： In this article, we will first review the pathology and pathways affected in the common forms of liSP subtypes by searching the PubMed database. We will then summarize the findings and insights gained from studies using iPSC-based models, and discuss challenges and future directions. RESULTS： HSPs, a heterogeneous group of genetic neurodegenerative disorders, exhibit similar pathological changes that result from retrograde axonal degeneration of cortical motor neurons. Recently, iPSCs have been generated from several common forms of HSP including SPG4, SPG3A, and SPG11 patients. Neurons derived from HSP iPSCs exhibit impaired neurite outgrowth, increased axonal swellings, and reduced axonal transport, recapitulating disease-specific axonal defects. CONCLUSIONS： These patient-derived neurons offer a unique tool to study the pathogenic mechanisms and explore the treatments for rescuing axonal defects in HSP, as well as other diseases involving axonopathy.
