Beta-nerve growth factor gene therapy alleviates pyridoxine-induced neuropathic damage by increasing doublecortin and tyrosine kinase A in the dorsal root ganglion

Beta-nerve growth factor(β-NGF) is known to be a major leading cause of neuronal plasticity. To identify the possible action mechanisms of β-NGF gene therapy for sciatic nerve recovery, experimental dogs were randomly divided into control, pyridoxine, and pyridoxine + β-NGF groups. We observed chronological changes of morphology in the dorsal root ganglia in response to pyridoxine toxicity based on cresyl violet staining. The number of large neurons positive for cresyl violet was dramatically decreased after pyridoxine intoxication for 7 days in the dorsal root ganglia and the neuron number was gradually increased after pyridoxine withdrawal. In addition, we also investigated the effects of β-NGF gene therapy on neuronal plasticity in pyridoxine-induced neuropathic dogs. To accomplish this, tyrosine kinase receptor A(TrkA), βIII-tubulin and doublecortin(DCX) immunohistochemical staining was performed at 3 days after the last pyridoxine treatment. TrkA-immunoreactive neurons were dramatically decreased in the pyridoxine group compared to the control group, but strong TrkA immunoreactivity was observed in the small-sized dorsal root ganglia in this group. TrkA immunoreactivity in the dorsal root ganglia was similar between β-NGF and control groups. The numbers of βIII-tubulin-and DCX-immunoreactive cells decreased significantly in the pyridoxine group compared to the control group. However, the reduction of βIII-tubulin-and DCX-immunoreactive cells in the dorsal root ganglia in the β-NGF group was significantly ameliorated than that in the pyridoxine group. These results indicate that β-NGF gene therapy is a powerful treatment of pyridoxine-induced neuropathic damage by increasing the TrkA and DCX levels in the dorsal root ganglia. The experimental protocol was approved by the Institutional Animal Care and Use Committee(IACUC) of Seoul National University, South Korea(approval No. SNU-060623-1, SNU-091009-1) on June 23, 2006 and October 9, 2009, respectively.

Glucose metabolism and neurogenesis in the gerbil hippocampus after transient forebrain ischemia

Recent evidence exists that glucose transporter 3（GLUT3） plays an important role in the energy metabolism in the *** previous studies have been conducted using focal or hypoxic ischemia models and have focused on changes in GLUT3 expression based on protein and m RNA levels rather than tissue *** the present study,we observed change in GLUT3 immunoreactivity in the adult gerbil hippocampus at various time points after 5 minutes of transient forebrain *** the sham-operated group,GLUT3 immunoreactivity in the hippocampal CA1 region was weak,in the pyramidal cells of the CA1 region increased in a time-dependent fashion 24 hours after ischemia,and in the hippocampal CA1 region decreased significantly between 2 and 5 days after ischemia,with high level of GLUT3 immunoreactivity observed in the CA1 region 10 days after *** a double immunofluorescence study using GLUT3 and glial-fibrillary acidic protein（GFAP）,we observed strong GLUT3 immunoreactivity in the ***3 immunoreactivity increased after ischemia and peaked 7 days in the dentate gyrus after ischemia/*** a double immunofluorescence study using GLUT3 and doublecortin（DCX）,we observed low level of GLUT3 immunoreactivity in the differentiated neuroblasts of the subgranular zone of the dentate gyrus after ***3 immunoreactivity in the sham-operated group was mainly detected in the subgranular zone of the dentate *** results suggest that the increase in GLUT3 immunoreactivity may be a compensatory mechanism to modulate glucose level in the hippocampal CA1 region and to promote adult neurogenesis in the dentate gyrus.

Experimental and Computational Studies on Coanda Nozzle Flow for the Air Knife Application

To control the film thickness of zinc in the process of continuous hot-dip galvanizing, it is known from the early days that the gas wiping through an air knife is the most effective one. The gas wiping using in galvanizing process brings about a problem of splashing from the strip edge for a certain high speed of coating. So, in the present study, the effects of the deflection angle of Coanda nozzle on jet structure and the distribution of impinging pres- sure at the plate surface are investigated numerically and experimentally. In numerical analysis, the governing equations consisted of three-dimensional time dependent full Navier-Stokes equations, standard k-ε turbulence model to solve turbulent stTess and so on are employed. In experiment, 16 channel pressure scanning valve and 3-axis auto traversing unit are used to measure the impinging pressure at the strip surface. As a result, it is found that the smaller the deflection angle for the same nozzle slit of air knife is, the larger the impinging pressure is. To reduce the size of separation bubble and to enhance the cutting ability, it is recommendable to use an air knife with the Coanda nozzle.

Effectiveness and safety of auriculotherapy for breastfeeding: a systematic review

BACKGROUND Closed rupture of the little and ring finger flexor tendons caused by the hamate is mostly associated with a fracture or nonunion of the hamate *** one case of a closed rupture of the finger flexor tendon caused by osteochondroma in the hamate has been ***,we present a case study to highlight the possibility of hamate osteochondroma as a rare cause of finger closed flexor tendon rupture based on our clinical experience and literature *** SUMMARY A 48-year-old man who had been a rice-field farmer for 7–8 h a day for the past 30 years visited our clinic due to the loss of right little finger and ring finger flexion involving both the proximal and distal interphalangeal *** patient was diagnosed with a complete rupture of the ring and little finger flexors because of the hamate and was pathologically diagnosed with an *** surgery was performed,and a complete rupture of the ring and little finger flexors due to an osteophyte-like lesion of the hamate was observed,which was pathologically diagnosed as an *** One should consider that osteochondroma in the hamate may be the cause of closed tendon ruptures.

Effects of Nitrogen Source and Bacterial Elicitor on Isoflavone Accumulation in Root Cultures of Albizzia kalkora （Roxb.） Prain

Changes in cellular isoflavone （daidzein and genistein） contents were monitored in root cultures of Albizzia kalkora （Roxb.） Prain after feeding different ratios of NH4^＋/NO3^- and treatment with a biotic elicitor （three strains of Rhizobium sp.）. The NH4^＋/NO3^- ratio appears to be positively correlated with daidzein content in the roots and shows a negative correlation with genistein. Among the three different strains of Rhizobium used, the strain ATCC 15834 caused a 35% increase in daidzein production by infection. In the case of genistein, maximum production （94%） was obtained when cultures were treated on Day 6 by the strains ATCC 15834 and KCTC 1541. The biosynthetic pathway of the two isoflavones apparently reacts differently to the same culture conditions and the same strains of Rhizobium. Therefore, the present data suggest that the production of daidzein and genistein could be modulated by changing the NH4^＋/NO3^- ratio and the application of Rhizobium.

The somatosensory system plays a crucial role in executing precise movements by providing sensory feedback （Farrer et al., 2003; Rabin and Gordon, 2004）. Somatosensory dys- function is a common problem following stroke. In partic- ular, somatosensory impairments, such as impairment in touch, proprioception, light touch, and vibration have been frequently observed （Carey et al., 1993; Sullivan and Hed- man, 2008; Tyson et al., 2008）. Patients with somatosensory dysfunction show negative effects on motor control, and it sometimes becomes difficult to perform daily activities independently.
